Chromosome 17 abnormalities and mutation of the TP53 gene: correlation between cytogenetics, flow cytometry and molecular analysis in three cases of chronic myeloid leukemia

Abstract: chronic myeloid leukemia (cml) have been described. this chromosomal region contains the tumor suppressor gene tp53 that may be an important factor in the evolution of this disease. in this study, we used flow cytometry and western blotting to assess p53 protein expression and single stranded conformational polymorphism to examine tp53 gene alterations in three patients with cml who showed alterations in 17p. only the case with del(17)(p11) had p53 expression positive by flow cytometry and an abnormal migration pattern by sscp analysis. the importance of the correlation between the results obtained with these techniques, as well as the clinical course of the patients, are discussed.

Abstract: cows on diets deficient in p have not responded p supplementation during late pregnancy, although they may respond in early lactation. bone metabolism of range cows has been monitored. all cows received complete mineral mixtures during wet season; during dry season, the imposed treatments were: complete mineral mixtures (mmc) or mineral mixtures lacking dicalcium phosphate, supplemented (mmr) or not (mm) with concentrate. cows supplemented with the concentrate tended to show less variation on bone measurements. calcium concentrations on bone ash were always below 34% without any symptom of ca deficiency. phosphorus concentrations on bone ash were also marginal, even in cows that received additional p all year round, while plasma p was always normal. osteocalcin and bone alkaline phosphatase showed higher values (p<0.05) during late pregancy, when deoxypyridinoline concentrations were the lowest. deoxypyridinoline increased at parturition and during early lactation. cows that received concentrate also presented lower deoxypyridinoline mean values. after three years, beef cows did not show signs of bone loss in any of the treatments.

Abstract: this work aims to identify the production arrangements developed by family farmers within the family unit and in cooperatives, assessing its impact on the area in terms of creation and expansion of social capital. it also tries to identify the strategies used by cooperatives to deal with the local adversities, which provide them with particular dynamics. do the cooperatives stimulate the mobilization of their members towards organizational and technological innovations? what is the impact in terms of increase of the social capital and advancement of the territories? in the analysis of these issues, the article examines a qualitative survey conducted in southern france, with reference to two case studies - société coopérative agricole fromagerie des cévennes and coopérative origine cévennes. in both cases, the emphasis has been on the know-how and leadership of the local agents, which are seen as "subjects of their own development," rather than "objects of study." the study reveals the limits and the power of a cooperative; even if it is separated by a considerable distance from the centers of distribution, and suffers from high operating costs, arid lands and infrastructure problems. these cases are particularly important to reinforce the importance of the cooperatives, especially in areas considered disadvantaged.

Abstract: to contribute to the knowledge of oomycetes of the brazilian atlantic rainforest, water and soil samples were collected, every three months, from june/2005 to june/2006, in the parque estadual da serra da cantareira, s?o paulo state, brazil. the samples were treated with the multiple baiting technique, in the laboratory and in the field, to isolate these organisms. twentyone species were identified, two of leptomitales and 19 of saprolegniales, with 179 occurrences during the studied period.
Study on the origin and nature of the adenomatoid odontogenic tumor by immunohistochemistry

Abstract: the adenomatoid odontogenic tumor (aot) is a clinically benign lesion. discussions about the aot hamartomatous or neoplastic nature, and the probable odontogenic epithelial cell it originates from still exist. this research aimed to study and discuss the subject by the immunohistochemical detection of cytokeratins, laminin, collagen iv, pcna and p53 in 8 tumor samples and 8 dental follicle samples containing reduced enamel epithelium. the results have shown that ck14 labelling indicated differentiation grades for secreting ameloblasts or ameloblasts in the post-secreting stage in the adenomatoid structure of aot. laminin, found on the luminal surface of adenomatoid structures, was compatible with the reduced enamel epithelium during the "protective stage of amelogenesis". pcna specifically labelled the spindled areas and peripheral cords of the aot, indicating that these areas are responsible for tumor growth. after considerations about pathogenesis, the authors suggested that the nature of aot is hamartomatous with histogenesis from the reduced enamel epithelium.
Interactions between DNA purinic bases and amodiaquine: A theoretical approach

Abstract: We study theoretically the amodiaquine-adenine and amodiaquine-guanine adducts formation using Density Functional Theory (B3LYP) and the 6-31G(d) basis set for the geometry optimizations and 6-31+G(d,p) for the analysis of the global indexes: electrophilicity (w), electronic chemical potential (m), hardness (h) and softness (S), based in the Frontier Molecular Orbital Theory – FMO. Local softness for nucleophilic reaction (sk+) sites over guanine was evaluated using Fukui function (f k). We also evaluated the guanine Electrostatic Potential (EP) values using the (MSK) charge scheme. The theoretical calculations had demonstrated that the amodiaquine has greater electronic affinity for the guanine, with irreversible formation of the amodiaquine-guanine adduct, as reported before on a previous experimental work.

Abstract: a case of mid-dermal elastolysis is reported in a 37-year-old female patient, who presented skin areas with fine wrinkles, mainly in fold areas and also perifollicular papules similar to peau d'orange in the paravertebral area, lending her a prematurely elderly appearance. histological examination revealed fragmentation and rarefaction of elastic fibers, restricted to the mid-dermis portion.
Inter and intra-guild interactions in egg parasitoid species of the soybean stink bug complex

Abstract: the objective of this research was to evaluate the parasitism behavior of telenomus podisi ashmead, trissolcus basalis (wollaston) e trissolcus urichi crawford (hymenoptera: scelionidae) on eggs of nezara viridula l., euschistus heros f., piezodorus guildinii westwood and acrosternum aseadum rolston (heteroptera: pentatomidae), in no choice and multiple choice experiments. for all parasitoid species, the results demonstrated the existence of a main host species that maximizes the reproductive success. the competitive interactions among the parasitoid species were investigated in experiments of sequential and simultaneous release of different combinations of parasitoid pairs on the hosts n. viridula, e. heros and a. aseadum. exploitative competition was observed for egg batches at the genus level (telenomus vs. trissolcus) and interference competition at the species level (t. basalis vs. t. urichi). trissolcus urichi was the most aggressive species, interfering with the parasitism of t. basalis. generally, t. basalis showed an opportunistic behavior trying to parasitise eggs after t. urichi had abandoned the egg batch. the selection of parasitoid species for use in augmentative biological control programs should take into account the diversity of pentatomids present in soybean in addition to the interactions among the different species of parasitoids.
Molecular typing of Candida albicans strains isolated from nosocomial candidemia

Abstract: yeasts of the genus candida have been recognized as important microorganisms responsible for nosocomial fungemia. six blood-stream and two intravenous central catheter c. albicans strains were isolated from eight patients and studied by electrophoretic karyotyping of chromosomal dna by pulsed-field gel electrophoresis. seven chromosomal dna profiles were identified. two patients showed isolates with the same profile, suggesting nosocomial transmission. karyotyping of c. albicans revealed an excellent discriminatory power among the isolates and may therefore be useful in the study of nosocomial candidemia.
