Comparative cytogenetic analysis of two grasshopper species of the tribe Abracrini (Ommatolampinae, Acrididae)

Abstract: the grasshopper species orthoscapheus rufipes and eujivarus fusiformis were analyzed using several cytogenetic techniques. the karyotype of o. rufipes, described here for the first time, had a diploid number of 2n = 23, whereas e. fusiformis had a karyotype with 2n = 21. the two species showed the same mechanism of sex determination (xo type) but differed in chromosome morphology. pericentromeric blocks of constitutive heterochromatin (ch) were detected in the chromosome complement of both species. cma3/da/dapi staining revealed cma3-positive blocks in ch regions in four autosomal bivalents of o. rufipes and in two of e. fusiformis. the location of active nors differed between the two species, occurring in bivalents m6 and s9 of o. rufipes and m6 and m7 of e. fusiformsi. the rdna sites revealed by fish coincided with the number and position of the active nors detected by agno3 staining. the variability in chromosomal markers accounted for the karyotype differentiation observed in the tribe abracrini.

Evaluation of immunogenicity elicited from two DNA vaccine candidates that expresses the prM and E genes of the dengue-3 virus  [PDF]

Abstract: In this work, we report the evaluation of two DNA vaccines against dengue-3 virus (DENV-3). The first construction, called pVAC3DEN3, was engineered inserting the pre-membrane (prM) and envelope (E) gene of DENV-3 truncated with a restriction site between them, as previously described. The second construction was developed cloning the full gene sequence of prM and E from DENV-3 virus in pCI plasmid for mammalian expression and was denominated pVAC1WDEN3. The results showed that both constructions were capable of expressing the prM and E proteins, as demonstrated by ELISA and immunoblotting detection in cell culture transfected with the plasmids. After positive “in vitro” results, the vaccine candidates were used to immunize BALB/c mice and the elicited response was investigated. After immunization by intramuscular inoculation with three doses of each vaccinal clone the animals were sacrificed, the cytokine levels and T cell response were analyzed in the spleens, after three days of culture with stimulus, our analysis showed that the two constructions elicited T cell responses mea- sured by BrdU incorporation assay and high levels of IFN-γ, detected in the supernatant of the cultures. Moreover, both constructions induced detectable titers of neutralizing antibodies in mice. And finally the survival rate of the immunized animals after intracerebral challenge was analyzed, showing a better result in the pVAC3DEN3 group with an 80% survival compared with a 50% survival of the pVAC1 WDEN3. Thus, these data showed that our two constructions were able to induce specific immune response and protects mice against a lethal challenge with DENV-3, and these vaccine candidates can be employed to develop a viable dengue vaccine.

Abstract: this article's objective is to present the experience of women in their daily relationship with their partners, in a qualitative study. there were 14 women participating age between 20 to 24 years oíd, they all lived in the outskirts of maceio, capital of alagoas state. data collection was done through interviews, using gender as the theoretical reference and oral history as the thematic methodologic procedure. the pregnancy occurred during adolescence and it was provided a consensual unión, when it did not exist before, except for one of the participants. but in this kind of unión, some men acted more freely, having extramarital relations and many times attacking their wives at home. most women live with the problem of male infidelity and domestic violence, that increases with poverty. as we read their histories, we notice that they consider wrong their partner attitude of betraying and harming, but most of them have passive reactions when they know about, or when they complaint, it generates fights between the couple, but it d?es not modify men's behavior and it d?esn't grant women's physical and psy-chological integrity. this situation shows the male domination exerted in gender relations, it characterizes the gender asymmetries that permeate the couple relations, and gives visibility to the submission position to which these women are exposed, in this study.

Abstract: Este artigo tem como objetivo apresentar a experiência de mulheres na rela o vivida com seus parceiros, cuja natureza do estudo foi qualitativa. Participaram 14 mulheres na faixa etária de 20 a 24 anos, residentes na periferia de Maceió, capital do estado de Alagoas. A coleta de informa o foi realizada por meio de entrevistas, utilizando gênero como referêncial teórico e a historia oral temática como procedimento metodológico. A gravidez ocorrida na adolescência propiciou a uni o consensual, quando ela n o existia anteriormente, com exce o de urna das participantes. Porém nesta uni o alguns homens agiam mais livremente, maniendo rela es extraconjugais e chegavam a agredir suas mulheres, em casa. A maioria das mulheres convive com o problema da infidelidade masculina e da violência doméstica, que é acrescido das circunstancias de dificuldade de sobrevivência. No exame de suas historias observamos que elas consideram errada a atitude do parceiro, de trair e agredir, mas a maioria se submete a tomarem conhecimento e terem rea es passivas, ou queixas que geram brigas entre o casal, porém que n o garante a modifica o de comportamento do mesmo e a sua integridade física e psicológica. essa situa o revela a domina o masculina exercida as rela es entre os sexos, caracteriza as assimetrias de gênero que permeiam as rela es conjugáis, no grupo estudado, e dá visibilidade á posic o de submiss o a que essas mulheres est o sujeitas. El presente estudio, de carácter cualitativo, tuvo como objetivo presentar resultados de la experiência de mujeres en la relación con sus parejas, en su condición de madres y esposas. Participaron 14 mujeres con edad entre 20 y 24 a os, residentes en las afueras de Maceió, capital del estado de Alagoas, Brasil. La información se obtuvo a través de entrevistas, utilizando el gênero como una referência teórica y la historia oral como un procedimiento temático metodológico. El embarazo ocurrido en la adolescéncia determino la unión consensual, a excepción de una de las participantes que ya había contraído matrimonio. No obstante, ante esta unión algunos hombres actuaban con mayor libertad, manteniendo relaciones extraconyugales, y llegaban a agredir a sus esposas en el hogar. La mayor parte de las mujeres convive con el problema de la infidelidad masculina y de la violéncia doméstica, que resulta agravado por las circunstancias que dificultan la sobrevivência. Abstract: Objective: to describe and analyze prenatal care, according to criteria of PHPN. Methodology: this is about an exploratory and descriptive study, from quantitative approach. Study’s subjects were 4131 pregnant women registered in SISPRENATAL from 2002 to 2006. Data were collected through a form applied to SISPRENATAL after approval by the Ethics Committee on Research of the Centre Integrated Health Amauri de Medeiros/CISAM under protocol number CAAE - 0263.0.000.250-08. Results: 76.5% of pregnant women started prenatal care until the fourth month of pregnancy. As for exams, 60.3% of them made any classification blood, 24.7% had VDRL full, 59.9% had hemoglobin and hematocrit, 24.86% had complete glucose exam, 24.1%, 26.9% and 46 4%, respectively, underwent urinalysis, HIV testing and were immunized against tetanus. As indicators: consultations, examinations and postpartum period, only 11.1% had them all together, so this is performed six or more consultations, recommended tests and post-partum visit. Conclusion: we concluded that the percentages identified can cause negative impacts to the health of mother and baby, and to difficult, consistently, the reduction of maternal and perinatal mortality. Therefore, the study highlights the need to train professionals, to ensure the realization of exam and treatment of complications, to ensure a quality care, while respecting the rights established.

Abstract: the purpose of this work was to collect the main information from the literature about the biotyping of cryptococcus neoformans. the more up-to date research concerning the epidemiology of cryptococcosis comprising quite a few articles, mainly after the advent of aids, was also reviewed. the cryptococcus neoformans varieties neoformans and gattii are well defined biochemically nowadays chiefly through the c.g.b. medium, according to kwon-chung et al. (1982)24. the isolation of c. neoformans var. gattii from flowers and leaves of eucalyptus camaldulensis and eucalyptus tereticornis, specially in australia, through the works of ellis & pfeiffer (1990) 16 and pfeiffer & ellis (1992) 41 permitted very interesting epidemiological investigations on c. neoformans, a capsulaled yeast by which sanfelice 50, 51, in italy (1894; 1895) attracted attention of medical class. busse 8, in 1894, described the first human case of cryptococcosis under the presentation of a bone lesion simulating sarcoma. in this paper, the brazilian researchers focused on this subject were pointed out, followed by the author's experience with the c.g.b medium (l - canavanine, glycin and bromothymol blue) proposed by kwon-chung et al. (1982) 24 with very good results. it was possible with such medium the study of 50 c.n.s. liquor samples, being 39 from aids patients (78%) and 11 from non-aids ones (22%). thirty-seven out of the 39 hiv- positive patients (74%) were identified as c. neoformans var. neoformans and 2 (4%) as c. neoformans var. gattii. from the negative hiv, 8 (16%) were classified as c. neoformans var. neoformans and 3 (6%) as c. neoformans var. gattii. we could not perform the serotyping of the above referred samples. it is evident anyway that in brazil there exist both varieties gattii and neoformans, agents of neurocryptococcosis, including aids patients. the importance of neurocryptococcosis, mainly among aids patients, is stressed here, showing once more the value of c.g.b. medium in

Abstract: we review here the chemistry of reactive oxygen and nitrogen species, their biological sources and targets; particularly, biomolecules implicated in the redox balance of the human blood, and appraise the analytical methods available for their detection and quantification. those biomolecules are represented by the enzymatic antioxidant defense machinery, whereas coadjutant reducing protection is provided by several low molecular weight molecules. biomolecules can be injured by rons yielding a large repertoire of oxidized products, some of which can be taken as biomarkers of oxidative damage. their reliable determination is of utmost interest for their potentiality in diagnosis, prevention and treatment of maladies.
Hydralazine Hydrochloride: An Alternative Complexometric Reagent for Total Iron Spotometric Determination  [PDF]

Abstract: An alternative spectrophotometric method has been developed for total iron determination using flow injection analysis (FIA). The procedure is based on the coordination reaction between hydralazine and Fe2+ ions, which results in the formation of a purple complex monitored at 538 nm. For determination of total iron, Fe3+ ions were reduced using ascorbic acid. Under optimized conditions, a linear calibration graph (0.1 - 6.0 ?g?ml–1; n = 6) was obtained. The method allows LOD (3? of blank/slope = 0.06 ?g?ml–1) and LOQ (10? of blank/slope = 0.22 ?g?ml–1). The RSD ((s/ ) × 100) for a mixed standard containing 0.60 ?g?ml–1 Fe2+ and Fe3+ was 0.10% (n = 10). Recoveries of spiked samples were 94.3% - 106.0%. The analytical frequency was 60 h–1. The effect of possible interferences has been studied. The procedure was successfully applied for analysis of environmental samples. The real samples results were comparable with those obtained by the official method considering a paired t-test and 95% of confidence level.
