Integrin β3 Crosstalk with VEGFR Accommodating Tyrosine Phosphorylation as a Regulatory Switch

Abstract: Integrins mediate cell adhesion, migration, and survival by connecting intracellular machinery with the surrounding extracellular matrix. Previous studies demonstrated the importance of the interaction between β3 integrin and VEGF type 2 receptor (VEGFR2) in VEGF-induced angiogenesis. Here we present in vitro evidence of the direct association between the cytoplasmic tails (CTs) of β3 and VEGFR2. Specifically, the membrane-proximal motif around 801YLSI in VEGFR2 mediates its binding to non-phosphorylated β3CT, accommodating an α-helical turn in integrin bound conformation. We also show that Y747 phosphorylation of β3 enhances the above interaction. To demonstrate the importance of β3 phosphorylation in endothelial cell functions, we synthesized β3CT-mimicking Y747 phosphorylated and unphosphorylated membrane permeable peptides. We show that a peptide containing phospho-Y747 but not F747 significantly inhibits VEGF-induced signaling and angiogenesis. Moreover, phospho-Y747 peptide exhibits inhibitory effect only in WT but not in β3 integrin knock-out or β3 integrin knock-in cells expressing β3 with two tyrosines substituted for phenylalanines, demonstrating its specificity. Importantly, these peptides have no effect on fibroblast growth factor receptor signaling. Collectively these data provide novel mechanistic insights into phosphorylation dependent cross-talk between integrin and VEGFR2.
From policy to practice: education reform in Mozambique and Marrere Teachers’ Training College

Abstract: The Mozambican government has introduced reforms of basic education, notably the introduction of interdisciplinarity, learner-centredness and new teaching pedagogies. This is a case study of how these curriculum reforms have been implemented at Marrere Teachers’ Training College. We conducted interviews with lecturers, observed their teaching practices, and studied student results to assess teaching outcomes. The study is grounded in the literatures on educational change and globalization. The problems of policy and practice have focused attention on bottom-up and top-down research, and hybrid approaches. The study of globalization has highlighted the relationship between curriculum change and the world economy. There is a paucity of research on how these developments have affected underdeveloped countries. We found that practical issues influence implementation. Lecturers did not understand the meaning of interdisciplinarity. They could, however, articulate the meaning of learnercentredness. Lesson observations showed they did not implement it. Against the backdrop of these inter-related factors, final year students performed poorly in examinations. These analyses show the complexities of the moving from policy to practice, and the global to the local.
Nueva luz sobre viejos problemas: incidencia de la cuantificación en la historiografía sobre la industria temprana en Uruguay

Abstract: in this paper we analyze the impact of quantitative research on the historiography of early industry in uruguay. we distinguish two phases that are connected by a transitional period. we maintain that the diffusion of quantitative methods among economic historians, and in particular their estimates of gdp, made for a change in the way they approached industry before 1930. although the traditional problems remain -like the role of protectionism- the kind of evidence used to support the arguments changed. also, the key role off protectionism in the explanation of industrial growth has been challenged. the new evidence suggests that the factors behind early industrial growth were protectionist laws and the effects of international crises. quantitative historiography maintains that this is not accounted for in estimations of product, and stresses the importance of the economy as a whole and the linkages between the export and domestic sectors.

Abstract: El trabajo analiza la influencia que la introducción de métodos cuantitativos ha tenido en la historiografía de la industria temprana en Uruguay. Se distinguen dos etapas articuladas por un periodo transicional. Sostenemos que la difusión entre historiadores económicos de métodos cuantitativos y, en particular, la estimación de series de producto total e industrial, provocó un viraje en la forma de abordar la industria anterior a 1930. Si bien permanecen los antiguos problemas -como la importancia del proteccionismo-, se modifica el tipo de evidencia utilizada, y se cuestiona la tesis central y la periodización de la historiografía anterior. Según esta, la industria temprana creció al amparo de las leyes proteccionistas y en el entorno de las crisis internacionales. La historiografía cuantitativa sostiene que esto no se constata en las estimaciones de producto y destaca la importancia de la economía en su conjunto y los encadenamientos provenientes del sector exportador. Bound states in Yukawa theory

Abstract: A generalization of the Gell-Mann-Low Theorem is applied to bound state calculations in Yukawa theory. The resulting effective Schroedinger equation is solved numerically for two-fermion bound states with the exchange of a massless boson. The complete low-lying bound state spectrum is obtained for different ratios of the constituent masses. No abnormal solutions are found. We show the consistency of the non-relativistic and one-body limits and discuss the special cases of identical fermions and fermion-antifermion states. To our knowledge, this is the first consistent calculation of bound states in pure Yukawa theory (without UV cutoff).
The Generalized Gell-Mann--Low Theorem for Relativistic Bound States

Abstract: The recently established generalized Gell-Mann--Low theorem is applied in lowest perturbative order to bound-state calculations in a simple scalar field theory with cubic couplings. The approach via the generalized Gell-Mann--Low Theorem retains, while being fully relativistic, many of the desirable features of the quantum mechanical approaches to bound states. In particular, no abnormal or unphysical solutions are found in the model under consideration. Both the non-relativistic and one-body limits are straightforward and consistent. The results for the spectrum are compared to those of the Bethe-Salpeter equation (in the ladder approximation) and related equations.
Comparación de anticoagulantes de heparina de litio y sodio en la bioquímica plasmática del cocodrilo de pantano (Crocodylus moreletii), en Campeche, México

Abstract: lithium heparin and sodium heparin anticoagulants were compared with the aim to measure concentrations and activity of selected plasma biochemical analytes in wild and captive morelet's crocodiles (crocodylus moreletii). blood samples were obtained from the ventral caudal tail vein in 63 crocodiles (32 wild and 30captive) and each sample was placed in both lithium heparin and sodium heparin. plasma was removed and duplicate samples were assayed for the following: cholesterol, glucose, uric acid, creatinine and alanine aminotransferase (alt). the only analyte having a significant difference (t = -2.95, df = 56, p < 0.01) when comparing the two anticoagulants was alt. still, the difference was not considered to be clinically significant. the results of this study indicates that the type of heparin used as an anticoagulant, does not affect concentratrions and activity of certain plasma biochemicals of morelet's crocodiles, and perhaps other species of crocodilians.
Comparación de anticoagulantes de heparina de litio y sodio en la bioquímica plasmática del cocodrilo de pantano (Crocodylus moreletii), en Campeche, México

Abstract: Los anticoagulantes de heparina de litio y heparina de sodio se compararon con el objetivo de medir las concentraciones y actividad de analitos bioquímicos seleccionados en el plasma de cocodrilos de pantano (Crocodylus moreletii). Se obtuvieron muestras de sangre de la vena caudal de 63 cocodrilos (32 silvestres y 30 en cautiverio), cada muestra se analizó en heparina de litio y heparina de sodio. Se obtuvo el plasma y las muestras pareadas fueron analizadas para los siguientes analitos: Colesterol, glucosa, ácido úrico, creatinina y alanina aminotransferasa (ALT). El único analito que mostró una diferencia signifi cativa (t = -2.95, gl = 56, P < 0.01) fue el ALT para ambos anticoagulantes; sin embargo, la diferencia no se considera clínicamente signifi cativa. Los resultados de este estudio indican que el tipo de heparina usado como anticoagulante no afecta las concentraciones y actividades de la bioquímica plasmática del cocodrilo de pantano y quizá de otras especies de cocodrilianos.
Development and testing of an online method to measure ambient fine particulate Reactive Oxygen Species (ROS) based on the 2',7'-dichlorofluorescin (DCFH) assay

Abstract: An online, semi-continuous instrument to measure fine particle (PM2.5) reactive oxygen species (ROS) was developed based on the fluorescent probe 2'7'-dichlorofluorescin (DCFH). Parameters that influence probe response were first characterized to develop an optimal method for use in a field instrument. The online method used a mist chamber scrubber to collect total (gas plus particle) ROS components (ROSt) alternating with gas phase ROS (ROSg) by means of an inline filter. Particle phase ROS (ROSp) was determined by difference between ROSt and ROSg. The instrument was deployed in urban Atlanta, Georgia, and at a rural site during various seasons. Concentrations from the online instrument generally agreed well with those from an intensive filter measurement of ROSp. Concentrations of the ROSp measurements made with this instrument were lower than reported in other studies, often below the instrument's average limit of detection (0.15 nmol H2O2 equivalents m 3). Mean ROSp concentrations were 0.26 nmol H2O2 equivalents m 3 at the Atlanta urban sites compared to 0.14 nmol H2O2 equivalents m 3 at the rural site.
