Characteristics of the Alternative Phenotype of Microglia/Macrophages and its Modulation in Experimental Gliomas

Abstract: Microglia (brain resident macrophages) accumulate in malignant gliomas and instead of initiating the anti-tumor response, they switch to a pro-invasive phenotype, support tumor growth, invasion, angiogenesis and immunosuppression by release of cytokines/chemokines and extracellular matrix proteases. Using immunofluorescence and flow cytometry, we demonstrate an early accumulation of activated microglia followed by accumulation of macrophages in experimental murine EGFP-GL261 gliomas. Those cells acquire the alternative phenotype, as evidenced by evaluation of the production of ten pro/anti-inflammatory cytokines and expression profiling of 28 genes in magnetically-sorted CD11b+ cells from tumor tissues. Furthermore, we show that infiltration of implanted gliomas by amoeboid, Iba1-positive cells can be reduced by a systematically injected cyclosporine A (CsA) two or eight days after cell inoculation. The up-regulated levels of IL-10 and GM-CSF, increased expression of genes characteristic for the alternative and pro-invasive phenotype (arg-1, mt1-mmp, cxcl14) in glioma-derived CD11b+ cells as well as enhanced angiogenesis and tumor growth were reduced in CsA-treated mice. Our findings define for the first time kinetics and biochemical characteristics of glioma-infiltrating microglia/macrophages. Inhibition of the alternative activation of tumor-infiltrating macrophages significantly reduced tumor growth. Thus, blockade of microglia/macrophage infiltration and their pro-invasive functions could be a novel therapeutic strategy in malignant gliomas.

Abstract: Wprowadzenie i cel: Wobec zmieniaj cej si etiologii zaka eń skóry, przeprowadzono analiz zaka eń w obr bie pachwin u pacjentów badanych w latach 1997-2003 w Pracowni Mikologicznej WPSW w Kielcach. Materia i metody: W tym okresie w Laboratorium Mikologicznym by o przebadanych 25 128 osób. Pobieranie materia u i diagnostyk przeprowadzono na podstawie aktualnie obowi zuj cych metod laboratoryjnych. Wyniki i wnioski: Zaka enia grzybicze i zaka enia wywo ane przez C. minutissimum w obr bie pachwin stwierdzono u 727 pacjentów, cz ciej u zamieszkuj cych miasta (71%) ni wsie (29%), a tak e cz ciej u m czyzn (81%) ni kobiet (19%). Wi e si to prawdopodobnie z wi kszymi mo liwo ciami diagnostycznymi i wi ksz wiadomo ci ludno ci zamieszkuj cej miasta oraz predyspozycjami anatomicznymi i zaniedbaniami higienicznymi u pacjentów p ci m skiej. Szczytzachorowań m czyzn przypada na przedzia wiekowy mi dzy 11 a 20 r. . (26%), a u kobiet mi dzy 41 a 50 r. . (28%). Liczba zachorowań u obojga p ci w poszczególnych latach by a w miar ustabilizowana. Najcz stszymi czynnikami etiologicznymi zaka eń pachwin w tym okresie by y: £ floccosum (29,3%), T. rubrum (23,7%), C. minutissimum (19,0%) oraz C. albicans (16,9%). Na dalszych miejscach uplasowa y si kolejno: C. species, T. mentagrophytes v. gypseum, T. mentagrophytes v. quinckenum oraz M. canis, M. gypseum, T. mentagrophytes v. interdigitale. Stwierdzono równie , e zaka enia dermatofitami, takimi jak: £ floccosum, T. rubrum oraz T. mentagrophytes v. gypseum, wykazuj tendencj malej c , a zaka enia wywo ane przez grzyby dro d opodobne (C. albicans i C. species) i przez C. minutissimum - tendencj rosn c . Wynika z tego potrzeba zwrócenia baczniejszej uwagi na leczenie i profilaktyk zaka eń pachwin wywo anych przez dro d aki oraz przez C. minutissimum u pacjentów regionu wi tokrzyskiego.
The Useful art of the social-economic Rankings

Abstract: The purpose of the paper is to identify advantages and disadvantages of various methods of constructing rankings. The subject of our study is important due to the international debate on development and welfare measurement methods and ways of comparing results obtained for different countries. Because GDP per capita does not allow sufficient assessment, countries are compared on the basis of many criteria and results are usually presented in form of rankings. We discuss different outranking methods originating from multidimensional statistical analysis and multicriteria optimization and compare them taking into consideration the effect of each method and each set of criteria on the final result. Our remarks are illustrated by rankings of development and economic performance built for European Union countries. Our observations and results can be regarded also as an opinion in the discussion on the report of the International Commission onMeasurement of Economic Performance and Social Progress chaired by J.E. Stiglitz and A. Sen.

Abstract:

To provide enough space to carry all surface charges responsible for high cation exchange capacity of plant roots, large area of the root specific surface is necessary, however all experimental methods used up to date give too small surface area values. In this paper, we propose to measure the plant roots surface area using water vapor adsorption isotherm. This method gives roots specific surface areas compatible to CEC. Methodical aspects of the measurements are described along with theoretical background for calculating specific surface area on the example of roots of barley grown in nutrient solution.

Abstract: Wprowadzenie: W oportunistycznych zaka eniach grzybami dro d opodobnymi z rodzaju Candida opisuje si wiele czynników usposabiaj cych. W pi miennictwie dyskutuje si równie mo liwo jatrogennego oddzia ywania pola magnetycznego stosowanego w zabiegach rehabilitacyjno-leczniczych. Cel pracy: Ocena wp ywu rezonansowego pola magnetycznego stosowanego w zabiegach magnetostymulacji na aktywno wzrostow izolowanych grzybów dro d opodobnych z rodzaju Candida. Materia i metody: Badano 30 osób, kobiet i m czyzn, poddawanych zabiegom magnetostymulacji (w systemie QUANTRONIC MRS-2000) ze wskazań rehabilitacyjno-leczniczych, u których z jamy ustnej i przestrzeni mi dzypalcowych stóp izolowano grzyby dro d opodobne z rodzaju Candida. Okre lano: wielko kolonii, mo liwo wzrostu w hipertonicznych st eniach NaCI, a dla gatunku C. albicans -zdolno do wytwarzania chlamydospor i wypustek rostkowych. Badania prowadzono w dwu uk adach: I - materia do badań pobierano 3-krotnie: przed pierwszym zabiegiem, po pi tym zabiegu i po pi tnastym (ostatnim) zabiegu magnetostymulacji; II - materia pobierano przed pierwszym zabiegiem, nast pnie hodowle by y poddawane bezpo redniemu dzia aniu pola magnetycznego i oceniane równolegle z drugim i trzecim badaniem I uk adu badawczego. Wyniki: Obserwowano zmniejszenie wielko ci kolonii wszystkich izolowanych gatunków oraz zmniejszenie liczby wytwarzanych przez C. albicans chlamydospor i wypustek rostkowych. Zmiany te by y wprost proporcjonalne do sumarycznej dawki ekspozycji. Nie obserwowano wp ywu na mo liwo wzrostu w hipertonicznych st eniach NaCI. Wnioski: Rezonansowe pole magnetyczne stosowane w zabiegach magnetostymulacji obni a aktywno wzrostow grzybów dro d opodobnych z rodzaju Candida.
THE EFFECT OF H-FABP GENE POLYMORPHISM ON CARCASS AND MEAT QUALITY IN THE POLISH NATIVE Z OTNICKA SPOTTED PIG

Abstract: To estimate the relationships between the H-FABP/HinfI genotype and quality of carcass and meat traits, a total of 30 pigs Z otnicka Spotted breed were investigated. Blood samples were taken during slaughter into test tubes containing anticoagulant K3EDTA and stored at -25oC until analysed. The H-FABP genotypes were identified with PCR-RFLP method using HinfI restriction endonuclease. Meat physico-chemical traits were determined in the longissimus lumborum (LL) muscle. Two alleles (H and h) and three genotypes (HH, Hh and hh) were identified within studied pig sample. The effect of H-FABP gene polymorphism was showed on body weight at slaughter and cold carcass weight, where both weights of body were higher in hh group (113.33 kg and 84.27 kg) than in Hh (106.06 kg and 76.40 kg) and HH (103.75 kg and 73.40 kg); P≤0.05 and P≤0.01. Differences between the means of backfat thickness for three genotype groups were not significant. However, significant relations were found between the polymorphism at locus H-FABP/HinfI and basic chemical meat composition. The pigs meat with genotype hh were marked by a higher intramuscular fat content (2.64%) compared to HH group (1.99%); P≤0.05. Values of all analysed traits were within ranges indicating good meat quality of the Polish native Z otnicka Spotted pigs.

Abstract: e-government can be defined as internet applications for public administration processes and decisión making on local, regional, national as well as cross-national level. therefore, the strong necessity to ensure vertical and horizontal interoperation of administrative units is arrived. the main goal of the paper is to reveal how interoperability is built into the public administration engineering process. the paper covers explanations how interoperability is important for e-governmental processes covering documents flows and what activities have been realized with eu agencies to ensure interoperability development. the first part of the paper comprises explanation of e-government document engineering and administrative process development. the second part covers consideration on interoperability for e-business and public administration. the third part includes analysis of what standardization is demanded for interoperability of e-government agencies and how different countries' governments are prepared to ensure such interoperability in public administration in the light of documents, institutional norms and open standards.

Abstract: The status of two Monte Carlo generators, HELAC-PHEGAS, a program for multi-jet processes and VBFNLO, a parton level program for vector boson fusion processes at NLO QCD, is briefly presented. The aim of these tools is the simulation of events within the Standard Model at current and future high energy experiments, in particular the LHC. Some results related to the production of multi-jet final states at the LHC are also shown.
