Stability of isoenzyme and kinetoplast DNA (k-DNA) patterns in successively cloned Trypanosoma cruzi populations

Abstract: four trypanosoma cruzi strains from zymodermes a, b, c and d were successively clonedon bhi-lit-agar-blood blab). twenty clones from the first generation (f1), 10 from the second (f2) and 4 from the third (f3) from the strains a138, b147 and c23 were isolated. the d150 strain provied 29 f1 and f2 clones. the strains and clones had their isoenzyme and k-dna patterns determined. the clones from a138, bl47 and c231 strains presented isoemzyme and k-dna patterns identical between thewmselves and their respective parental strains. therefore showing the homogenety and stability of isoenzyme and k-dna patterns after successive cloning. the dl50 strain from zymodeme d (zd) showed heterogeneity. twenty-eight out of 29 clones of the first generation were of zymodeme a and only one was of zymodeme c, confirming previous reports that zd strains consisted of za and zc parasite populations. the only d150 strain clone of zymodeme c showed a k-dna pattern identical to its parental strain. the remining clones although similar among themselves were different from the parental strain. thus the t. cruzi strains had either homonogeneus or heterogeneous populations. the clones produced by successive cloning provided genetically homonogeous populations. their experimental use will make future results more reliable and reproducible.

Abstract: This paper presents an ergonomics work analysis study on firewood kettle operation. The workload was analyzed in qualitative and quantitative forms, by documents, direct observation, interviews, postural analysis and heart frequency. The results showed that musculoskeletal disorders and work accidents are related to the working time and operator age. Besides, considering the movements performed to realize the activity, the major pain complaints concerns the right side independent to the dominant hand. Heart frequency measurements indicate a medium to high workload according to the physical conditions. Training and workstation changes are firstly suggested to improve work conditions. Este artigo apresenta o estudo da análise ergon mica do trabalho na opera o de caldeira à lenha. O trabalho pesado foi analisado de forma qualitativa e quantitativa, através de documentos, observa o direta, entrevistas, avalia es de problemas posturais e medi o da freqüência cardíaca. Os resultados mostraram que as les es musculares e os acidentes de trabalho est o diretamente relacionados com o tempo de trabalho do operador e com a idade do mesmo. Além disso, devido aos movimentos necessários para a realiza o da atividade, o lado direito é o que apresenta maiores queixas de dores, independentemente da lateralidade do operador. As medi es de frequência cardíaca indicaram que o trabalho apresenta carga de regular a alta, conforme as condi es físicas do operador. Abstract: this paper presents a taxonomic study of the genera chaetomorpha and rhizoclonium occurring in the littoral of bahia. the species were defined based on habit, presence or absence of lateral rhizoids, basal cell type, shape and cells dimensions. the presence of crystalline inclusions in cells was also verified. it were identified nine species, seven to the genus chaetomorpha (c. aerea (dillwyn) kütz., c. antennina (bory) kütz., c. brachygona harv., c. clavata kütz., c. crassa (c. agardh) kütz., c. minima collins & herv. and c. nodosa kütz.) and two to the genus rhizoclonium (r. africanum kütz. and r. riparium (roth) kütz. ex harv.). chaetomorpha crassa, c. minima and c. nodosa are new additions to the northeastern of brazil and c. aerea, c. clavata and r. africanum are additions to the bahia. three crystals types were found: 1. clusters of fine needle-shaped of silica crystals presents in c. aerea, c. antennina, c. brachygona, c. clavata and c. crassa; 2. octahedral of calcium oxalate crystals found in c. antennina and c. clavata and, 3. globular aggregates of cone-shaped calcium carbonate crystals observed in c. clavata. fertile thallus of c. antennina, c. clavata, c. nodosa, r. africanum and r. riparium are being illustrated for the first time for the brazilian coast. the vegetative and reproductive structures are described in detail and a comparison with similar species is provided.

Abstract: phosphorus is an important mineral nutrient for vegetables. this work was conducted at universidade federal da paraíba, in areia, brazil, from december/2001 to july/2002, aiming to evaluate the effect of levels of p2o5 on the yield of lima bean, cv. "orelha de vó". the experimental design was a randomized block with five treatments (0; 100; 200; 300; 400 and 500 kg ha-1 of p2 o5), and four replications. each plot consisted of 40 plants spaced 1.0 m between rows and 0.50 m between plants in each row. the estimated maximum yield of green and dry beans (5.2 and 2.7 t ha-1, respectively) corresponded, to the dosages of 309 and 302 kg ha-1 of p2o5. the levels 291 kg ha-1 and 281 kg ha-1 of p2o5 provided, respectively, the greater economic return for the yield of green (4.1 t ha-1) and dry (1.8 t ha-1) beans. the most economic levels included more than 80% of those dosages responsible for the maximum yields and indicated the economic viability of phosphorus utilization on lima bean crop. the p2o5 levels which promoted the maximum yield and the maximum economic return for green and dry beans yield were respectively, with 57.1; 56.3; 55.0 and 53.8 mg dm-3 of available p for the extractor of melich 1. the lima bean response to the phosphorus fertilization in soils with similar fertility used in the present study, for green and dry beans yield, will be reduced at levels of available p superior to 55.0 and 53.8 mg dm-3. the average concentration of p in the leaves of lima bean, 120 days after sowing, in function of levels of p2o5 was of 3.4 g kg-1. for the soil of this study the application of 291 and 281 kg ha-1 of p2o5 is more recommended to establish the lima bean respectively, for green and dry beans yield.

Abstract: the aim of this study was to investigate the feasibility of a co-processing technique for improving the manufacturing properties of maytenus ilicifolia (schrad.) planch., celastraceae, and cassia angustifolia vahl, fabaceae, extracts in order to obtain tablets containing a high dose of such extracts. an experimental mixture design was used to optimise the formulation composition. flowability parameters, such as compressibility index, time flow and angle of repose, were determined. additional important industrial parameters, such as granulometry, bulk density and moisture stability, were also studied. the results demonstrated that co-processing technique was able to improve the flowability of vegetal extracts, making these materials suitable for a direct compression process. the contour plots revealed that formulations with a higher amount of lactose produced the best flow results as well as a larger particle size and a greater bulk density. tablets from co-processed extracts containing lactose as majority diluent showed appropriate physical-chemical characteristics and presented a more stable moisture sorption behaviour compared to commercial gelatine capsules.

Abstract: Presently, several different graft materials are employed in regenerative or corrective bone surgery. However current misconceptions about these biomaterials, their use and risks may compromise their correct application and development. To unveil these misconceptions, this work briefly reviewed concepts about bone remodeling, grafts classification and manufacturing processes, with a special focus on calcium phosphate materials as an example of a current employed biomaterial. Thus a search on the last decade was performed in Medline, LILACS, Scielo and other scientific electronic libraries using as keywords biomaterials, bone remodeling, regeneration, biocompatible materials, hydroxyapatite and therapeutic risks. Our search showed not only an accelerated biotechnological development that brought significant advances to biomaterials use on bone remodeling treatments but also several therapeutic risks that should not be ignored. The biomaterials specificity and limitations to clinical application point to the current need for developing safer products with better interactions with the biological microenvironments.

Abstract: in this article the construction of a simple calorimeter for the development of a protocol with which it is possible to determine the enthalpy of dissolution of salts is described. thereunto, salts solutions such as sodium carbonate, sodium bicarbonate, copper sulfate and sodium chloride besides one thermometer and one expanded-polystyrene (styrofoam) container were used. the experiment allows to assimilate thermochemistry concepts and can be applied to any high school, since it uses materials easily obtainable at low cost.

Abstract: the employment of spad index for n status diagnosis of crops has been largely studied in recent years. however, conclusive studies about its usefulness in zucchini plants were not found in literature. the objective of this study was to evaluate the employment of spad index for n status diagnosis of zucchini plants (cv. caserta). the experiment was carried out in vi?osa, minas gerais state, brazil, in a randomized block design with five rates of mineral n (0, 50, 100, 200 and 400 kg/ha), with four replications. at the early flowering stage, total chlorophyll concentration, spad index and total n concentration in the fourth fully expanded leaf from the apex of zucchini plants were evaluated. the total chlorophyll concentration, spad index and total n concentration in zucchini leaves increased following a quadratic model as a function of the n rates. the estimated critical levels of total chlorophyll concentration, spad index and total n concentration on zucchini leaves responsible for maximum fruit yield were 5.12 mg/g of fresh matter, 55.62 units and 3.97 dag/kg of dry matter, respectively. significant linear and positive correlation between the evaluated characteristics was observed. the results indicate that the spad index can be used for n status diagnosis of zucchini plants
Viability, production and morphology of pollen grains for different species in the genus Manihot (Euphorbiaceae)

Abstract: the objective of this work was to characterize the viability, production and morphology of pollen for different species in the genus manihot. floral buds from manihot accessions were collected from two germplasm banks at embrapa cassava & fruits. the viability of the pollen was assessed via colorimetric, in vitro and in vivo assays. the diameter of the pollen grains was determined by measuring the transversal length of the grain. the experimental design was entirely randomized. studies on pollen ultrastructure were performed via scanning electron microscopy. pollen viability was high in the colorimetric tests and intermediate in vivo tests; there was no germination in the in vitro tests. the average production for all accessions was 1,253 pollen grains per floral bud. the size of the pollen grains varied from 132 to 163 μm in the wild accessions, and 129 to 146 μm in the cultivated accessions. the pollen grains for all accessions were very large, apolar, spherical as well as inaperturate, with an exine ornamented with pila organized in a croton pattern. the wild accessions, in general, produced more and larger pollen grains compared with the cultivated accessions.
