Simultaneous circulation of all four dengue serotypes in Manaus, State of Amazonas, Brazil in 2011

Abstract: introduction: manaus, the capital city of the state of amazon with nearly 2 million inhabitants, is located in the middle of the amazon rain forest and has suffered dengue outbreaks since 1998. methods: in this study, blood samples were investigated using reverse transcriptase-polymerase chain reaction (rt-pcr), aimed at identifying dengue virus serotypes. results: acute phase sera from 432 patients were tested for the presence of dengue virus. out of the 432 patients, 137 (31.3%) were found to be positive. all the four dengue virus serotypes were observed. conclusions: the simultaneous circulation of the four dengue serotypes is described for the first time in manaus and in brazil.

Abstract: hantavirus disease is caused by the hantavirus, which is an rna virus belonging to the family bunyaviridae. hantavirus disease is an anthropozoonotic infection transmitted through the inhalation of aerosols from the excreta of hantavirus-infected rodents. in the county of itacoatiara in the state of amazonas (am), brazil, the first human cases of hantavirus pulmonary and cardiovascular syndrome were described in july 2004. these first cases were followed by two fatal cases, one in the municipality of maués in 2005 and another in itacoatiara in 2007. in this study, we investigated the antibody levels to hantavirus in a population of 1,731 individuals from four different counties of am. sera were tested by igg/igm- enzyme-linked immune-sorbent assay using a recombinant nucleocapsid protein of the araraquara hantavirus as an antigen. ten sera were igg positive to hantavirus (0.6%). among the positive sera, 0.8% (1/122), 0.4% (1/256), 0.2% (1/556) and 0.9% (7/797) were from atalaia do norte, careiro castanho, itacoatiara and lábrea, respectively. none of the sera in this survey were igm-positive. because these counties are distributed in different areas of am, we can assume that infected individuals are found throughout the entire state, which suggests that hantavirus disease could be a local emerging health problem.
Clinical and Virological Descriptive Study in the 2011 Outbreak of Dengue in the Amazonas, Brazil

Abstract: Background Dengue is a vector-borne disease in the tropical and subtropical region of the world and is transmitted by the mosquito Aedes aegypti. In the state of Amazonas, Brazil during the 2011 outbreak of dengue all the four Dengue virus (DENV) serotypes circulating simultaneously were observed. The aim of the study was to describe the clinical epidemiology of dengue in Manaus, the capital city of the state of the Amazonas, where all the four DENV serotypes were co-circulating simultaneously. Methodology Patients with acute febrile illness during the 2011 outbreak of dengue, enrolled at the Funda??o de Medicina Tropical Dr. Heitor Viera Dourado (FMT-HVD), a referral centre for tropical and infectious diseases in Manaus, were invited to participate in a clinical and virological descriptive study. Sera from 677 patients were analyzed by RT-nested-PCRs for flaviviruses (DENV 1–4, Saint Louis encephalitis virus-SLEV, Bussuquara virus-BSQV and Ilheus virus-ILHV), alphavirus (Mayaro virus-MAYV) and orthobunyavirus (Oropouche virus-OROV). Principal Findings Only dengue viruses were detected in 260 patients (38.4%). Thirteen patients were co-infected with more than one DENV serotype and six (46.1%) of them had a more severe clinical presentation of the disease. Nucleotide sequencing showed that DENV-1 belonged to genotype V, DENV-2 to the Asian/American genotype, DENV-3 to genotype III and DENV-4 to genotype II. Conclusions Co-infection with more than one DENV serotype was observed. This finding should be warning signs to health authorities in situations of the large dispersal of serotypes that are occurring in the world.

Abstract: Rocio (ROCV), Saint Louis encephalitis (SLEV) and West Nile (WNV) are Flavivirus (Flaviviridae) probably carried by birds and transmitted by Culex mosquitoes. We show here a review on infections of the central nervous system by St. Louis Encephalitis, Rocio and West Nile Flaviviruses in Brazil, 2004-2014. In the last 10 years, serologic surveys in horses showed high proportions of seropositive animals which point out that SLEV and ROCV have circulated infecting horses in west-central, southeast and other regions of Brazil and that WNV has been introduced into Brazil and circulates mostly in Pantanal region. However humans infected by WNV have not been reported. In the State of Sao Paulo: SLEV was isolated from a case clinically diagnosed as dengue in 2004; in 2006, 6 SLEV patients including 3 cases of menigoencephalitis were reported in the middle of a large epidemic of dengue type 3; and in 2008, 1 patient with acute febrile illness that was IgM-positive for dengue was found infected by SLEV by detection of the virus genome. In 2010, ROCV genome was detected in the cerebrospinal fluids of 2 patients from the northern region with meningoenchephalitis and also AIDS. This was the first report of infections by ROCV in the last 34 years and curiously, it occured more than 2000 km from where the virus was firstly found. It is necessary to improve the surveillance of SLEV, ROCV and WNV in Brazil.

Abstract: description of a case of acute acquired ocular toxoplasmosis following hantavirus pulmonary syndrome. a 41-year-old man presenting hantavirus pulmonary syndrome, confirmed in the laboratory by detection of igm antibodies to the virus, was submitted to high doses of intravenous corticosteroids for two months. after clinical improvement of hantavirus pulmonary syndrome the patient presented visual loss in both eyes that was secondary to a toxoplasmosis retinitis. the retinitis resolved with anti-toxoplasma therapy. acquired toxoplasmic retinochoroiditis can occur following steroid therapy for hantavirus pulmonary syndrome.
Detection of Brazilian hantavirus by reverse transcription polymerase chain reaction amplification of N gene in patients with hantavirus cardiopulmonary syndrome

Abstract: we report a nested reverse transcription-polymerase chain reaction (rt-pcr) assay for hantavirus using primers selected to match high homology regions of hantavirus genomes detected from the whole blood of hantavirus cardiopulmonary syndrome (hcps) patients from brazil, also including the n gene nucleotide sequence of araraquara virus. hantavirus genomes were detected in eight out of nine blood samples from the hcps patients by rt-pcr (88.9% positivity) and in all 9 blood samples (100% positivity) by nested-pcr. the eight amplicons obtained by rt-pcr (p1, p3-p9), including one obtained by nested-pcr (p-2) and not obtained by rt-pcr, were sequenced and showed high homology (94.8% to 99.1%) with the n gene of araraquara hantavirus. although the serologic method elisa is the most appropriate test for hcps diagnosis, the use of nested rt-pcr for hantavirus in brazil would contribute to the diagnosis of acute hantavirus disease detecting viral genomes in patient specimens as well as initial genomic characterization of circulating hantaviruses.
Efeito do hipotireoidismo induzido na resposta superovulatória em novilhas de corte mesti as = Induced hypothyroidism on the superovulatory response in crossbred beef heifers

Abstract: Estudou-se o efeito do hipotireoidismo induzido sobre a resposta ovariana à superovula o, à atividade do corpo lúteo e ao ganho de peso em novilhas de corte mesti as ( Bos TaurosxBos Indicus). Nas análises estatísticas realizadas, n o foi observada a influência dos tratamentos sobre a varia o de peso (P>0,05). Os níveis de T3, T4 e TSH foram, respectivamente, 1,31+0,77, 16,71+0,44 e 10,35+5,74 para o grupo tratado e 2,08+0,77, 57,46+0,44 e 2,08+5,74 para o controle (P<0,05). O número de embri es viáveis e de estruturas totais colhidas foi, respectivamente, de 2,52 e de 4,86 nos animais hipotireóideos e de 2,67 e 5,60 nos controles, n o havendo diferen a (P>0,05). Desse modo, concluiu-se que o hipotireoidismo n o afetou a resposta superovulatória, a atividade do corpo lúteo e a varia o de peso. This study was aimed at the effect of hypothyroidism induced on the ovarian superovulation response, corpus luteum activity, and the weight gain in crossbred heifers ( Bos taurosxBos indicus). The data showed that the females weight remained the same (P>0.05) even after the treatment. The levels of T3, T4 e Tsh values were respectively1.31+0.77, 16.71+0.44 and TSH 10.35+5.74 to treated animals, and 2.08+0.77, 57.46+0.44, 2.08+5.74 to control group (P<0.05). There was no difference between them (P>0.05) considering the number of viable embryos and total number of structures 2.52 and 4.86 for the induced animals, and 2.67 and 5.60 for the control group respectively. Therefore, the results demonstrated that the hypothyroidism did not affect the superovulatory response, the corpus luteum activity, or the weight variation.

Abstract: the effect of various concentrations and exposure times of two plant growth regulators on root formation of stem cuttings of `tahiti' lime (citrus latifolia tanaka) was evaluated. additional trials evaluated and compared root formation efficiency of stem cuttings of two sweet orange cultivars (citrus sinensis l. osbeck). the research was carried out in piracicaba, sp, brazil, in an screen-house with an intermitent water spray system, using 10 cm long, five month old terminal semihardwood stem cuttings, without the three basal leaves. the experimental design consisted of randomized blocks, with four replications in all trials. data was collected 90 days after the beginning of each trial. the parameters analysed were: average number of roots per rooted cutting, percentage of rooting, percentage of callus induction, average length of the longest root, and average number of leaves per rooted cutting. the results showed that: a) rooting of `tahiti' lime cuttings was high, above 50%, reaching up to 96% in some cases, with no effect of the application of any plant growth regulator; b) sweet orange cuttings presented low percentage of root formation (around 0,5%), with no effect of the application of any plant growth regulator; c) there were no significant differences among the treatments for all the other parameters; d) the present study showed that plant propagation of `tahiti' lime through stem cuttings is an efficient and fast propagation method for this citrus species.

Abstract: Avaliou-se o efeito de várias concentra es e tempos de exposi o de dois reguladores de crescimento no enraizamento de estacas de lima ácida `Tahiti' (Citrus latifolia Tanaka). Experimentos complementares também avaliaram a eficiência do enraizamento de estacas de laranjas `Pêra' e `Valencia' (Citrus sinensis L. Osbeck) comparando-a com a da lima ácida `Tahiti'. Os trabalhos experimentais foram conduzidos, em Piracicaba, SP, em camara de nebuliza o intermitente, com estacas ponteiro, semi-lenhosas, coletadas 5 meses após a brota o, com 10 cm de comprimento, sem as 3 folhas basais. O delineamento experimental adotado foi o de blocos casualizados com 4 repeti es para todos os ensaios realizados. A avalia o dos tratamentos foi realizada 90 dias após a instala o de cada ensaio através da coleta dos seguintes dados biométricos (número médio de raízes/estaca enraizada-NMR, porcentagem de enraizamento-PE, porcentagem de forma o de calos-PC, comprimento médio da maior raiz-CMMR e número médio de folhas/estaca enraizada-NMF). Os resultados dos ensaios permitiram as conclus es: a) O enraizamento de estacas de lima ácida `Tahiti' ocorreu com relativa facilidade (PE acima de 50%, podendo chegar a 96%), independentemente da aplica o de reguladores de crescimento; b) Estacas de laranja doce revelaram baixa PE (0,5%), independentemente da utiliza o de reguladores de crescimento; c) Para cada espécie estudada, n o houve diferen as significativas para os parametros PC, NMR, CMMR e NMF. Observou-se que a obten o de mudas de lima ácida `Tahiti' por enraizamento de estacas é um método eficiente e rápido de propaga o para essa espécie.
Serosurvey of hantavirus infection in humans in the border region between Brazil and Argentina

Abstract: introduction: according to reports by the ministry of health, in the far western region of the state of santa catarina, there have been no reports of hantavirus pulmonary syndrome, a zoonotic disease transmitted by feces of infected rodents. a seroepidemiological study of residents of this region, was conducted, with the aim of determining the presence of hantavirus infections. a total of 340 volunteers of both genus, from the towns of belmonte and paraíso, were studied. methods: the serum of these patients was collected and used to detect igg antibodies against recombinant n protein of araraquara hantavirus, by elisa assay. the positive samples were then titrated and confirmed by immunofluorescence assay. results: this study demonstrated the presence of igg antibodies against hantavirus n protein in 3.5% of the population. the most frequent occupation was farm worker, 81% had direct and indirect contact with rodents, 91.7% of positive cases were farm workers, indicating that the probable cause of infection occurred during barn cleaning. these antibodies are noteworthy, given that the levels of antibodies were verified in individuals whose contact with hantavirus may have occurred many years ago. conclusions: this study shows the circulation of hantavirus in the region, a fact that until now, had not reported. all the serum reagents had contact with the pathogen, but did not develop pulmonary and cardiovascular syndrome. it is important to remain alert, because hantavirus is a serious and emerging disease of some relevance.
