Use of radiation in strains of Saccharomyces cerevisiae: A new technique for industrial applications  [PDF]

Abstract:
During the industrial fermentation process in the production of fuel ethanol, yeasts are subject to several stressing conditions. The survival and the permanence of strains introduced in the process correlate with the capability of these yeasts in resisting to physical and chemical stresses, as well as their recovering ability to compete with contaminating micro-organisms commonly present in this industrial process. We aim at the selection of Saccharomyces cere visiae strains having this capability and ability. In this sense, cultivations of strains with industrial interest were irradiated with gammas ray at a wide dose interval. Growing curves for the strains were analyzed by means of their relative growth, a new concept here introduced, which allows a better understanding of the growing and recovering processes following radiative stress. It was found that gamma radiation could be used as an alternative method to quantify growing capabilities of S. cerevisiae strains under stressing conditions. It was also shown that this radiological method could be utilized as an additional procedure to select best robust industrial strains. This radiological method simplifies traditional analysis of strain viability, by avoiding the great number of necessary and consecutive fermentation assays.

Abstract: six hundred and nine cases of meningitis occurred from july 1st to december 31st in the rio de janeiro municipal area; 311 were classified as bacterial and 298 as viral. the majority of the viral cases were part of the september-december outbreak caused by the echo-9 virus. three aspects are presented and discussed in this paper: first, the contradiction between the political and epidemiological significance of the disease; second, one of the main problems of the brazilian health information system - the lack of possible epidemic disease warnings to the public health authorities; and, finally, the importance given to the geographical rather than to the social aspects of disease distribution in the organization of health services. this last approach may distort the overall pattern of the disease and hence, may thwart the effectiveness of health actions set up to control it.

Abstract: this study was conducted in an area of 14 km in length oriented in the nne direction by 7.3 km wide, covering the totality of the serra do carambeígranite and adjacent units. located in the northern region of the alagados dam, paraná state, this granite contains relatively high concentrations of k, eu and eth, known since the 1970s through airborne and terrestrial gamma-ray spectrometry surveys. recent radiochemical tests conducted on 61 samples of geological materials such as rocks (17 samples) and material of the weathering mantle and alluvial deposits (44 samples), confirmed the occurrence of radioactive anomalies in this granite, especially in thorium and uranium. the contents of k, eu and eth obtained in gamma-ray spectrometry survey and radiochemical tests, converted to annual radiation dose (ard), allowed to evaluate the intensity of natural radiation, whose levels relatively high in certain regions of the study area, can offer hazard to the local populations. the analysis and interpretation of data, as well the preparation of contour maps of k, eu and eth were fundamental to understand the behavior and mobility of radionuclides in different environmental compartments of the area.

Abstract: nitrate in soils and plants can be determined by the method of ultraviolet absorption of nitrowyl chloride. the extracts of plants need to be treated with hydrogen peroxide before the determination in order to exidize the organic matter. for the soil extracts the blank is obtained by eliminating nitrates with hydrazine sulfate wich server to correct the absorption of the soil solutions. the determination is carried on adding perchloric, sulfuric and hudrochloric acids to the solutions obtained from vegetable and soil materials to form nitrowyl chloride. the absorbances of solutions are read at 230 miliraicrons.
Effect of conjugated equine estrogens and tamoxifen administration on thyroid gland histomorphology of the rat

Abstract: objective: the aim of this study was to evaluate the action of the conjugated equine estrogens and tamoxifen on the morphology of thyroid gland in ovariectomized (ovx) rats. methods: conjugated equine estrogens (cee), clinically used as estrogen therapy, is a complex formulation containing multiple estrogens that decrease menopausal symptoms. thirty ovariectomized rats were randomly divided into 3 treatment groups: gi, vehicle (propylene glycol); gii, cee 200 μg/kg per day; and giii, tamoxifen 1 mg/kg per day. another group of 10 rats with intact ovaries (giv) was included, treated with the vehicle, and sacrificed during estrous. all animals were treated by gavage for 50 days, after which they were sacrificed. blood samples were collected, and the thyroid was removed for morphological analysis and pcna evaluation through immunohistochemical study. results: the thyroid follicular cell height was increased in animals treated with cee (14.90 ± 0.20 μm), with tam (14.90 ± 0.10 μm), and in rats with intact ovaries (15.10 ± 0.50 μm) in comparison to that of the vehicle group (9.90 ± 0.20 μm) (p < 0.001). the follicular area was larger in the cee (2,225 ± 51 μm2) and tam (2,127 ± 67 μm2) groups compared to that of the vehicle group (5,016 ± 53 μm2). the levels of t4 and t3 in rats treated with cee, with tamoxifen and in rats with intact ovaries, were higher than those those in the vehicle group (p < 0.001). the pcna index in the vehicle group was lower than in other groups. conclusion: our data suggest that estrogen and tamoxifen administration has a proliferative effect on the thyroid.
Characterization of pseudarthrosis with ultrasound backscattered signals in rats

Abstract: purpose: to propose a novel model of pseudarthrosis in a small animal and to investigate the ability of backscatter parameters from ultrasound signals in differentiating normal bone from those ones with pseudarthorsis. methods: twelve rattus norvegicus albinus free from pathogenic species (spf) were randomly divided in two groups, with six animals each. in the control group a surgical approach to the femur was made, followed by the synthesis of the muscle and skin layers. the experimental group was submitted to an osteotomy of the femur and a vascularized flap of the fascia lata was interposed in the line of the fractured bone. then the alignment and bone stabilization were accomplished, by using nylon stitch in u shape introduced in holes made in the proximal and distal fractured bone. bone samples were scanned with ultrasound and signals were collected for each one to analyze the parameter apparent integrated backscatter - aib. results: radiological and anatomopathologic studies revealed the absence of bone consolidation with persistence of fiber-osteoid tissue. values of the ultrasound parameter aib from normal bones were statistically different from those with pseudarthrosis. conclusion: the experimental model was suitable for pseudarthrosis development in rats and the ultrasound backscatter parameters were able to identify such a bone disease in vitro.

Abstract: zygomycosis (mucormycosis) is a rare but highly invasive infection caused by fungi belonging to the order mucorales, which includes the genera rhizopus, mucor, rhizomucor, absidia, apophysomyces, saksenaea, cunninghamella, cokeromyces and syncephalastrum. this type of infection is usually associated with hematologic diseases, diabetic ketoacidosis and organ transplantation. the most common form of presentation is rhinocerebral mucormycosis, with or without pulmonary involvement. pulmonary zygomycosis is more common in patients with profound, prolonged neutropenia and can present as segmental or lobar infiltrates, isolated nodules, cavitary lesions, hemorrhage or infarction. the clinical and radiological manifestations are often indistinguishable from those associated with invasive aspergillosis. this article describes the general characteristics of pulmonary zygomycosis, emphasizing laboratory diagnosis, and illustrates the morphology of some lesions.

Abstract: the biology of trichogramma pretiosum riley (hymenoptera, trichogrammatidae) "strain bonagota" reared in eggs of bonagota salubricola (meyrick) (lepidoptera, tortricidae) was studied in laboratory estimating the thermal requirements and the number of annual generations in some apple production regions of the rio grande do sul and santa catarina states. eggs of b. salubricola were submitted to parasitism by t. pretiosum for 24h, and transferred to incubators regulated to 18, 20, 22, 25, 28, 30 and 32oc. the duration of the biological cycle (egg-adult) was influenced by temperature ranging from 8.18 to 18.12 days at 32 and 18°c, respectively. the parasitoid emergency was higher (66.2%) at 22°c. the sex ratio of t. pretiosum ranged from 0.61 to 0.72 being not influenced by temperature. the highest relation of parasitoids/egg was found at 30°c (1.41). the thermal constant (k) and the threshold temperature (tb) was 187.7 degrees days and 7.42oc, respectively. the number of annual generations estimated for t. pretiosum for fraiburgo and s?o joaquim, sc and vacaria, rs was 17.5; 11.6 and 16.8, respectively.

Abstract: red spider mite, panonychus ulmi (acari: tetranychidae) is a significant pest in apple tree in fraiburgo, santa catarina (sc) and applied biological control was implemented in the mid-nineties. the objective of this study was to demonstrate the economic benefits of biological control in the management of red spider mite. the assessment was carried out in two commercial orchards, one of which was subjected to biological control of spider mite by releasing the predator neoseiulus californicus, insecticide selection and weed management, and the other to conventional arthropod management, based on the application of chemicals to control insects, phytofagous mites and weed. economic analysis showed that the costs for labor and machines were similar in both orchards; however acaricide costs were significantly lower in the orchard subjected to biological control, demonstrating that despite the need for investment in predator mite breeding apparatus and maintenance costs, the biological strategy was economically viable.
Parasitism on Eriosoma lanigerum (Homoptera: Aphididae) by Aphelinus mali (Hymenoptera: Encyrtidae) on apple orchards, in Fraiburgo county, state of Santa Catarina, Brazil

Abstract: the parasitism of the woolly apple aphid (eriosoma lanigerum hausmann) by aphelinus mali (hald.) was evaluated in apple orchards cultivated with the varieties gala and fuji in fraiburgo county, state of santa catarina, southern brazil. colonies of the woolly apple aphid were evaluated during one year on 16 apple trees of each variety, starting in august 1999. the number of aphids at the mummy stage or presenting the orifice of the parasitoid emergence was recorded. results have shown that a. mali parasitized more than 50% of the woolly apple aphids and occurred during the four seasons. it was concluded that that under these conditions no chemical control against the aphid is necessary for the apple varieties gala and fuji.
