Abstract: Background Dengue includes a broad range of symptoms, ranging from fever to hemorrhagic fever and may occasionally have alternative clinical presentations. Many possible viral genetic determinants of the intrinsic virulence of dengue virus (DENV) in the host have been identified, but no conclusive evidence of a correlation between viral genotype and virus transmissibility and pathogenicity has been obtained. Methodology/Principal Findings We used reverse genetics techniques to engineer DENV-1 viruses with subsets of mutations found in two different neuroadapted derivatives. The mutations were inserted into an infectious clone of DENV-1 not adapted to mice. The replication and viral production capacity of the recombinant viruses were assessed in vitro and in vivo. The results demonstrated that paired mutations in the envelope protein (E) and in the helicase domain of the NS3 (NS3hel) protein had a synergistic effect enhancing viral fitness in human and mosquito derived cell lines. E mutations alone generated no detectable virulence in the mouse model; however, the combination of these mutations with NS3hel mutations, which were mildly virulent on their own, resulted in a highly neurovirulent phenotype. Conclusions/Significance The generation of recombinant viruses carrying specific E and NS3hel proteins mutations increased viral fitness both in vitro and in vivo by increasing RNA synthesis and viral load (these changes being positively correlated with central nervous system damage), the strength of the immune response and animal mortality. The introduction of only pairs of amino acid substitutions into the genome of a non-mouse adapted DENV-1 strain was sufficient to alter viral fitness substantially. Given current limitations to our understanding of the molecular basis of dengue neuropathogenesis, these results could contribute to the development of attenuated strains for use in vaccinations and provide insights into virus/host interactions and new information about the mechanisms of basic dengue biology.

Abstract: the pequi hulls is a waste of pequi processing, found in large volume in the brazilian cerrado, which may provide an alternative to ruminants' diet. this experiment was developed to evaluate the ruminal degradation kinects of dry matter (dm), crude protein (cp) and neutral detergent fiber (ndf) of pequi hulls bran (phb) and experimental diets, with different substitution levels of elephant grass (eg) by pequi hulls bran. four goats castrated with ruminal cannulas were used in a randomized complete block as experimental design. it was evaluated the ruminal degradation of phb, elephant grass and four diets with 0, 10, 20 e 30% of phb in substitution to elephant grass. the feeds were incubated in the rumen at times 4, 8, 12, 24, 48, 72 and 96 hours. the results showed that the phb had high degradation potential dm, cp and ndf, over 90, 80 and 80% respectively. the addition of the phb, replacing the eg, resulted in a higher degradability of dry matter and neutral detergent fiber, because the pequi waste had higher soluble and potentially degradable fraction of dm than elephant grass. the phb addition was negatively correlated with the soluble cp and with the potential and effective degradation of the cp, and positively correlated with insoluble potentially digestible cp. the eg substitution for phb allows better utilization of the diets, increasing the nutrient supplies to the animal.

Abstract: foodborne diseases have been deserving special attention by researchers of the whole world. viral etiology outbreaks of foodborne diseases have been improperly treated as less important than bacterial ones. rotavirus is the most reported cause of gastroenteritis and accounts for thousands of deaths annually. the present work reports a rotavirus-induced disease outbreak in a commercial establishment, where 51 employees related symptoms like diarrhea, vomits, nausea. water, food and beverages were analyzed microbiologically. no pathogenical bacterial contamination were detected, neither any modification on sensorial characteristics. according to the "attributable risk" of each food involved, bread and butter was considered suspicious food, and epidemiological approach pointed to the fact that the employee responsible for preparing this food was rotaviruses sicked just before the outbreak spreading.

This article uses some excerpts from the Genesis selected from adapted Bibles for children to discuss whether the choice for adaptations is due to the possibility the translator has to manipulate the biblical discourse differently from traditional Bibles and it also attempts to investigate whether her decisions are analogous with the biblical ideology.
