Abstract: Food irradiation has the purpose of destroying insects or microorganisms, thereby increasing the safety and shelf life of foods. Flavonoids are ubiquitous plant secondary products with radical scavenger ability. In the present study their antioxidant stability after gamma irradiation was evaluated. The flavonoids showed fast scavenger ability measured with the 2.2-diphenyl-1-picrylhydrazyl radical (DPPH) after high doses of radiation. The low damage caused by irra- diation on antioxidant capacity of the flavonoids shows their potential use in combating chemical oxidation of bio- molecules in irradiated foods.

Abstract: Lectin Cramoll-1,4, obtained from Cratylia mollis seeds (beans camaratu) was structurally characterized, biologically and pharmacologically, but its use as a biopharmaceutical is not well documented. The objective of this study is to propose a biopharmaceutical formulation lectin Cramoll-1,4, test their hemagglutinating properties in vitro as well as the use of gamma radiation as a continuous process of decontamination formulation. It was made of the extraction and purification Cramoll-1,4, was developed a gel formulation using Carbopol? as a vehicle, at concentrations of 50, 100, 200, 300 and 600 μg was irradiated with 60Co gamma rays in a dose of 7.549 kGy·h–1. The proposed formulation at a concentration of 300 μg produced an increase in the hemagglutinating units Cramoll-1,4 due to the synergistic effect caused by gamma radiation. Considering the diverse use of lectins, specific molecular and structural factors, as well as changes resulting from its formulation, concentration, irradiation and route of administration is of utmost importance to continue the studies in vitro, for subsequent application in vivo to characterize the physiological and molecular processes involved in the response and cellular effects.
Antimicrobial Activity of Cladonia verticillaris Lichen Preparations on Bacteria and Fungi of Medical Importance

Abstract: Cladonia verticillaris lichen lectin (ClaveLL) was purified using a previously established protocol and then evaluated for its potential antimicrobial activity. Initially, the autochthonous lichen was submitted to extraction with sodium phosphate buffer pH 7.0, followed by filtration and centrifugation to obtain crude extract. A salt fractionation was performed with 30% ammonium sulfate. After centrifugation, the protein fraction was loaded onto molecular exclusion chromatography using Sephadex G-100 matrix to purify active lectin. ClaveLL showed antibacterial activity against Gram-positive (Bacillus subtilis, Staphylococcus aureus, and Enterococcus faecalis) and Gram-negative (Escherichia coli and Klebsiella pneumoniae) assayed strains, with greater inhibitory effect on growth of E. coli (MIC of 7.18?μg?mL?1). The lowest minimum bactericidal concentration (MBC, 57.4?μg?mL?1) was detected against E. faecalis. The antifungal assay performed with Trichophyton mentagrophytes, Microsporum gypseum, Trichophyton rubrum, Trichosporon cutaneum, and Trichosporon asahi evaluated crude extract, fraction, and ClaveLL preparations. ClaveLL was the most active against T. rubrum with an inhibition percentage of 35% compared to negative control (phosphate buffer). Extract and fraction showed better activity on growth inhibition of T. mentagrophytes (35%). The results indicate the potential of ClaveLL and other C. verticillaris preparations as antimicrobial agents useful for applications focusing on human health. 1. Introduction The treatment of diseases caused by bacteria and fungi is becoming an issue of concern, due to the growing emergence of microorganism strains resistant to drugs and opportunistic fungi that cause serious infections in humans [1, 2]. Microbial resistance is a genetic phenomenon, in which the microorganisms have genes that encode biochemical mechanisms which prevent drug actions. It may be caused by mutations in the reproductive process of the microorganisms or by imported genes acquired through transduction, conjugation, and transformation mechanisms [3]. Microbial resistance is sometimes due to natural evolution of microorganisms; however, it gains importance through the excessive use of antimicrobial substances in medical, agricultural, and veterinary practices [4]. Oviposition-Stimulant and Ovicidal Activities of Moringa oleifera Lectin on Aedes aegypti

Abstract: Background Natural insecticides against the vector mosquito Aedes aegypti have been the object of research due to their high level of eco-safety. The water-soluble Moringa oleifera lectin (WSMoL) is a larvicidal agent against A. aegypti. This work reports the effects of WSMoL on oviposition and egg hatching of A. aegypti. Methodology/Principal Findings WSMoL crude preparations (seed extract and 0–60 protein fraction), at 0.1 mg/mL protein concentration, did not affect oviposition, while A. aegypti gravid females laid their eggs preferentially (73%) in vessels containing isolated WSMoL (0.1 mg/mL), compared with vessels containing only distilled water (control). Volatile compounds were not detected in WSMoL preparation. The hatchability of fresh eggs deposited in the solutions in the oviposition assay was evaluated. The numbers of hatched larvae in seed extract, 0–60 protein fraction and WSMoL were 45±8.7 %, 20±11 % and 55±7.5 %, respectively, significantly (p<0.05) lower than in controls containing only distilled water (75–95%). Embryos were visualized inside fresh control eggs, but not within eggs that were laid and maintained in WSMoL solution. Ovicidal activity was also assessed using stored A. aegypti eggs. The protein concentrations able to reduce the hatching rate by 50% (EC50) were 0.32, 0.16 and 0.1 mg/mL for seed extract, 0–60 protein fraction and WSMoL, respectively. The absence of hatching of stored eggs treated with WSMoL at 0.3 mg/mL (EC99) after transfer to medium without lectin indicates that embryos within the eggs were killed by WSMoL. The reduction in hatching rate of A. aegypti was not linked to decrease in bacteria population. Conclusions/Significance WSMoL acted both as a chemical stimulant cue for ovipositing females and ovicidal agent at a given concentration. The oviposition-stimulant and ovicidal activities, combined with the previously reported larvicidal activity, make WSMoL a very interesting candidate in integrated A. aegypti control.

Abstract: the incidence of deep venous thrombosis (dvt) in children (0-18 years old) is low. the aim of this study was to investigate the case of a 12 year-old child that had dvt after a trauma. protein c and protein s activities, antithrombin and resistance to activated protein c were analyzed in coagulometer. factor v leiden (fvl) was studied. the patient and his father were heterozygotes for fvl. his mother was normal homozygote. we concluded that the presence of fvl associated with other medical conditions tends to multiply the occurrence of thrombosis, which is a multifactorial disease.
Topical Application Effect of the Isolectin Hydrogel (Cramoll 1,4) on Second-Degree Burns: Experimental Model

Abstract: This study aimed at evaluating the use of hydrogel isolectin in the treatment of second-degree burns. Twenty male rats were randomly divided into two groups (G1 = treatment with hydrogel containing 100 μg/mL Cramoll 1,4 and G2 = Control, hydrogel). After 7, 14, 21, 28, and 35 days, animals were euthanized. On the 7th day, G1 showed intense exudates, necrosis and edema. On the 14th day, G1 showed tissue reepithelialization and moderate autolysis. On the 21st day, G1 showed intense fibroblastic proliferation, presence of dense collagen, and moderate fibrosis. On the 28th day, G1 showed complete tissue epithelialization. On the 35th day, G1 showed modeled dense collagen. The significant wound contraction was initiated from day, 14 in the G1. There were no significant differences in biochemical and hematological parameters analyzed. These results extend the potential of therapeutic applications for Cramoll 1,4 in the treatment of thermal burns.
Evaluation of Cytotoxic and Anti-Inflammatory Activities of Extracts and Lectins from Moringa oleifera Seeds

Abstract: Background The extract from Moringa oleifera seeds is used worldwide, especially in rural areas of developing countries, to treat drinking water. M. oleifera seeds contain the lectins cmol and WSMoL, which are carbohydrate-binding proteins that are able to reduce water turbidity because of their coagulant activity. Studies investigating the ability of natural products to damage normal cells are essential for the safe use of these substances. This study evaluated the cytotoxic and anti-inflammatory properties of the aqueous seed extract, the extract used by population to treat water (named diluted seed extract in this work), and the isolated lectins cmol and WSMoL. Methodology/Principal Findings The data showed that the aqueous seed extract and cmol were potentially cytotoxic to human peripheral blood mononuclear cells, while WSMoL and diluted seed extract were not cytotoxic. The M. oleifera aqueous seed extract and the lectins cmol and WSMoL were weakly/moderately cytotoxic to the NCI-H292, HT-29 and HEp-2 cancer cell lines and were not hemolytic to murine erythrocytes. Evaluation of acute toxicity in mice revealed that the aqueous seed extract (2.000 mg/kg) did not cause systemic toxicity. The aqueous seed extract, cmol and WSMoL (6.25 μg/mL) and diluted seed extract at 50 μg/mL exhibited anti-inflammatory activity on lipopolyssaccharide-stimulated murine macrophages by regulating the production of nitric oxide, TNF-α and IL-1β. The aqueous seed extract reduced leukocyte migration in a mouse model of carrageenan-induced pleurisy; the myeloperoxidase activity and nitric oxide, TNF-α and IL-1β levels were similarly reduced. Histological analysis of the lungs showed that the extract reduced the number of leukocytes. Conclusion/Significance This study shows that the extract prepared according to folk use and WSMoL may be non-toxic to mammalian cells; however, the aqueous seed extract and cmol may be cytotoxic to immune cells which may explain the immunosuppressive potential of the extract.
Economic Viability of Semi-Confined and Confined Milk Production Systems in Free-Stall and Compost Barn  [PDF]

Abstract: The objective of this research was to survey the productivity, costs, agricultural income and economic viability of Semi-Confined and confined milk production systems in Free-Stall and Compost Barn in Rio Grande do Sul, Brazil. In order to perform the research, four milk production units were selected in each of the three systems studied. Compared to the others, the Semi-Confined system had greater return of capital invested per year, as well as higher agricultural income per animal and per area, while the confined system in Compost Barn had higher farm incomes per man work unit and total farm incomes due to greater productive scale of properties. The Free-Stall system had the worst economic results. It was not possible to define an ideal system for the state, since the specificities of each property should be analyzed for choosing the system, especially the availability of the factors of production land, capital and labor.

Abstract: the objective of this study was to evaluate the effects of perennials legumes, in n, p and k foliar concentration and maize productivity in alley cropping system, without phosphorus fertilization. field experiments were carried out for two cycles, with legumes intercropping maize (uenf 506-8) in field research ccta/uenf in campos dos goytacazes - rj - brazil. the treatments consisted of alley cropping system with the species: albizia lebbeck (l.) benth., peltophorum dubium (spreng.) taub., leucaena leucocephala (lam.) de wit., cajanus cajan (l.) millsp., sesbania virgata (cav.) pers., mimosa caesalpiniaefolia benth., gliricidia sepium (jacq.) pers. and two control treatments: sole maize with or without fertilizer npk. eight months after the plantation, the legumes species were pruned and incorporated in soil. maize was sown interows legumes, in rows spaced 80 cm. new prune carried on sixty days after the first cut. in the second cycle, the crop handlings were similar to the first cycle. the experimental design consisted of randomized blocks with four replicates. in the alley cropping with cajanus cajan the maize show the larger n foliar concentration, in first cycle. in the second cycle, maize+gliricidia sepium, maize+cajanus cajan and sole maize with npk produced more grain than others treatments.
