Expression of the SIRT2 Gene and Its Relationship with Body Size Traits in Qinchuan Cattle (Bos taurus)

Abstract: Silent information regulator 2 (SIRT2) is a member of the sirtuin family of class III NAD (nicotinamide adenine dinucleotide)-dependent protein deacetylases and may regulate senescence, metabolism and apoptosis. The aims of this study were to investigate whether the SIRT2 gene could be used as a candidate gene in the breeding of Qinchuan cattle. Real-time polymerase chain reaction (RT-PCR) results showed that among all types of tissue that were analyzed, the highest mRNA expression levels of the gene were found in subcutaneous fat. DNA sequencing of 468 individual Qinchuan cattle identified two novel, single nucleotide polymorphisms (g.19501 C > T and g.19518 C > T) in the 3' untranslated region (3'UTR) of the SIRT2 gene. The frequencies of SNP g.19501 C > T and g.19518 C > T were in Hardy-Weinberg disequilibrium in all the samples (chi-square test, χ 2 < χ 0.05 2). An association analysis showed that the two loci were significantly correlated with some body size traits and the H 2H 2 (-CT-CT-) diplotypes performed better than other combinations. These results indicated that the variations in the SIRT2 gene and their corresponding genotypes may be considered as molecular markers for economic traits in cattle breeding.
Parametrized Post-Newtonian Orbital Effects in Extrasolar Planets

Abstract: Perturbative Post-Newtonian variations of the standard osculating orbital elements are obtained by using the two-body equations of motion in the Parameterized Post-Newtonian theoretical framework. The results obtained are applied to the Einstein and. Brans - Dicke theories. As a results, the semi-major axis and eccentricity exhibit periodic variation, but no secular changes.. The longitude of periastron and mean longitude at epoch experience both secular and periodic shifts. The Post-Newtonian effects are calculated and discussed for six extrasolar planets.
Orbit and spin evolution of the synchronous binary stars on the main sequence phase

Abstract: The sets of the synchronous equations are derived from the sets of non-synchronous equations The analytical solutions are given by solving the set of differential equations. The results of the evolutionary tendency of the orbit-spin are that the semi-major axis shrinks gradually with time: the orbital eccentricity dereacses gradually with time until the orbital circularization; the orbital period shortens gradually with time and the rotational angular velocity of primary component speed up with time gradually before the orbit-rotation achieved the circularization The theoretical results are applied to evolution of the orbit and spin of synchronous binary stars Algol A, B on the main sequence phase The circularization time and life time (age) and the evolutional numerical solutions of orbit and spin when circularization time are estimeted for Algol A, B. The results are discussed and concluded.

Abstract: 采用PCR—SSCP方法对186头24月龄秦川牛IGF2基因进行了SNPs多态性检测,并将其与部分胴体和肉质性状进行关联分析。在IGF2基因120碱基处发现C—T突变,在279碱基处发现A—G突变。方差分析结果表明:BB、DD两个位点与胴体性状中与宰前活重、胴体重、胴体长、胴体胸深、眼肌面积显著相关(p〈0.05),其中背部皮下脂肪厚达到差异极显著(P〈0.01);与肉质性状大理石花纹、嫩度、pIa24(牛肉排酸24h后的酸度值)显著相关(P〈0.05)。但是在胴体深、系水力指标中差异不显著(p〉0.05)。A、D等位基因是群体中的优势等位基因,AA、DD基因型是优势基因型,而含有B、D等位基因的个体的胴体和肉质性状优于其他个体,尤其有着极强脂肪沉积能力

Abstract: In this paper the effect of gravitational radiation damping on the variation of the orbital elements of binary system is studied using the method of perturbation. This effect results in both the secular and periodic variation of the semimajor axis and the eccentricity, however, the longitude of periastron exhibits no secular variation, but only periodic variation In addition, the deduced expression is applied to calculate the variation of secular and periodic terms of the orbital elements of binary system, Y cyg, PSR 1913 + 16 and PSR 2303 + 24, and the obtained results are discussed and compared with the relativistic effects.
Effect of Microsatellite Marker on Bull Meat Traits

Abstract: The aim of the present study was to verify associations between bull meat traits and 6 microsatellite loci. A sample of 66 weaned young bulls from Grassland Red Cattle (GLRC) and its improved hybrids by Limousine (LM) was characterized for BM2113, TGLA44, ETH225, IDVGA46, BM1824 and IDVGA55. The muscularity evaluation traits in the regulation of linear valuation and slaughtering meaty traits were used to evaluate their meat traits. The relationship between bull meat traits and 6 microsatellite loci was analyzed by SPSS. The results showed that for microsatellite BM2113, the allele 142 had a positive correlation with net meat weight and net meat rate; for microsatellite IDVGA46, the allele 211 had a negative correlation with the 5 muscularity evaluation and 4 slaughter meaty traits; for BM1824, the allele 171 had a positive correlation with Loin thickness; for microsatellite TGLA44, the allele 221 had a positive correlation with some muscularity evaluation traits such as withers, loin thickness, hip shape and some slaughtering meaty traits such as body weight, carcass weight and net meat weight.

Abstract: Retinoic X receptor-gamma (RXRG) gene was studied as a candidate gene for the twinning trait of bovine. A new SNP A1941G was detected by sequencing at 3'UTR. Different genotypes were determined in Luxi monotocous cows, Luxi twinning cows, Chinese Simmental cows, Angus cows and Simmental x Mongolia cows by restriction fragment length polymorphism (RFLP). The value of polymorphism information content indicated that this was a moderate polymorphism in Luxi monotocous cows and Luxi twinning cows. The chi(2) test indicated that the polymorphic locus in Luxi twinning cows did not fit Hardy-Weinberg equilibrium (Plt;0.05). The chi(2) test of associational analysis between genotypic distribution and twinning or monotocous trait in Luxi cows showed that the difference was very significant (Plt;0.01).

Abstract: The mitochondrial DNA 12S rRNA in seven Chinese local yellow cattle breeds, one cultivated breed and three imported breeds was studied by single-strand conformation polymorphism (SSCP). Three haplotypes (I, II , III) were detected, with haplotype I found only in one cattle of the Jinnan breed. Haplotype II did not appear in the foreign cattle breeds and low in Chinese-Simmental breed but was rich in Chinese local breeds. Haplotype III had the widest distribution and was present in ten cattle breeds. The average polymorphism information content (PIC) for Chinese cattle between 0.232-0.423 was modest, which suggested relative abundance of genetic resources for the Chinese native cattle.
SGO1 Maintains Bovine Meiotic and Mitotic Centromeric Cohesions of Sister Chromatids and Directly Affects Embryo Development

Abstract: Shugoshin (SGO) is a critical factor that enforces cohesion from segregation of paired sister chromatids during mitosis and meiosis. It has been studied mainly in invertebrates. Knowledge of SGO(s) in a mammalian system has only been reported in the mouse and Hela cells. In this study, the functions of SGO1 in bovine oocytes during meiotic maturation, early embryonic development and somatic cell mitosis were investigated. The results showed that SGO1 was expressed from germinal vesicle (GV) to the metaphase II stage. SGO1 accumulated on condensed and scattered chromosomes from pre-metaphase I to metaphase II. The over-expression of SGO1 did not interfere with the process of homologous chromosome separation, although once separated they were unable to move to the opposing spindle poles. This often resulted in the formation of oocytes with 60 replicated chromosomes. Depletion of SGO1 in GV oocytes affected chromosomal separation resulting in abnormal chromosome alignment at a significantly higher proportion than in control oocytes. Knockdown of SGO1 expression significantly decreased the embryonic developmental rate and quality. To further confirm the function(s) of SGO1 during mitosis, bovine embryonic fibroblast cells were transfected with SGO1 siRNAs. SGO1 depletion induced the premature dissociation of chromosomal cohesion at the centromere and along the chromosome arm giving rise to abnormal appearing mitotic patterns. The results of this study infer that SGO1 is involved in the centromeric cohesion of sister chromatids and chromosomal movement towards the spindle poles. Depletion of SGO1 causes arrestment of cell division in meiosis and mitosis.

Abstract: DNA samples from 60 Qinchuan cattle (Bos taurus) were analyzed with PCR-RFLPs and sequencing for insulin-like growth factor binding protein 3 (IGFBP3) gene. Fragments of 651 bp were amplified with two primers and the products of PCR were digested with restriction endonuclease HaeIII. The produced fragments showed three genotypes, namely AA,AB and BB after electrophoresis. Frequencies of the genotype AA,AB,BB and allele A,B were 0.7,0.28,0.02,and 0.84,0.16,respectively. Sequence analysis showed that a transversion of C-->A at 299 nt resulted in loss of the cleaved site of restriction endonuclease HaeIII and produced this polymorphism. This polymorphic locus of IGFBP3 gene was at Hardy-Weinberg equilibrium (P>0.05). The genotypes of AA,AB,BB slightly affected several slaughter and carcass traits of Qinchuan cattle. Dressing percentage,net meat percentage, striplion percentage, tenderloin percentage, ribeye percentage and tender shoulder percentage were decreased with the genotypes of AA,AB and BB in Qinchuan cattle, but it was not significant (P>0.05). Average ribeye area in individuals of AA genotype was significantly higher than that in individuals of BB genotype (P<0.05), and beef fat content in individuals of genotype AB and BB was significantly higher than that in individuals of AA genotype (P<0.01).
