MtsR, an Iron-Dependent Regulator in Streptococcus iniae  [PDF]

Abstract: Streptococcus iniae (S. iniae) is a major pathogen that is capable of resulting severe economic loss to cultured fish. Steadily iron availability from micro-environment is an important virulence factor for pathogens, and S. iniae encodes the iron-transporter MtsABC to accomplish heme utilization, but very little was known about the mechanisms involved in regulating and maintaining iron balance in S. iniae. In this study, the role of a putative iron-dependent transcriptional regulator MtsR was investigated, and the results showed that MtsR regulated the expression of iron-transport mtsABC to control iron homeostasis in S. iniae.
A solute-binding protein for iron transport in Streptococcus iniae

Abstract: An ABC transporter system, named as mtsABC (metal transport system) was cloned from S. iniae HD-1, and was found to be involved in heme utilization. mtsABC is cotranscribed by three downstream genes, i.e., mtsA, mtsB, and mtsC. In this study, we cloned the first gene of the mtsABC transporter system (mtsA), and purified the corresponding recombinant protein MtsA. The analysis indicated that MtsA is a putative lipoprotein which binds to heme that can serve as an iron source for the microorganism, and is expressed in vivo during Kunming mice infection by S. iniae HD-1.This is believed to be the first report on the cloning the ABC transporter lipoprotein from S. iniae genomic DNA. Together, our data suggested that MtsA is associated with heme, and is expressed in vivo during Kunming mice infection by S. iniae HD-1 which indicated that it can be a potential candidate for S. iniae subunit vaccine.Streptococcus iniae (S. iniae) is a hemolytic Gram-positive coccus that is a major pathogen of culture fish. It has been associated with disease outbreak in several species of freshwater and marine fish cultured worldwide, including tilapia [1,2], barramundi [3], channel catfish [4], hybrid striped bass [1,5], Japanese flounder [6,7], olive flounder [8], rabbitfish [9], and rainbow trout [9,10]. Streptococcal infection can lead to serious symptoms such as meningoencephalitis and generalized septicaemia with high mortality rates of up to 50% [9,11]. S. iniae is also known to be an opportunistic pathogen that can cause fulminant soft tissue infection in humans, such as bacteremic cellulitis, septicarthritis, and endocarditis [12]. Abstract: Skopos theory for a long time remains the core of Functional School of translation. The major principle of Skopos theory is that translation should be processed from the perspective of the intended audiences. Its major theory structure makes it as the guiding principle of business English translation which requires communicative and social textual equivalence. Based on the analysis of Skopos theory and business English translation features, this paper focuses on developing business English translation strategies, particularly in the case of advertisement translation. It aims to play a positive role in promoting the research and practice of business English translation.

Abstract: The functional theory of translation, with Skopostheorie as a core, advocates that the translation process should not only be based upon equivalence but more importantly upon the prospective functions or Skopos of the target text. Translation errors study was also put forward and carried out as a result. Based on the explanation of this translation theory, this paper makes a thorough analysis of business English translation errors, with the major reasons raised and corresponding solutions and strategies presented. Besides shedding lights on the theoretical study of translation in this field, the analysis aims to play a positive role in improving the substantial quality of business English translation in practice.

Abstract: Compared with the in-place pile, the pore-forming pouring pile is more simple and convenient, with a wider range of construction. In the actual construction process, it is able to pass through complex bottom layer and water layer underground without very high requirements in equipment. The actual bearing capacity of single pile is very strong, so that it can be better to adapt to the actual needs of different scales or the different geological conditions in building. And it has been promoted and used greatly in building construction work [1]. This paper introduces the concept of the pore-forming pouring pile technology, analyzes the pore-forming construction technology and the pile construction technology, then talks about prevention problems of the pore-forming pouring pile construction in House Building Project, at last draws a conclusion that the pore-forming pouring pile technology is the most basic construction technology and is the most effective and convenient way of construction.

Abstract: After the Opinions of the State Council on Solving Problems of Migrant Workers is carried out, the migrant workers’ situation has been improved, especially that their income has been increased greatly, the problem of wages in arrears has been abated, and the services provided by the government have been enhanced. However, some basic problems such as the migrant workers’ low income and difficult life have not been changed fundamentally. Thus, in order to carry out all of the policies of solving migrant workers’ problems, we should promote the institutional innovation, increase related investment, and gradually eliminate the institutional and financial obstacles of the citizenization of migrant workers; only in these ways can we solve the migrant workers’ problems.
Differential expression of DKK-1 binding receptors on stromal cells and myeloma cells results in their distinct response to secreted DKK-1 in myeloma

Abstract: Primary multiple myeloma (MM) cells were isolated from BM mononuclear cells of 12 MM patients. Human bone marrow stromal cells (SCs) were obtained from BM adherent cells of these MM patients and 10 healthy donors. The mRNA expression levels of DKK-1 binding receptor LRP5/6 and Kremen1/2 (Krm1/2) were analyzed by Real-time PCR in human myeloma cell line (HMCL) RPMI-8226, NCI-H929, U266, LP-1, CZ-1, KM-3, Sko-007, primary myeloma cells and SCs from 12 MM patients and SCs from 10 healthy donors. The binding capability of DKK-1 binding receptors to DKK-1 on primary myeloma cells and SCs was detected by flow cytometry assay.The mRNA expression levels of DKK-1 binding receptor LRP5/6 and Krm1/2 in SCs from patients with MM were significantly higher than those in myeloma cells and in SCs from healthy donors. The binding capability to DKK-1of DKK-1 binding receptors on SCs from MM patients was obviously higher than those on myeloma cells and SCs from healthy donors by flow cytometry assay. Similar to the effects of coculture with rhDKK1, coculture of SCs from healthy donors with myeloma cells in the presence or absence of a Transwell insert did up-regulate SCs' mRNA levels of LRP5/6 and Krm1/2, and down-regulate their mRNA levels of β-catenin.Compared with myeloma cells, the SCs from MM patients overexpress DKK-1 binding receptors LRP5/6 and Krm1/2 in response to DKK-1 secreted by myeloma cells, which results in intracellular Wnt signaling inhibition.
