Ceramide Mediates Ox-LDL-Induced Human Vascular Smooth Muscle Cell Calcification via p38 Mitogen-Activated Protein Kinase Signaling

Abstract: Vascular calcification is associated with significant cardiovascular morbidity and mortality, and has been demonstrated as an actively regulated process resembling bone formation. Oxidized low density lipoprotein (Ox-LDL) has been identified as a regulatory factor involved in calcification of vascular smooth muscle cells (VSMCs). Additionally, over-expression of recombinant human neutral sphingomyelinase (N-SMase) has been shown to stimulate VSMC apoptosis, which plays an important role in the progression of vascular calcification. The aim of this study is to investigate whether ceramide regulates Ox-LDL-induced calcification of VSMCs via activation of p38 mitogen-activated protein kinase (MAPK) pathway. Ox-LDL increased the activity of N-SMase and the level of ceramide in cultured VSMCs. Calcification and the osteogenic transcription factor, Msx2 mRNA expression were reduced by N-SMase inhibitor, GW4869 in the presence of Ox-LDL. Usage of GW4869 inhibited Ox-LDL-induced apoptosis in VSMCs, an effect which was reversed by C2-ceramide. Additionally, C2-ceramide treatment accelerated VSMC calcification, with a concomitant increase in ALP activity. Furthermore, C2-ceramide treatment enhanced Ox-LDL-induced VSMC calcification. Addition of caspase inhibitor, ZVAD-fmk attenuated Ox-LDL-induced calcification. Both Ox-LDL and C2-ceramide treatment increased the phosphorylation of p38 MAPK. Inhibition of p38 MAPK by SB203580 attenuated Ox-LDL-induced calcification of VSMCs. These data suggest that Ox-LDL activates N-SMase-ceramide signaling pathway, and stimulates phosphorylation of p38 MAPK, leading to apoptosis in VSMCs, which initiates VSMC calcification.

Structural OH in mantle-derived clinopyroxene megacrysts from Nushan

Abstract: Thirteen clinopyroxene megacrysts from Nushan Cenozoic basalts have been investigated by Fourier transform infrared spectrometer(FTIR). The results demonstrate that all samples contain trace amount of structural OH. Clinopyroxenes from Nushan show four groups of absorption bands, in which, 3 472–3 506 cm 1 is the major characteristic one. It is suggested that the position of major band is probably related to AI content: the position of major characteristic band for high-AI samples is around 3 470 cm 1, whereas that for low-AI samples around 3 620 cm 1.

AF17 Facilitates Dot1a Nuclear Export and Upregulates ENaC-Mediated Na+ Transport in Renal Collecting Duct Cells

Abstract: Our previous work in 293T cells and AF17-/- mice suggests that AF17 upregulates expression and activity of the epithelial Na+ channel (ENaC), possibly by relieving Dot1a-AF9-mediated repression. However, whether and how AF17 directly regulates Dot1a cellular distribution and ENaC function in renal collecting duct cells remain unaddressed. Here, we report our findings in mouse cortical collecting duct M-1 cells that overexpression of AF17 led to preferential distribution of Dot1a in the cytoplasm. This effect could be blocked by nuclear export inhibitor leptomycin B. siRNA-mediated depletion of AF17 caused nuclear accumulation of Dot1a. AF17 overexpression elicited multiple effects that are reminiscent of aldosterone action. These effects include 1) increased mRNA and protein expression of the three ENaC subunits (α, β and γ) and serum- and glucocorticoid inducible kinase 1, as revealed by real-time RT-qPCR and immunoblotting analyses; 2) impaired Dot1a-AF9 interaction and H3 K79 methylation at the αENaC promoter without affecting AF9 binding to the promoter, as evidenced by chromatin immunoprecipitation; and 3) elevated ENaC-mediated Na+ transport, as analyzed by measurement of benzamil-sensitive intracellular [Na+] and equivalent short circuit current using single-cell fluorescence imaging and an epithelial Volt-ohmmeter, respectively. Knockdown of AF17 elicited opposite effects. However, combination of AF17 overexpression or depletion with aldosterone treatment did not cause an additive effect on mRNA expression of the ENaC subunits. Taken together, we conclude that AF17 promotes Dot1a nuclear export and upregulates basal, but not aldosterone-stimulated ENaC expression, leading to an increase in ENaC-mediated Na+ transport in renal collecting duct cells.

Abstract: Conserved Ser354, Ser357 in the seventh transmembrane domain of NET were mutated and the function of mutants was studied by3H-NE influx measurement. Double mutation at these two serine sites reduced the activity of NET to base level. It is inferred that these two sites may be involved in the recognition and transport activity of the NET protein.
CD44+/CD105+ Human Amniotic Fluid Mesenchymal Stem Cells Survive and Proliferate in the Ovary Long-Term in a Mouse Model of Chemotherapy-Induced Premature Ovarian Failure

Abstract: Objectives: Stem cell transplantation has been reported to rescue ovarian function in a preclinical mouse model of chemotherapy-induced premature ovarian failure (POF); however, maintaining the survival and self-renewal of transplanted seed cells in ovarian tissues over the long-term remains a troublesome issue. In this study we aimed to determine whether the CD44+/CD105+ human amniotic fluid cell (HuAFCs) subpopulation represent potential seed cells for stem cell transplantation treatments in POF. Materials and methods: The CD44+/CD105+ subpopulation were isolated from HuAFCs, cultured in vitro, and injected into a cyclophosphamide-induced mouse model of POF. Results: Under continuous subculture in vitro, CD44+/CD105+ cells proliferated rapidly and expressed high levels of the proliferative markers Ki67 and survivin, as well as high levels of a number of mesenchymal stem cell biomarkers. Moreover, when red fluorescence protein (RFP)-transduced CD44+/CD105+ HuAFCs were transplanted into the ovaries of POF mice, the cells could be detected by fluorescence microscopy up to three weeks after injection. Furthermore, the BrdUrd incorporation assay and immunofluorescent staining demonstrated that CD44+/CD105+ HuAFCs underwent normal cycles of cell proliferation and self-renewal in the ovarian tissues of POF mice over the long-term. Conclusions: The mesenchymal stem cell properties and long-term in vivo survival of CD44+/CD105+ HuAFCs make them ideal seed cells for stem cell transplantation to treat POF.

Abstract: Let $\Omega$ be a pseudoconvex domain with $C^2$-smooth boundary in $\mathbb CP^n$. We prove that the $\bar\partial-Neumann operator $N$ exists for $(p,q)$-forms on $\Omega$. Furthermore, there exists a $t_0>0$ such that the operators $N$, $\bar\partial^*N$, $\bar\partial N$ and the Bergman projection are regular in the Sobolev space $W^t (\bar{\Omega}) $ for $t 0$.

Abstract: In this paper, several widely applied fracture criteria were first numerically examined and the crack-tip-region Jintegral criterion was confirmed to be more applicable to predict fracture angle in an elastic-plastic multiphase material. Then, the crack propagation in an idealized dendritic two-phase Al-7%Si alloy was modeled using an elastic-plastic finite element method. The variation of crack growth driving force with crack extension was also demonstrated. It is found that the crack path is significantly influenced by the presence of α-phase near the crack tip, and the crack growth driving force varies drastically from place to place. Lastly, the simulated fracture path in the two-phase model alloy was compared with the experimentally observed fracture path.
