Abstract: BACKGROUND: Although most studies have focused on the hepatocytes, all the hepatic cells participate in the regenerative process, among them the stellate cells. The stellate cells are mesenchymal cells involved in local neurotransmission and paracrine regulation of several liver functions. Acute hepatic tissue loss promotes the proliferation and activation of stellate cells from a quiescent state to myofibroblast-like cells. AIM: Investigate the effects of antihypertensive agents on the stellate cell population during the liver regenerative phenomenon in rats. METHODS: Adult male Wistar rats received lisinopril, losartan, bradykinin, or saline solution in a proportional volume, intraperitoneally, before and after 70% partial hepatectomy. Animals from the experimental and saline groups were sacrificed at 36 hours after partial hepatectomy. The alpha-smooth muscle actin labelled stellate cells population was counted in the periportal and pericentral zones of the liver specimen. RESULTS: The labelled stellate cells were more numerous in the control group both in the periportal and pericentral zones at 36 hours after partial hepatectomy than at the other times. The population of stellate cells was significantly lower in the losartan group and higher in the bradykinin and lisinopril groups than in the control group. CONCLUSIONS: These results suggest that losartan can inhibit and bradykinin and lisinopril can stimulate the stellate cell population during liver regeneration in rats. These cells synthesize several substances to stimulate liver regeneration.

Abstract: O efeito de um inibidor da enzima conversora da angiotensina (lisinopril), de um antagonista do receptor da angiotensina II (losartan) e da bradicinina na popula o de células estreladas (CE) durante o fen meno regenerativo hepático foi estudado. Ratos machos Wistar receberam lisinopril, losartan, bradicinina ou solu o salina em volumes proporcionais, intraperitonealmente, antes e após hepatectomia parcial a 70% (HP). Cinco animais de cada grupo experimental e controle foram sacrificados sob anestesia com éter em 36 horas após a HP. A popula o de CE marcadas para alfa-actina de músculo liso foi estimada nas zonas periportal e pericentral das amostras hepáticas. A popula o de CE foi menor no grupo tratado com losartan, e maior nos grupos tratados com bradicinina e lisinopril que no grupo controle. Estes resultados sugerem que o losartan pode inibir, e a bradicinina e o lisinopril podem estimular a popula o de CE durante a regenera o hepática em ratos.
Vascular Relaxation Induced by C-Type Natriuretic Peptide Involves the Ca2+/NO-Synthase/NO Pathway

Abstract: Aims C-type natriuretic peptide (CNP) and nitric oxide (NO) are endothelium-derived factors that play important roles in the regulation of vascular tone and arterial blood pressure. We hypothesized that NO produced by the endothelial NO-synthase (NOS-3) contributes to the relaxation induced by CNP in isolated rat aorta via activation of endothelial NPR-C receptor. Therefore, the aim of this study was to investigate the putative contribution of NO through NPR-C activation in the CNP induced relaxation in isolated conductance artery. Main Methods Concentration-effect curves for CNP were constructed in aortic rings isolated from rats. Confocal microscopy was used to analyze the cytosolic calcium mobilization induced by CNP. The phosphorylation of the residue Ser1177 of NOS was analyzed by Western blot and the expression and localization of NPR-C receptors was analyzed by immunohistochemistry. Key Findings CNP was less potent in inducing relaxation in denuded endothelium aortic rings than in intact ones. L-NAME attenuated the potency of CNP and similar results were obtained in the presence of hydroxocobalamin, an intracellular NO0 scavenger. CNP did not change the phosphorylation of Ser1177, the activation site of NOS-3, when compared with control. The addition of CNP produced an increase in [Ca2+]c in endothelial cells and a decrease in [Ca2+]c in vascular smooth muscle cells. The NPR-C-receptors are expressed in endothelial and adventitial rat aortas. Significance These results suggest that CNP-induced relaxation in intact aorta isolated from rats involves NO production due to [Ca2+]c increase in endothelial cells possibly through NPR-C activation expressed in these cells. The present study provides a breakthrough in the understanding of the close relationship between the vascular actions of nitric oxide and CNP.

Abstract: objective: the use of medicinal plants has increased worldwide. therefore, it is necessary to know their possible effects in order to establish their correct use on human beings. taking it into account, the objective of this study was to assess the effects of the aqueous extract of hyptis pectinata leaves, popularly known as "sambacaitá" or "canudinho", on liver regeneration after partial hepatectomy in rats. methods: ten rats were divided into 2 groups: hp group, which underwent 67% hepatectomy after 4 days of distilled water administration; and hp100 group, which was submitted to partial hepatectomy and oral administration of 100 mg/kg of the aqueous extract of hyptis pectinata for the same period of time. all animals were sacrificed 24 hours after the surgical procedure. liver regeneration was evaluated by immunohistochemical staining for proliferating cell nuclear antigen (pcna) using the pc-10 monoclonal antibody. results: the hepatic regeneration index was significantly increased in hp100 group (p<0.05). conclusions: the present study shows that the aqueous extract of hyptis pectinata leaves can stimulate liver regeneration at 100 mg/kg.

Abstract: purpose: the aim of this study was to investigate alterations compatible with hepatic ischemia-reperfusion after bilioduodenal shunt (bd) in rats with obstructive jaundice (ob) . methods: thirty six animals were divided into 6 experimental groups: co1 and co2 - control groups, sham-operated (so) and evaluated 6 and 24 hours after, respectively; ob1 and ob2, - obstructive jaundice groups, sham-operated 15 days after bile duct ligature and evaluated 6 and 24 hours after so, respectively; dbd1and dbd2 - obstructive jaundice groups evaluated ,respectively, 6 and 24 hours after bd performed 15 days after bile duct ligature. the parameters evaluated were serum total bilirubin, aminotransferase activity (ast, alt), tnfa, liver mitochondrial functions and parenchymatous injury. results: bilirubin decreased while aminotransferase activity increased 6 hours after bd (p<0.01); tnfa determination at the 6th hour after bd was higher than the one at the 24th hour (p<0.05); oxygen consumption in states 3 and 4 remained elevated in the bd initial phase , and liver cell damage worsened 24 hours after bd. conclusion: the results demonstrated that surgical biliary decompression in obstructive jaundice is followed by alterations related to hepatic ischemia- reperfusion.
The effect of anti-hypertensive drugs on the obstructive pancreatitis in rats

Abstract: purpose: to investigate the effect of ace inhibitor, lisinopril and at1 blocker, losartan, on the obstructive pancreatitis in rat. methods: acute pancreatitis in rats (n=21) was induced for a common hepatic duct were ligated proximal to its entry into the pancreas and the common bile - pancreatic duct were also ligated near its junction with the duodenum, under ether anesthesia, after which the abdomen were closed. the animals was divided in tree groups, being two treated and control group. the animals was treated with losartan and lisinopril at the dose of 10μg/kg body weight per day, i.p., in a proportional volume, for five days, before and after treatement. results: the inflammation, collagen deposition in the pancreas of treated animals were smaller, suggesting that the use of antihypertensive agents interfered positively in the depletion of the injury of the pancreas. scythe showed a correlation between activity of pancreatic stellate cells (pscs) lower in treated animals when compared to control. conclusion: the pancreatic stellate cells strength are involved in collagen production during acute pancreatitis and why antihypertensive drugs such as lisinopril and losartan may possibly have beneficial effects in reducing pancreatic fibrosis in models of experimental obstructive pancreatitis.

Abstract: O presente estudo relata os efeitos da aplica o de LASER sobre fígados cirróticos de ratos Wistar. A cirrose hepática foi induzida através da ligadura do ducto biliar. Aproximadamente vinte e cinco dias após a cirurgia inicial, os animais foram submetidos à irradia o LASER com exposi o hepática direta. O experimento comp s-se da análise morfométrica dos constituintes do fígado após 24 horas da irradia o. Os resultados revelaram maior prolifera o vascular nos animais irradiados sem altera o dos demais componentes.
