Transcriptional regulation of metabotropic glutamate receptor 2/3 expression by the NF-κB pathway in primary dorsal root ganglia neurons: a possible mechanism for the analgesic effect of L-acetylcarnitine

Abstract: Activation of group II metabotropic glutamate receptors (mGlu2 and mGlu3) induces antinociception in several pain models in rodents [1-5]. Consistent with these studies, we have shown that L-acetylcarnitine (LAC), a drug clinically effective in the treatment of neuropathic pain of various origins [6-9], up-regulates the expression of mGlu2 in the dorsal root ganglia (DRG) and in the dorsal horn (DH) of the spinal cord [10,11]. This has challenged the previous view that LAC increases pain thresholds and relieves neuropathic pain by enhancing brain acetylcholine synthesis [12] or by increasing the trophism of peripheral nerves [13,14]. Consistent with the "mGlu2 hypothesis of LAC-induced analgesia," the mGlu2/3 receptor antagonist, LY341495, prevents LAC-induced analgesia in rodents [10]. Interestingly, LAC selectively enhances the expression of mGlu2 receptors and has no effect on the expression of mGlu3 receptors [10,15], although these two receptor subtypes are highly homologous and share similar functions in the CNS [16]. This suggests that the expression of mGlu2 and mGlu3 is differentially regulated and that unraveling the nature of this difference may lead to the identification of new targets for the treatment of neuropathic pain.Analysis of the 5'-region upstream of the coding sequence of the human GRM2 gene (encoding mGlu2) [GenBank: AB045011], using the Transcription Factor Binding Sites Database TRANSFACT and TFSEARCH, revealed the presence of many potential regulatory elements for transcription factors of the NF-κB family, including p50 and p65/Rel-A, and for the coactivator p300. In contrast, only one binding site for the NF-κB family protein, c-Rel, and no binding sites for p65/RelA and p300 have been described in the putative promoter region of the human GRM3 gene encoding mGlu3 [17]. Baryon and Sunyaev-Zel'dovich effect properties of MareNostrum and MultiDark simulated clusters (MUSIC)

Abstract: We report the first results of the MUSIC project. It consists of two data sets of resimulates clusters extracted from two large dark matter only simulations: Marenostrum Universe and Multidark. In total, the MUSIC contains more than 400 clusters resimulated with high resolution both with radiative and non-radiative physics included. Here we present the first results on the properties of the baryon content and the Sunyaev Zeldovich scaling relations.

Abstract: Since 2003, New European Union Member States have made large capital inflows, which led to a credit crunch and recession. Whether they are foreign direct investment, or banking flows, capital inflows ultimately affect GDP, depending on how they are invested. In the specialty literature, analysis of capital flows was done especially in terms of their structure, with a lack of analysis in terms of final destination of capital inflows. Therefore, we analyzed the effect of capital inflows on GDP in the New Member States of the European Union (Bulgaria, Czech Republic, Estonia, Hungary, Latvia, Lithuania, Poland, Romania and Slovakia) over the last economic cycle. Based on experiences of the new Member States during the recent boom and crisis, the paper studies the impact of capital inflows on GDP growth, inflows channeled to economic sectors, such as real estate and corporate investment sector. The results of this research tries to highlight the extent to which the final destination of capital flows is important for the evolution of GDP.

Abstract: Until recently, capital mobility was encouraged across national borders, because it was considered that such capital can seek the highest rate of return. However, recent global financial developments have shown that, due to contagion, the mobility of capital flows can cause severe financial imbalances. In the context of globalization, liberalization or maintaining controls on capital flows is a current topic, more debated by economists. This topic is very important, due to the impact of liberalization decision or maintaining controls on capital flows has on the overall macroeconomic framework. The paper analyzes the relationship between capital flowsa€ control and the income per capita, the degree of central bank independence, democracy country, the foreign exchange regime. Also, it analyzes the effectiveness in time of capital controls, taking account of financial system development and potential risks of instability. Over time, it was observed that a period in which they have imposed restrictions on capital movements was followed by a removal of such restrictions, and vice versa. Cyclic change of capital movements regime corresponds to the cyclic evolution of the global economy. Full capital account liberalization led to the emergence of currency and financial crises, so that the idea of maintaining controls on capital is not rejected by economists. After a full liberalization of capital flows, there is a change in the mentality of an increasing number of economists, who support the maintenance of controls, in a gradual liberalization.
An R Package for Dynamic Linear Models

Abstract: We describe an R package focused on Bayesian analysis of dynamic linear models. The main features of the package are its flexibility to deal with a variety of constant or time-varying, univariate or multivariate models, and the numerically stable singular value decomposition-based algorithms used for filtering and smoothing. In addition to the examples of "out-of-the-box" use, we illustrate how the package can be used in advanced applications to implement a Gibbs sampler for a user-specified model.
A Bayesian framework for functional time series analysis

Abstract: The paper introduces a general framework for statistical analysis of functional time series from a Bayesian perspective. The proposed approach, based on an extension of the popular dynamic linear model to Banach-space valued observations and states, is very flexible but also easy to implement in many cases. For many kinds of data, such as continuous functions, we show how the general theory of stochastic processes provides a convenient tool to specify priors and transition probabilities of the model. Finally, we show how standard Markov chain Monte Carlo methods for posterior simulation can be employed under consistent discretizations of the data.
X-ray calibration of SZ scaling relations with the ACCEPT catalogue of galaxy clusters observed by Chandra

Abstract: We explore the scaling relation between the flux of the Sunyaev-Zel'dovich (SZ) effect and the total mass of galaxy clusters using already reduced Chandra X-ray data present in the ACCEPT (Archive of Chandra Cluster Entropy Profile Tables) catalogue. The analysis is conducted over a sample of 226 objects, examining the relatively small scale corresponding to a cluster overdensity equal to 2500 times the critical density of the background universe, at which the total masses have been calculated exploiting the hydrostatic equilibrium hypothesis. Core entropy (K0) is strongly correlated with the central cooling time, and is therefore used to identify cooling-core (CC) objects in our sample. Our results confirm the self-similarity of the scaling relation between the integrated Comptonization parameter (Y) and the cluster mass, for both CC and NCC (non-cooling-core) clusters. The consistency of our calibration with recent ones has been checked, with further support for Y as a good mass proxy. We also investigate the robustness of the constant gas fraction assumption, for fixed overdensity, and of the Yx proxy (Kravstov et al. 2007) considering CC and NCC clusters, again sorted on K0 from our sample. We extend our study to implement a K0-proxy, obtained by combining SZ and X-ray observables, which is proposed to provide a CC indicator for higher redshift objects. Finally, we suggest that an SZ-only CC indicator could benefit from the employment of deprojected Comptonization radial profiles.
Biased total mass of cool core galaxy clusters by Sunyaev-Zel'dovich effect measurements

Abstract: The Sunyaev Zel'dovich (SZ) effect from galaxy clusters is one of the most powerful cosmological tools for investigating the large-scale Universe. The big advantage of the SZ effect is its redshift independence, which is not the case for visible and X-ray observations. It allows us to directly estimate the cluster's total mass from the integrated comptonization parameter Y, even for distant clusters. However, not having a full knowing intra-cluster medium (ICM) physics can affect the results. By taking self-similar temperature and density profiles of the ICM into account, we studied how different ICM morphologies can affect the cluster total mass estimation. With the help of the high percentage of cool core (CC) clusters, as observed so far, the present analysis focuses on studying this class of objects. A sample of eight nearby (0.1 < z < 0.5) and high-mass (M > 10^(14) M_sun) clusters observed by Chandra was considered. We simulated SZ observations of these clusters through X-ray derived information and analyzed the mock SZ data again with the simplistic assumption of an isothermal beta-model profile for the ICM. The bias on the recovered cluster total mass using different sets of assumptions is estimated to be 50% higher in the case of hydrostatic equilibrium. Possible contributions to the total bias due to the line-of-sight integration and the considered ICM template are taken into account. The large biases on total mass recovery firmly support, if still necessary, cluster modeling based on more sophisticated universal profiles as derived by X-ray observations of local objects and hydrodynamical simulations.
Atmospheric monitoring in the mm and sub-mm bands for cosmological observations: CASPER2

Abstract: Cosmological observations from ground at millimetre and sub-millimetre wavelengths are affected by atmospheric absorption and consequent emission. The low and high frequency (sky noise) fluctuations of atmospheric performance imply careful observational strategies and/or instrument technical solutions. Measurements of atmospheric emission spectra are necessary for accurate calibration procedures as well as for site testing statistics. CASPER2, an instrument to explore the 90-450 GHz (3-15 1/cm) spectral region, was developed and verified its operation in the Alps. A Martin-Puplett Interferometer (MPI) operates comparing sky radiation, coming from a field of view (fov) of 28 arcminutes (FWHM) collected by a 62-cm in diameter Pressman-Camichel telescope, with a reference source. The two output ports of the interferometer are detected by two bolometers cooled down to 300 mK inside a wet cryostat. Three different and complementary interferometric techniques can be performed with CASPER2: Amplitude Modulation (AM), Fast-Scan (FS) and Phase Modulation (PM). An altazimuthal mount allows the sky pointing, possibly co-alligned to the optical axis of the 2.6-m in diameter telescope of MITO (Millimetre and Infrared Testagrigia Observatory, Italy). Optimal timescale to average acquired spectra is inferred by Allan variance analysis at 5 fiducial frequencies. We present the motivation for and design of the atmospheric spectrometer CASPER2. The adopted procedure to calibrate the instrument and preliminary performance of the instrument are described. Instrument capabilities were checked during the summer observational campaign at MITO in July 2010 by measuring atmospheric emission spectra with the three different procedures.
