Finasteride Inhibits Human Prostate Cancer Cell Invasion through MMP2 and MMP9 Downregulation

Abstract: Introduction The use of the 5-alpha reductase inhibitors (5-ARIs) finasteride and dutasteride for prostate cancer prevention is still under debate. The FDA recently concluded that the increased prevalence of high-grade tumors among 5-ARI-treated patients must not be neglected, and they decided to disallow the use of 5-ARIs for prostate cancer prevention. This study was conducted to verify the effects of finasteride on prostate cell migration and invasion and the related enzymes/proteins in normal human and tumoral prostatic cell lines. Materials and Methods RWPE-1, LNCaP, PC3 and DU145 cells were cultivated to 60% confluence and exposed for different periods to either 10 μM or 50 μM finasteride that was diluted in culture medium. The conditioned media were collected and concentrated, and MMP2 and MMP9 activities and TIMP-1 and TIMP-2 protein expression were determined. Cell viability, migration and invasion were analyzed, and the remaining cell extracts were submitted to androgen receptor (AR) detection by western blotting techniques. Experiments were carried out in triplicate. Results Cell viability was not significantly affected by finasteride exposure. Finasteride significantly downregulated MMP2 and MMP9 activities in RWPE-1 and PC3 cells and MMP2 in DU145 cells. TIMP-2 expression in RWPE-1 cells was upregulated after exposure. The cell invasion of all four tested cell lines was inhibited by exposure to 50 μM of finasteride, and migration inhibition only occurred for RWPE-1 and LNCaP cells. AR was expressed by LNCaP, RWPE-1 and PC3 cells. Conclusions Although the debate on the higher incidence of high-grade prostate cancer among 5-ARI-treated patients remains, our findings indicate that finasteride may attenuate tumor aggressiveness and invasion, which could vary depending on the androgen responsiveness of a patient’s prostate cells.

Abstract: Psychotherapeutic practice in hospitals in Brazil started in the 1950s even before the profession of psychologist in the country was formalized, and has been building a fairly rich history because it is differentiated according to the development of the practice over the decades as a function of the various clinical methods associated with it, as a consequence of the policies and/or different theoretical orientations that have underpinned it. Twenty years after this practice began, during the 1970s, the field of mental health was configured as a major center of absorption of psychologists in an attempt to change the prevailing medical model. It is at that time that we can first see the formation of multidisciplinary teams linked to psychotherapy treatments. When we examine the relationship between the advancement of the practice and its theoretical foundations, in a field where political orientations toward the practice carried no less weight, we realize that this entire movement has led to important developments in the history of psychology in general. In order to specify what clinical practice in hospitals has contributed to psychology in general, it is necessary to follow its guidelines over the decades, where we can confirm that a certain prevalence of psychoanalysis has been and is being established. To the extent that it is characterized as a discourse directed at the subject, it is fundamental that it allows for situating the work in the hospital in an exchange with the medical field in such a way that each field may sustain its own particularities, respect their differences, and contribute, each in their own way, toward the advancement of clinical work. From the 1950s through today, clinical practice highlights a concern with asserting subjectivity in hospitals where the contributions of some psychoanalytical developments to different forms of psychotherapy have not been lacking.

Abstract: this paper highlights some aspects regarding the supply of pre-school education in brazil, these last 10 years, bearing in mind the goals of the 2001 national plan for education (pne). after the period characterized by the implementation of policies aimed at integrating creches (daycare center) and pre-schools into the teaching systems, in line with the legal goals proposed after the national education law of guidelines and basis (ldben, 1996), we have perceived permanencies, redefinitions and tensions. the elements to draw up the new pne, deliberated within the conference on national education (conae), last april, resize the goals for this stage of basic education, with a more general purpose of establishing bases for a new federative pact aimed at the development of a national system of education. we wonder if the goals suggested by the conae for the education of little children consider the tendencies of access coverage observed these last 10 years and if they promote investments in the perspective of the legal goals that have been developed these last two decades.

Abstract: Spin-lattice and spin-spin relaxation times are one of the most attractive tools in the solid-state nuclear magnetic resonance spectroscopy to evaluate the level of clay dispersion in the nanocomposite matrices. The efficiency of the relaxation processes can be used to evaluate the nanoparticles intermolecular interactions and, consequently, the dispersion of them in the polymer matrix, the molecular dynamic of the hybrid compounds, as well as the molecular domains formation in an organic material. The determination of relaxation parameters was carried out to evaluate the organoclay exfoliation and intercalation process in the polymeric matrix, in addition to their dispersion and distribution in the matrix. The proton NMR relaxation data showed that the polymeric nanomaterials investigated presented good intermolecular interaction that promoted good nanoparticles dispersion and distribution in the hybrid materials. The proportion of 2% clay promoted a greater heterogeneity in the matrix compared to other ratios; 1% clay influenced only to the higher molecular rigidity phase; and 3% clay had a decrease in heterogeneity compared to 2% though still influenced the matrix as a whole. These results prove the efficiency of NMR technique in the evaluation of nanofillers interaction with polymer matrices, as well as their dispersion and distribution.

Abstract: the use of disqualifies ginger rhizomes as raw material for fresh fruit market is a way to allow the increase of ginger production . the starch is not directly fermentable, which requires prior hydrolysis to obtain glucose. this study aimed to evaluate the effect of concentrations of α-amylase and amyloglucosidase on the yield and sugar profile in the process of hydrolysis-saccharification of ginger. the process followed the central composite design for two factors, totaling 11 treatments. the results showed the effect of concentrations of α-amylase (termamyl 2x) and amyloglucosidase (amg 300l) on the glucose content and amyloglucosidase concentration effect on the content of dextrin. the higher yields of hydrolysis of the ginger suspension were obtained under conditions of high concentration of -amylase and amyloglucosidase.

Abstract:
Green coconut fiber (Cocos nucifera L.) has been largely used in solid-phase extraction. The effect of the initial dye concentration (1.91, 3.02 and 4.02) × 10-5 mol·L-1 and solid phase contact (SPE) time were evaluated at different temperatures (283, 298 and 313) K at pH 2. Kinetic experimental data were applied to three simplified kinetic models: pseudo-first order, pseudo-second order, and intraparticle diffusion. The adsorption of 5.5’-DI onto the solid phase showed excellent fit to the pseudo-second order model. At 283, 298 and 313 K the maximum sorption, qmax,exp, for the lowest initial concentration (Co = 1.91 × 10-5 mol·L-1) of 5.5’-DI were (5.01, 5.24 and 6.14) × 10
