Otimiza??o da técnica da PCR para a detec??o de Actinobacillus pleuropneumoniae

Abstract: the use of molecular methods based on pcr is important in actinobacillus pleuropneumoniae detection, being able to identify the infection before the establishment of the disease in the herd. these methods have larger sensitivity when compared with traditional methods of bacteriological isolation, but they can suffer influence of substances that reduce the specificity of the test and resulting in inespecific amplifications. in order to reduce inespecific amplifications, observed when applied the pcr technique for the gene cpx in tonsil's tissue samples, the optimization was performed, in which different annealing temperatures were analyzed and introduced, in the technique, an antibody that binds to the enzyme taq dna polimerase, increasing its specificity. in parallel, an experiment was performed in order to verify the inhibiting effect of the tonsil's tissue on the pcr results. for that, portions of tonsil from animals negative to the a. pleuropneumoniae were artificially contaminated with the reference sample of the sorotype 5b. the addition antibody for the enzyme taq dna polimerase and the increase of the primers anneling temperature to 57°c reduced the inespecific amplifications. the results obtained in the experiment demonstrated a possible inhibiting effect of the tonsil's tissue in the pcr amplifications. besides, amplifications depend on at least 675 ufc present in the aliquot of samples that will be used in pcr (equivalent to 1.35 x 105 ufc ml-1), therefore, samples tissue's fragments in initial infections and/or with few cells can result in false-negative.

Abstract: swine pleuropneumonia is one of the most important pig respiratory diseases and has been found in all producer countries. for control and monitoring of pleuropneumonia, it is necessary the development of fast and specific methods of diagnosis. to validate pcr based on the cpx gene of actinobacillus pleuropneumoniae in positive pigs, an experimental infection with a. pleuropneumoniae serotype 5b was performed and samples were obtained by tonsil swab, tonsil biopsy and blood for pcr, bacterial isolation and elisa, respectively. these tests were then performed in naturally infected pigs from three herds with different sanitary situations of clinical disease. in each herd, five groups of different ages were analyzed. tonsil biopsy for bacterial isolation and pcr and blood to determine the herd serological status was collected. the results obtained in the experimental infection confirmed that, even with the infection establishment, proved with bacterial isolation, it was not possible to detect the agent by pcr 45 days after infection. in naturally infected animals, pcr was more sensitive than bacterial isolation. the association between pcr and elisa is a good alternative to define the herd sanitary status regarding the infection with a. pleuropneumoniae.
Pharmacopuncture with acepromazine to tranquilize swine/ Farmacopuntura com acepromazina para tranquiliza o de suínos

Abstract: In order to tranquilize swine with acepromazine subdose in yin tang acupoint (located in the medium point of a line joined lateral cantus of the eyes in frontal region) was made this study. It was delineated in four steps in different days with five pigs. In first step (S1) acepromazine was administrated at 0.03mg/kg by intramuscular route. In second step (S2), the same drug was administrated at 0.003mg/kg at same route. In the third step (S3), acepromazine was administrated in yin tang acupoint at same dose utilized in S2. In the last step (S4) acupuncture needle was introduced in yin tang acupoint. Twenty minutes after S1, all animals presented spontaneous decubitus, palpebral ptosis, and sleepiness. In S2, only one animal had spontaneous decubitus and three showed palpebral ptosis, and sleepiness. In S3 all animals had palpebral ptosis, and sleepiness, and four spontaneous decubitus. In S4 occurred spontaneous decubitus, and sleepiness in all animals, two had palpebral ptosis. In conclusion, acepromazine at sub-dose in yin tang acupoint tranquilizes pigs. The acupuncture needle introduction in yin tang acupoint tranquilizes pigs too, but with technical difficulties because the needle must remain in acupoint for the necessary time. Foi realizado um estudo para tranqüilizar suínos com subdose de acepromazina no acuponto yin tang, localizado no ponto médio frontal de uma linha tra ada entre os cantos laterais dos olhos. O estudo foi delineado em quatro etapas, realizadas em dias diferentes, utilizando-se cinco suínos. Na primeira etapa (E1) administrou-se acepromazina por via intramuscular (IM), na dose de 0,03mg/kg. Na segunda etapa (E2) administrou-se acepromazina na dose de 0,003mg/kg, IM. Na terceira etapa (E3) foi administrada acepromazina no acuponto yin tang, na mesma dose empregada em E2. Na última etapa (E4) foi introduzida agulha de acupuntura no acuponto yin tang. Vinte minutos após E1, todos os animais apresentaram decúbito espontaneo, ptose palpebral e sonolência. Em E2, apenas um animal teve decúbito espontaneo, três apresentaram sonolência e ptose palpebral. Na E3, quatro animais tiveram decúbito espontaneo e todos apresentaram sonolência e ptose palpebral. Em E4 ocorreu decúbito espontaneo e sonolência em todos os animais; dois tiveram ptose palpebral. Concluiu-se que a administra o de acepromazina em subdose no acuponto yin tang tranqüiliza suínos. A introdu o da agulha no acuponto yin tang também tranquilizou os suínos, mas com dificuldades técnicas, pois a agulha deve permanecer no acuponto durante o tempo requerido para traquiliza

Abstract: O amido de arroz apresenta características favoráveis a muitas aplica es industriais; no entanto, a natureza hidrofílica do amido na forma nativa pode apresentar algumas limita es para determinados tipos de processamento. Neste estudo, amido de arroz com médio teor de amilose foi acetilado sob catálise alcalina em duas condi es reacionais para produzir acetatos de amido com diferentes graus de substitui o (GS). A introdu o de grupos acetila ao amido foi confirmada por espectroscopia de infravermelho com transformada de Fourier (FT-IV) e os acetatos de amido produzidos foram avaliados quanto às suas propriedades de pasta em viscoamilógrafo (RVA). A acetila o ocasionou redu es em todas as propriedades de pasta avaliadas (temperatura de pasta, viscosidade mínima, pico de viscosidade, viscosidade final e tendência à retrograda o), sendo a redu o mais intensa no amido acetilado com maior GS. Rice starch has characteristics suitable to many industrial applications, however, the hydrophilic nature of the starch in native form may present some limitations for some uses. In this study, rice starch with medium amylose content was acetylated under alkaline catalysis on two reaction conditions to produce starch acetates with different degrees of substitution (DS). The introduction of acetyl groups to the starch was confirmed by infrared Fourier transform (FT-IR) and starch acetates produced were evaluated for their paste properties in viscoelastograf (RVA). Acetylation caused reductions in all properties paste (paste temperature, minimum viscosity, peak viscosity, final viscosity and retrogradation tendency) being the most intense reduction in acetylated starch with greater GS.

Abstract: La tecnología como una expresión del progreso de la ciencia acompa a la evolución histórica de la humanidad, mostrándose cada vez más extraordinaria y abierta en las diferentes áreas del conocimiento. A pesar de que su inserción es cada vez mayor en las diversas formas de las relaciones entre los seres y de ellos con el medio ambiente, este es un tema que todavía necesita de mucha reflexión. Por ese motivo, nosotras nos propusimos cuestionar, reflexionar y aproximar los fenómenos de la "tecnología y el cuidado", no como una crítica a las tecnologías disponibles, sino más bien como una reflexión a cerca de las tecnologías del cuidado y sus perspectivas filosóficas. En este ejercicio reflexivo destacamos la responsabilidad ética como un imperativo moral a ser seguido por los profesionales del área de la salud, como participantes de la civilización del desarrollo tecnológico. La responsabilidad no presupone la bondad y la perfección del hombre donde, siendo el responsable, procurará siempre actuar para el bien, sino que también indica la capacidad individual de asumir con anticipación aquello que va a hacer, teniendo conciencia de todas las consecuencias de sus propias acciones y omisiones.
Revasculariza o miocárdica: estratégias para o enfrentamento da doen a e do processo cirúrgico Myocardial revascularization: strategies for coping with the disease and the surgical process

Abstract: OBJETIVO: Conhecer as estratégias utilizadas por pacientes no enfrentamento da doen a cardíaca coronária e do processo cirúrgico de revasculariza o miocárdica. MéTODOS: Utilizou-se a Teoria Fundamentada nos Dados como referencial metodológico para a releitura dos dados de um estudo mais amplo intitulado: Significando a experiência cirúrgica e o processo de viver do paciente submetido à revasculariza o do miocárdio. A coleta de dados foi realizada de outubro de 2010 a agosto de 2011, por meio de entrevista semiestruturada, com três grupos amostrais (pacientes, familiares de pacientes e profissionais de saúde) e 23 participantes. RESULTADOS: Os resultados apontam as estratégias utilizadas pelos pacientes submetidos ao processo cirúrgico de revasculariza o miocárdica, quais sejam: apoio familiar, espiritual e profissional. CONCLUS O: A experiência da cirurgia cardíaca modifica o processo de viver do paciente cardíaco e as estratégias utilizadas tornam a vivência menos traumática ao paciente. Logo, tais dados fornecem base teórica para o cuidado de enfermagem. OBJECTIVE: To understand the strategies used by patients in coping with heart coronary disease and surgical process of myocardial revascularization. METHODS: It was applied the Data analyses in Grounded Theory studies as a methodological reference to perform a re-reading of data from a larger study entitled as: "Signifying the surgical experience and the life process in patients undergoing myocardial revascularization". Data collection was carried out based on semi-structured interviews, with 23 participants (patients, their families and health professionals), from October, 2010 to August, 2011. RESULTS: The results showed the strategies used by patients undergoing myocardial revascularization in addressing the surgical procedure and coronary heart disease such as family, spiritual and professional support. CONCLUSION: The experience of cardiac surgery modifies the living process of cardiac patients and their strategies to let this experience less traumatic to them. Thus, such data have provided a theoretical basis for nursing care.
Biology of Triatoma klugi Carcavallo, Jurberg, Lent & Galv o 2001 (Heteroptera: Reduviidae) under Laboratory Conditions: Effects of Distinct Blood Sources and Susceptibility to Trypanosoma cruzi and Trypanosoma rangeli

Abstract: The life cycle of Triatoma klugi Carcavallo, Jurberg, Lent & Galv o 2001 was compared under laboratory conditions using two groups of the F1 generation obtained from field-collected bugs. Among the 100 nymphs weekly fed on mice (Group A) or chicken (Group B), 77% of Group A and 67% of Group B reached the adult stage, and the mean time from the first nymphal stage to adult was 190.08 ± 28.31 days and 221.23 ± 40.50, respectively. The average span in days for each stage per group and the number of blood meals required for each stage were also evaluated. The overall mortality rate was 23% and 33% for Groups A and B, respectively. The mean number of eggs laid per month in a three-month period was of 56.20, 51.70 and 73.20 for Group A, and 64.50, 53.50 and 38.71 for Group B. Despite the blood source, comparative analysis revealed no statistically significant differences in the life cycle of T. klugi under laboratory conditions. Infection rates over 60% were observed for both Trypanosoma cruzi strains tested. Even revealing high infection rates of the hemolymph by T. rangeli strains, T. klugi revealed no salivary gland infections and was not able to transmit the parasite.
