In Silico Pharmacokinetics Studies for Quinazolines Proposed as EGFR Inhibitors  [PDF]

Abstract: In silico pharmacokinetics studies can aid the search for molecules with potential ability to be drug candidates. In this paper, a number of quinazoline candidates for epidermal growth factor receptor inhibitors—EGFR, important targets for the treatment of cancer, are computationally analyzed. The literature described that 69 quinazoline molecules were synthesized and the respective half maximum inhibitory concentrations (IC50) were obtained. A bilinear parabolic model was built to investigate the druglikeness by correlating the corresponding lipophilicities, which can be represented by the ideal Log P , with the optimal biological activity in terms of pIC50 values. Structural characteristics leading to improved pharmacokinetics parameters were then analyzed. Compound 56 exhibited the lowest IC50 and, therefore, it had the highest ability to inhibit the EGFR. In the present work, the most potent inhibitor 56 is not calculated to be the most promising drug candidate, since it’s out of the parabolic model obtained due to a Log P above 5, which is not within the expected optimum range. Finally, this work is an example of computational prediction that an experimentally, highly active EGFR inhibitor can be unsuccessful as drug candidate because of pitfalls in pharmacokinetics parameters.

Abstract: the ethanolic extract of "guaco" (mikania laevigata) traditionally cultivated in the soil or in a hydroponic system were tested for allelopathic and antimicrobial activities. allelopathic activity was evaluated by the inhibition of germination assay using lettuce seeds, and antimicrobial activity by the disc diffusion assay. a notable allelopathic effect was observed for both extracts, although a more expressive activity of traditional "guaco" was verified, since the inhibition of seeds germination was 100% even in the lower concentration. none ethanolic extract of "guaco" presented significant antimicrobial activity against staphylococcus aureus, escherichia coli, pseudomonas aeruginosa, enterococcus faecalis and enterococcus faecium.
Sir Ernst Gombrich and the Barber from Tuscany

Abstract: In the spirit of Sir Ernst Gombrich, this essay uses an anecdote—a chat between Gombrich and a barber from Tuscany—to illustrate a deeper point, namely, how cultural memory, tradition, and a canon give rise to an implied language of culture and cultural value. Gombrich staunchly defended tradition against relativism. By relativism, he meant something like "radical subjectivism." To his mind, subjectivism (in the cultural and social sense of the term) is not only impossible, since meaning is conferred through culture and society, but it is also dangerous (in the cultural and ethical sense of the term), since it denies the existence of shared values. Against consensus on the one hand and radical subjectivism on the other, Gombrich advocated a middle way: criticism and self-criticism to ensure latitude; the search for "truth" to ensure a limited plurality of interpretations.

Abstract: Von den Sozialwissenschaften in Gro britannien wird immer st rker erwartet, dass sie sich nicht nur hinsichtlich wissenschaftlicher und intellektueller Kriterien durch Exzellenz auszeichnen. Vielmehr müssen sie ihre Relevanz auch für unterschiedliche Interessengruppen und Forschungsnutzer aufweisen. Dieses Klima des Wandels macht sich an einer noch nie da gewesenen Zahl an strategischen Initiativen fest, die derzeit laufen. Der Zweck des Artikels besteht darin, die verschiedenen strategischen Entwicklungen zu überblicken und aufzuzeigen, wie sie zur F rderung der avancierten Entwicklungen in der Methodologie der Sozialwissenschaften beitragen. Dabei liegt das Augenmerk besonders auf den Entwicklungen im Bereich der qualitativen Methodologien und Methoden. In einem Teil des Beitrags werden wir Ergebnisse einer Erhebung bei britischen Sozialwissenschaftler(inne)n über die Ressourcen qualitativer Forschung bieten. Der Artikel stellt sich zur Aufgabe, die expliziten und impliziten Ziele zu formulieren, die die so genannte "methodologische Wende" in den ffentlich gef rderten Sozialwissenschaften Gro britanniens im frühen 21. This "climate of change" is seen in the un-pre-ced-ented level of strategic initiatives that are currently under-way. The purpose of this article is to consider some of these strategic developments and how they seek to promote leading edge developments in social science methodology. Attention is focus-sed, in particular, on the place of qualitative method-ologies and methods within these develop-ments. One of the sections of the article reports on the main findings of a consultation exercise with UK social scientists on qualitative research resources. Summer sea-surface temperatures and climate events on the North Icelandic shelf through the last 3000 years

Abstract: Diatom data from core MD992271 on the North Icelandic shelf record a cooling trend through the last 3000 years. This is indicated by a general decrease in warm water species and an increase in cold water taxa. The relative abundance of these two diatom groups changed periodically, suggesting that the climate also fluctuated within this time period. The results of diatom-based transfer function calculation show that the summer sea-surface temperatures (SSTs) before 1400 cal. a BP were generally higher than the mean value for the last 3000 years and the summer SSTs fluctuated around the mean between 1400 and 700 cal. a BP, and dropped to the values below the mean after 700 cal. a BP. Four cooling events were distinguished, centered at around 2600, 1900, 1300 and 600 cal. a BP respectively. The results are not only consistent with the data from neighbouring cores HM107-03 and MD992275, but also comparable with those from the GISP2 ice core and from other marine sediment records in the North Atlantic. This suggests that changes in the summer SSTs reflect regional climate variations in the North Atlantic. On the North Icelandic shelf, the summer SST variation is a result of changes in the interaction between the cold and the warm currents in the area.

Abstract: 冰岛北部陆架MD992271孔沉积硅藻中喜暖硅藻数量持续减少以及喜冷藻类数量不断增加,反映了该区近3000年来气候变冷的趋势,而硅藻组合中这两类硅藻数量呈周期性的此消彼长,指示在趋冷的背景下,气候的冷暖交替过程.转换函数计算结果表明,该孔夏季表层海水温度变化大致可分3个阶段,在1400cal.aBP以前,温度基本高于平均值;在1400~700cal.aBP之间,温度在平均值上下起伏,而在700cal.aBP后,温度均处于平均值之下.在这3000多年间,还存在着4次明显的降温事件,降温事件的中心分别位于约2600,1900,1300和600cal.aBP.上述夏季表层海水温度变化和冰岛北部陆架HM107-03和MD992275孔资料相对比,明显具有一致性,同时也可与GISP2冰芯氧同位素及北大西洋深海沉积记录对比,说明冰岛北部陆架晚全新世夏季表层海水温度变化,受区域性气候变化的控制.直接导致上述海水温度变化,是影响该区暖流和寒流作用此消彼长的结果,而冰岛周边海域3000年来海流分布的基本格局未有明显改变.

A Comparative Review Over the Pension Systems’ Performance in Central and Eastern European Countries

Abstract: Towards the late 90’s, together with overcoming the shock of transition, most countries in Central and Eastern Europe began a process of profound reform of the pension systems. The aim of these reforms was mainly ensuring the financial sustainability of the pension systems often without taking into account their primary goal: to provide adequate retirement incomes, to allow the elderly to maintain a decent living standard after retirement and to have economic independence. However, a successful pension system is not the one that involves little spending but the one achieving its primary goal while limiting the future pressures on public finances.The economic crisis has increased the vulnerability of the pension systems. The economic recessions or slow economic growth, budget deficits and debt burdens, low employment rates have led to rising the concern about the ability of pension systems to fulfill their purpose. In this context, most Central and Eastern European countries have recently been forced to take up new changes in pensions. In our study we accomplished a comparative analysis of the pension systems’ performance based on calculating a composite indicator that includesthree components: the indicator of adequacy, the financial sustainability indicator and the indicator of modernity. Each of these components is obtained by aggregating a selection of sub-indicators. The purpose of this analysis is to identify best practices for successful reform, but also to highlight potentialhazards or imbalances that might face various emerging countries.
Stroma Regulates Increased Epithelial Lateral Cell Adhesion in 3D Culture: A Role for Actin/Cadherin Dynamics

Abstract: Cell shape and tissue architecture are controlled by changes to junctional proteins and the cytoskeleton. How tissues control the dynamics of adhesion and cytoskeletal tension is unclear. We have studied epithelial tissue architecture using 3D culture models and found that adult primary prostate epithelial cells grow into hollow acinus-like spheroids. Importantly, when co-cultured with stroma the epithelia show increased lateral cell adhesions. To investigate this mechanism further we aimed to: identify a cell line model to allow repeatable and robust experiments; determine whether or not epithelial adhesion molecules were affected by stromal culture; and determine which stromal signalling molecules may influence cell adhesion in 3D epithelial cell cultures.
