Informa??o & Informa??o , 2012, DOI: 10.5433/1981-8920.2011v16n3p96-117
Technique to Obtain Mitotic Chromosomes of Conyza bonariensis L. Cronquist (Asteraceae)  [PDF]
Juliana Maria de Paula, Cecília Alzira Ferreira Pinto-Maglio
American Journal of Plant Sciences (AJPS) , 2015, DOI: 10.4236/ajps.2015.69145
Abstract: Cytogenetic analysis requires cytological preparations that unequivocally reveal the chromosome number and permit optimal visualizations of chromosome morphology for the construction of karyotypes and ideograms. Chromosomal characterization is possible only by establishing these two parameters. To cytogenetically characterize the weed Conyza bonariensis (L.) Cronquist (Asteraceae), it was necessary to improve cytological analysis techniques to obtain optimal results. This species belongs to a genus whose plants have wide phenotypic plasticity. These plants can be morphologically differentiated by other types of analysis, and thus the application of this technique will serve as a reference for cytogenetical analysis of other groups of plants that have cytogenetic characteristics similar to those of C. bonariensis. The methodology described here highlights three main protocol steps: 1) root tip collection from newly germinated seed radicles and from young root tips of mature plants, 2) pretreatment of meristems with antimitotic agents, both isolated and combined, and 3) acid and enzymatic hydrolytic processes.

Abstract: This work aimed to evaluate methods to breaking seed dormancy of Brachiaria brizantha cv. Marandu. An experiment was carried out at the Laboratory of Seeds from Instituto Federal Goiano – Campus Rio Verde. A completely randomized design with eight treatments to breaking seed dormancy was used. The treatments were: 1) control; 2) immersion in gibberellic acid (100 mg L-1) for 48 h; 3) immersion in gibberellic acid (200 mg L-1) for 48 h; 4) immersion in concentrated sulphuric acid for 5 min; 5) immersion in concentrated sulphuric acid for 10 min; 6) immersion in hot water (100 oC) for 1 min; 7) immersion in hot water (100 oC) for 2 min and 8) immersion in hot water (100 oC) for 3 min with four replicates. Daily, germination was evaluated to determine the germination speed index. After the complete germination (25 days), the seed germination percentage was quantified. Hot water treatments, independently of the seed immersion time, were the most effective to breaking the seed dormancy of B. brizantha cv. Marandu. Marandu.

Abstract: The “cachaça” is currently one of the fastest growing agro-industrial industries in the country as the drink considered as a symbol of Brazilian nationality. This study aimed to quantify the phenolic compounds, evaluate the antioxidant and antibacterial activity of different samples of aged “cachaça”. The physic-chemical analysis was performed at the Brandy Quality Laboratory of the Federal University of Lavras, and the determination of phenolic compounds was performed by High Performance Liquid Chromatography (HPLC). The antimicrobial activity evaluation was held at the Food Mycology Laboratory through the cavity agar diffusion technique and by steam using the micro-organisms Escherichia coli, Staphylococcus aureus, Salmonella choleraesuis, Listeria monocytogenes and Pseudomonas aeruginosa. The antioxidant activity was evaluated using the DPPH radical sequestering method. The values obtained for the phenolic compounds ranged from 0.41 to 9.69 mg/L, observing the predominance of syringaldehyde, gallic acid and vanillic acid. The phenolic extracts showed a satisfactory antibacterial activity for both Gram-negative and Gram-positive but did not show inhibition against the bacteria Pseudomonas aeruginosa. The antioxidant activity was evidenced by the DPPH test for cachaças tested and the “cachaça” aged in oak barrel showed the highest activity, followed by “cachaça” aged in jequitibá, amburana and balm barrels.
