Multifunctional Polypeptide EQCN Sensors: Probing the Cysteamine-Glutathione Film Permeability with Hg(II) Ions

Abstract: Multifunctional films are the basis of biosensors and play an important role in the emerging field of nanobioelectronics. In this work, films of a tripeptide glutathione (GSH) immobilized on a self-assembled monolayer of cysteamine (CA-SAM) on a quartz crystal Au piezosensor have been synthesized and characterized using electrochemical quartz crystal nanogravimetry (EQCN) with a Hg(II) ion probe. It has been found that in contrast to previously studied Au/GSH films, the Au/CA-GSH films strongly hinder the formation of Hg0 with bulk properties while still allowing for relatively easy permeation by Hg(II) ions. This results in complete disappearance of the sharp Hg0 electrodissolution peak which is observed on bare Au and Au/GSH piezosensors. The multiple-peak anodic behavior of Au/CA and bare Au is replaced by a single high-field anodic peak of mercury reoxidation in the case of Au/CA-GSH sensors. The mass-to-charge plots indicate predominant ingress/egress of Hg(II) to/from the film. The strong hindrance of CA-SAM to bulk-Hg0 formation is attributed to film-stabilizing formation of surface (CA)2Hg2+ complexes with conformation evaluated by ab initio quantum mechanical calculations of electronic structure using Hartree-Fock methods. The associates CA-GSH provide an additional functionality of the side sulfhydryl group which is free for interactions, e.g. with heavy metals. It is proposed that in the film, the CA-GSH molecules can assume open (extended) conformation or bent hydrogen-bonded conformation with up to four possible internal hydrogen bonds.

Abstract: five different antigens from cysticercus cellulosae, a vesicular fluid, a saline and an alkaline total extracts, an escolex and membrane, were studied in the elisa immunoenzymatic assay to demonstrate igg antibodies in cerebrospinal fluid (csf) and serum samples. for the 5 antigens the 20mg/ml concentration was selected for polyvinyl plates sensitization for csf and serum assays. the igg fraction of a sheep anti-human igg antiserum was labeled with horseradish peroxidase and revealed with hydrogen peroxide-5-aminosalicilic acid. for positive results 10 elisa units for sera and 5 eu for csf were taken. a total of 182 serum samples and 115 csf samples were tested. the elisa sensitivity for sera were 88.6% for vesicular fluid, escolex and membrane; 85.7% for saline extract and 62.8% for alkaline extract. the elisa sensitivity for csf was 100% for vesicular fluid and saline total extract, 91.7% for membrane, 89.6% for excolex and 77.1% for alkaline. the elisa specificity for sera was 100% to the 5 antigens studied; to the csf was 100% for the alkaline and escolex and 98.5% for the other antigens. an elisa geometric mean titer of sera and csf was respectively the 121.1 and 27.3 for vesicular fluid, 111.6 and 31.1 for saline total extract, 30.3 and 5.3 for alkaline total extract, 63.2 and 14.8 for escolex and 69.1 and 18.8 for membrane antigen. elisa was then compared to immunofluorescence, hemagglutination and complement fixation tests in csf and sera. elisa immunoenzymatic assay with saline total extract is recommended for the easy preparation and for the high quantity of antigens obtained, for the high sensitivity and great specificity for sera and csf; we suggest that this test may be used as a substitute for immunofluorescence, hemagglutination and complement fixation in sera or csf for the diagnosis of neurocysticercosis.

Abstract: this study discusses the main themes and research related to thanatology, studies of death and dying. the pioneer experts who wrote the early works that systematize the area are presented: herman feifel, robert kastenbaum and elizabeth kübler-ross, as well as the main themes of study: bereavement, violence and war, death and tv, palliative care, training of health and education professionals to deal with people experiencing loss and death. further studies to develop thanatology in brazil are proposed.

Abstract: this article discusses fundamental questions concerning the end of life and the proximity of death. based on the bioethical approach, issues such as quality of life, dignity in the process of dying, and autonomy of choices regarding life in its final moments are discussed. the improvement of medical technology favoured the cure of many diseases and the prolongation of life. when exaggerated, though, it may add suffering to what is supposed to be a benefit, stimulating discussion about complex issues such as: the right to decide about one's own death, euthanasia, assisted suicide and dysthanasia. clarification and presentation of these topics from various angles are objectives of this article. the possibility of multidisciplinary discussion of the issues presented is also suggested.

Perguntas têm assoberbado a humanidade, e respostas foram trazidas pelas religi es, ciências, artes, filosofias, entretanto, nenhuma delas é completa e universal. é sobre esses questionamentos e reflex es, sobre a busca de sentido à vida que a morte pode oferecer, sobre essa característica ou qualidade humana de questionamentos, de autoconhecimento, que pretendemos falar. A educa o é entendida como desenvolvimento pessoal, aperfei oamento e cultivo do ser, que também pressup e uma prepara o para a morte, envolvendo comunica o, relacionamentos, perdas, situa es-limite, como, por exemplo: fases do desenvolvimento, perda de pessoas significativas, doen as, acidentes, até o confronto com a própria morte. Ser o discutidas várias propostas de educa o para a morte tanto para o público leigo quanto para os profissionais. Death is part of the human development since the first years of life and goes along with the human being throughout the existence, leaving its marks. How could we prepare people to this subject so important in life? This is a challenge even more urgent to health and education professionals. Many questions overwhelm Humanity and answers are given by religions, sciences, arts and phylosophy, but none is universal. We shall consider these questions and reflections, the search for meaning, about life that the questions about death promote. Education here is understood as personal development, self improvement, and involves communication, relationships, losses, limit situations such as: development phases, loss of close people, diseases, accidents and confronting own death. Several death education formats will be discussed including layman and health and professionals' education.

Abstract: Die M nnerforschung ist innerhalb der Gender Studies eine recht junge, aber in den letzten Jahren gleichwohl verst rkt ernstzunehmende Disziplin auch in Deutschland. Der vorliegende Sammelband gew hrt einen Einblick in gegenw rtige Forschungen zu M nnlichkeitskonstruktionen und -strategien in Bildung und Gesellschaft. Es wird aufgezeigt, wie vielf ltig, kritisch und aktuell die M nnerforschung jenseits von politischer Vereinnahmung ist. Gleichwohl wird auch ersichtlich, wie viel Nachholbedarf‘ es noch in der deutschsprachigen Forschung gibt. Men’s studies is still a rather young discipline, yet nevertheless has become increasingly serious in recent years, even in Germany. The anthology at hand offers insights into current research on constructions and strategies of masculinity in education and society. It shows how diverse, critical, and topical men’s studies can be beyond political monopolization. At the same time, however, it also shows how much ‘catching up’ still needs to be done in German-language research.
