Reverse Engineering the Neuroblastoma Regulatory Network Uncovers MAX as One of the Master Regulators of Tumor Progression

Abstract: Neuroblastoma is the most common extracranial tumor and a major cause of infant cancer mortality worldwide. Despite its importance, little is known about its molecular mechanisms. A striking feature of this tumor is its clinical heterogeneity. Possible outcomes range from aggressive invasion to other tissues, causing patient death, to spontaneous disease regression or differentiation into benign ganglioneuromas. Several efforts have been made in order to find tumor progression markers. In this work, we have reconstructed the neuroblastoma regulatory network using an information-theoretic approach in order to find genes involved in tumor progression and that could be used as outcome predictors or as therapeutic targets. We have queried the reconstructed neuroblastoma regulatory network using an aggressive neuroblastoma metastasis gene signature in order to find its master regulators (MRs). MRs expression profiles were then investigated in other neuroblastoma datasets so as to detect possible clinical significance. Our analysis pointed MAX as one of the MRs of neuroblastoma progression. We have found that higher MAX expression correlated with favorable patient outcomes. We have also found that MAX expression and protein levels were increased during neuroblastoma SH-SY5Y cells differentiation. We propose that MAX is involved in neuroblastoma progression, possibly increasing cell differentiation by means of regulating the availability of MYC:MAX heterodimers. This mechanism is consistent with the results found in our SH-SY5Y differentiation protocol, suggesting that MAX has a more central role in these cells differentiation than previously reported. Overexpression of MAX has been identified as anti-tumorigenic in other works, but, to our knowledge, this is the first time that the link between the expression of this gene and malignancy was verified under physiological conditions.

Abstract: objective. to update concepts related to feline hyperthyroidism. materials and methods. information from available literature from the last 50 years including the bbcs-lilacs, fuente académica, ib-psycinfo, ib-ssci, ib-scielo, scopus and scirus, databases as well as historical articles, texts and references cited in published papers was analyzed. results. pertinent information related to the objectives proposed in the present review was obtained and it can be classified in five sections as follows: etiology, physiopathology, clinical signs, diagnosis and management. conclusion. feline hyperthyroidism is a disease caused by high levels of circulating t4, t3 or both. it is the most common endocrine disorder in domestic cats and affects approximately 1 in 300 cats showing multisystem complications. currently, there are both medical and surgical treatments. medical therapy is a conservative alternative which given the appropriate facilities has an effectiveness of 90%. however, depending on the etiology of the disease, thyroidectomy may be more effective for example in the case of tumors.

Abstract: objective: to update concepts related to normal function of the thyroid gland in domestic felines and to review the basic concepts of feline hypothyroidism. materials and methods: literature available in the bbcs-lilacs data bases, fuente académica, ib-psycinfo, ib- ssci, ib-scielo, scopus and scirus, databases from the last 50 years, as well as historical articles, texts and references cited in works public to date were analyzed. results: pertinent information related to the objectives proposed in the present review was found and analyzed. as a consequence, the article is classified in three sections as follows: production and storage of thyroid hormones, functions of thyroid hormones, feline hypothyroidism. conclusion: the thyroid gland plays an important role as thyroid hormones producer which are necessary for cellular differentiation and organic growth. feline hypothyroidism is not frequent and it is classified according to the time of appearance in congenital way or acquired, and depending if the alteration site is primary, secondary or tertiary. depending on the severity of its presentation it can also be classified as clinic or sub-clinic forms.
Técnicas de pós-colheita e expans?o da cultura da manga no Estado de S?o Paulo

Abstract: the worldwide demand for mango has been increasing over the last few years. in brazil, the mango production for exportation has been showing a great possibility of growth, where the use of floral induction and post harvest techniques have helped to participate in new market shares while the supply from other countries has been reduced. in the last decade, the mango production and exportation have been increasing in the s?o paulo state. the aim of this work was to analyze the relation between the adoption of new techniques for improving the mango quality required for exportation and the production expansion in the s?o paulo state. the producer adaptation degree to the required consuming market norms was used as a parameter for the quality analysis of the produced mango and was performed in two towns of the state. the field survey has shown that there are regions in the state that are adapting themselves to the exportation requirements and this has been having a great influence on the production growth. in addition, this activity has shown to be considerably profitable for producers.

Abstract: morphological characterization of fruits and seeds provides subsidies for species differentiation and characterization of ecological aspects of plants, such as dispersion, seedling establishment and ecological succession phase . thereare few studies reported in regard to erythrina variegata l. and in the literature there is no information about its behavior, especially about its system of propagation in the state of espírito santo. the objective of this study was to investigate the morphology of the fruits, seeds and seedlings of this species, characterizing the fruits externally, the seeds internally and externally, and describing the process of development and differentiation of the phases of the seedlings, as well as collecting information regarding the seed germination, using a completely randomized experimental design with the following treatments: intact seeds (control); mechanical scarification with sandpaper number 120, on the opposite side to the embryo; scarification with sandpaper and posterior soaking in water at ambient temperature for 6, 12 and 24 hours; thermal shock; and soaking in water at ambient temperature for 12 and 24 hours. the seeds did not show themselves impermeable to water through their teguments, thus, not requiring pre-germination treatments.

Abstract: the objective of this study was to determine the optimum germination temperature for seeds of the erythrina variegata l. the experiments were carried out at the seed analysis laboratory at cca-ufes (laboratório de análise de sementes of cca-ufes) in alegre-es. the seeds were placed in plastic seed germination containers (gerbox), with sterilized sand as the substrate. random samples of 25 seeds each were incubated separately at constant temperature of 12oc, 14oc, 16oc, 18oc, 20oc, 22oc, 24oc, 26oc, 28oc, 30oc, 32oc, 34oc, 36oc, 38oc and 40oc, with 8-hour photoperiod. this procedure was repeated four times to obtain a meaningful result. it was found that e. variegata seeds have a broad range of temperature tolerance with seed germination observed between 14oc and 38oc. temperatures between 22oc and 36oc seemed most favorable with germination in excess of 67%. temperatures between 32oc and 34oc produced the optimum average rate and time for germination. the highest percentage of e. variegata seed germination was found to be at the temperature of 34oc.

Abstract: the objective was to study the morphology of the seeds and seedlings of dimorphandra wilsonii rizz. characterizing the seeds internally and externally, as well as describing the processes of development and differentiation of the seedlings phases. the soaking curve, the thousand-seed weight and the number of seeds per kilogram were also determined. one kilogram of the seeds corresponded to a number of 2.560 seeds in average. the seeds have endosperm, in which the tegmen is strongly attached and the teguments are hard and impermeable to water. the seed and seedling morphology gives informations that facilitates the recognition of this species.
Técnicas de pós-colheita e expans o da cultura da manga no Estado de S o Paulo

Abstract: A demanda por manga tem apresentado crescimento significativo no mercado internacional. No Brasil, a produ o de manga apresenta grande potencial de crescimento para exporta o. A utiliza o de técnicas de indu o floral e póscolheita tem permitido explorar brechas de mercado, no momento em que se reduz a oferta dos países concorrentes. O Estado de S o Paulo tem aumentado a produ o e a exporta o de manga na última década. O objetivo desse trabalho foi analisar a rela o entre a ado o de técnicas de melhoria da qualidade da manga produzida, exigidas na exporta o, e a expans o da cultura no Estado de S o Paulo. Para a análise da qualidade da manga produzida, utilizou-se como parametro o grau de adequa o do produtor às normas exigidas nos mercados consumidores, em duas cidades do Estado. A pesquisa de campo mostrou que a adequa o aos requisitos exigidos de qualidade tem conformado regi es no Estado de S o Paulo, onde o aumento de produ o está diretamente relacionado com as exporta es. Adicionalmente, verificou-se que essa atividade tem apresentado um retorno econ mico atraente aos produtores.

Abstract: Luiz Felipe Cunha Mattos (21/02/1956 – 04/12/2012) “Se antes ajudava a construir a história da Medicina de Família, hoje é parte fundamental desta história.” Luiz Felipe formou-se em Medicina pela Universidade de Passo Fundo (UPF-RS), desde este período apresentava interesse pela saúde pública. Nas apresenta es e atividades, quando perguntado sobre qual o professor que marcara sua forma o, sempre se referia ao seu professor de saúde coletiva da UPF. Formado em 1981, e contrariando a “moda” da época, realizou sua Residência em Medicina Geral e Comunitária na Unidade Sanitária S o José do Murialdo (Porto Alegre, 1982-1983), sendo colega de Airton Stein, Elizabeth Wartchow, Magda Costa, Maria Teresa Pinho e Cristina Lemos, os quais - juntamente com ele e Wanda Loguércio, José Mauro C. Lopes e Hermes Cattani - em 1984 iriam compor o grupo dos primeiros médicos contratados da Unidade de Medicina de Família do Hospital Nossa Senhora da Concei o (HNSC). Este fato teve dois aspectos inovadores: foi, talvez, a primeira contrata o de médicos de família como tais, e realizada através de concurso que envolveu um processo seletivo rigoroso (prova da Associa o Médica do Rio Grande do Sul (AMRIGS), entrevista e prova oral). Desde 26 de abril de 1984, Luiz Felipe atuou como médico da Unidade Concei o, “na ponta”, sem interrup es na atividade clínica, sendo um dos médicos de maior longitudinalidade no Brasil. Desde o início envolveu-se ativamente na constru o do Servi o de Saúde Comunitária, participando de sua expans o. Ocupou cargos como Chefe de Unidade, Preceptor e Supervisor da Residência em MFC e Coordenador da COREME do HNSC. Alguns destes cargos foram ocupados por vários mandatos. Uma de suas preocupa es constantes era com a qualifica o clínica do MFC, participando ativamente das discuss es e debates dos anos 80 e 90 sobre qual o perfil do MFC necessário para o Sistema de Saúde do Brasil. Um aspecto talvez desconhecido seja sua atua o - durante a fase da Reforma Sanitária – no Conselho Municipal de Saúde de Porto Alegre, como representante do HNSC, tendo feito parte de sua Camara Técnica. Fez especializa o em Saúde Pública pela Escola de Saúde Pública do RS, no final dos anos 80, tendo como tema de seu trabalho de conclus o “O paciente frequente”. Desde o início desempenhou importante papel nas atividades associativas, fazendo parte do grupo de MFCs que assumiu pela primeira vez o Departamento de Saúde Coletiva da Associa o Médica do Rio Grande do Sul (anos 80).
