Solubility enhancement of aggregation-prone heterologous proteins by fusion expression using stress-responsive Escherichia coli protein, RpoS

Abstract: We analyzed the Escherichia coli proteome response to the exogenous stress of guanidine hydrochloride using 2-dimensional gel electrophoresis and found that RpoS (RNA polymerase sigma factor) was significantly stress responsive. While under the stress condition the total number of soluble proteins decreased by about 7 %, but a 6-fold increase in the level of RpoS was observed, indicating that RpoS is a stress-induced protein. As an N-terminus fusion expression partner, RpoS increased significantly the solubility of many aggregation-prone heterologous proteins in E. coli cytoplasm, indicating that RpoS is a very effective solubility enhancer for the synthesis of many recombinant proteins. RpoS was also well suited for the production of a biologically active fusion mutant of Pseudomonas putida cutinase.RpoS is highly effective as a strong solubility enhancer for aggregation-prone heterologous proteins when it is used as a fusion expression partner in an E. coli expression system. The results of these findings may, therefore, be useful in the production of other biologically active industrial enzymes, as successfully demonstrated by cutinase.Escherichia coli have been widely used as a host to produce valuable commercial, industrial, and therapeutic proteins. There are several disadvantages with this system, however, especially for the expression of eukaryotic proteins. Lactobacillus brevis Strains from Fermented Aloe vera Survive Gastroduodenal Environment and Suppress Common Food Borne Enteropathogens

Abstract: Five novel Lactobacillus brevis strains were isolated from naturally fermented Aloe vera leaf flesh. Each strain was identified by Random Amplified Polymorphic DNA (RAPD) analysis and 16S rRNA sequence comparison. These strains were highly tolerant to acid, surviving in pH2.5 for up to 4 hours, and resistant to 5% bile salts at 37°C for 18 hours. Due to its tolerance to acid and bile salts, one strain passed through the gastric barrier and colonised the intestine after oral administration. All five strains inhibited the growth of many harmful enteropathogens without restraining most of normal commensals in the gut and hence named POAL (Probiotics Originating from Aloe Leaf) strains. Additionally, each strain exhibited discriminative resistance to a wide range of antibiotics. The L. brevis POAL strains, moreover, expressed high levels of the glutamate decarboxylase (GAD) gene which produces a beneficial neurotransmitter, γ-aminobutyric acid (GABA). These characteristics in all suggest that the novel L. brevis strains should be considered as potential food additives and resources for pharmaceutical research.
H pylori eradication: A randomized prospective study of triple therapy with or without ecabet sodium

Abstract: AIM: To investigate whether adding ecabet sodium to the standard triple therapy for H pylori infection improve eradication rate.METHODS: Two hundred and fifty-seven H pylori-infected patients were randomly assigned to standard triple therapy (group A, n = 129) or triple therapy plus ecabet sodium (group B, n = 128). Successful eradication was defined as a negative 13C-urea breath test 6-8 wk after completion of treatment.RESULTS: After completion of therapy, 194/257 patients showed negative 13C-urea breath test results. According to intention-to-treat analysis, the infection was eradicated in 93/129 (72.1%) patients in group A and 101/128 (78.9%) in group B (P = 0.204). Per-protocol analysis showed successful eradication in 93/118 (78.8%) patients from group A and 101/114 (88.6%) from group B (P = 0.044). There were no significant differences in the side effects experienced by the patients in the two treatment groups.CONCLUSION: Our results suggest that the addition of ecabet sodium improves the efficacy of the standard triple therapy for H pylori.

Abstract:

This study focuses on the geometry and kinematics of the Sinnyeong Fault which is the most conspicuous fault among the WNW-trending Gaeum Fault System in the Gyeongsang Basin,SE Korea. The fault is traced for over ca.70 kmand has a consistent WNW-trending strike with a nearly vertical dip. It has an asymmetric fault damage zone of several meters to several tens of meters in width and a several meter-thick fault core. Its main movement is interpreted as sinistral-reverse oblique-slip or sinistral strike-slip under NE-SW compressional stress regime, although it could have experienced other faultings with different senses before/after this movement. Cylindrical folds, having the NW-trending fold axes of low angle plunge, are only observed along the southern damage zone of the fault with a continuous narrow width of several tens of meters. It is thus interpreted that the formation of the folds and sinistral movement of the fault were almost contemporaneously generated due to the concentration of the regional NE-SW compressional stress along pre-existing WNW-trending faults or densely populated fracture zone in a relatively stable intraplate region.

Abstract: Which slopes can or cannot appear as Seifert fibered slopes for hyperbolic knots in the 3-sphere S^3? It is conjectured that if r-surgery on a hyperbolic knot in S^3 yields a Seifert fiber space, then r is an integer. We show that for each integer n, there exists a tunnel number one, hyperbolic knot K_n in S^3 such that n-surgery on K_n produces a small Seifert fiber space.
Development of interpersonal problem solving inventory for high school students: The validity and reliability process

Abstract: The current study investigated reliability and validity of an inventory to measure high school students' problem solving strategies and abilities with Interpersonal Problem Solving Inventory (IPSI), which is originally developed to assess college students' problem solving strategies and abilities. IPSI consist of 50 items constructed in a form of 5 sub-scale. These five sub-scales are called; Approaching problems in a negative way, Constructive problem solving, Lack of self-confidence, Unwilling to take responsibility, and Insistent-persevering approach. The sample consist of 482 pupils, 48.1 % of boys (232), 51.9 % of girls (250). % 34.2 of these students were in 9th grade, %33.8 of them were in 10th grade, and %32 of them were in 11th grade. The results of confirmatory factor analysis (CFA) show that the scale consists of five interpersonal problem solving. The relationships between sub-scale scores of high school student showed similar results to the college students' sub-scale score. Confirmatory factor analysis, criterion validity, similar and different structure validity of the sub-scales were proved. The internal consistency values of Cronbach Alfa for the sub-scales are found between .67 and .89; the test-retest reliability is found between .67 and .84. The reliability and validity of the inventory showed that five sub-scales have the ability to measure the interpersonal problem solving ability and behavior for high school students.
Effect of Autologus Platelet-Rich Plasma on IL-6, MMP-3 and MCP-1 Expression in Synoviocytes from Osteoarthritic Patients Knees  [PDF]

Abstract: Nowadays, some studies reported promising results of platelet-rich plasma (PRP) for the treatment of osteoarthritis (OA). However, the effects of PRP on prevention of osteoarthritis in knee joints have been debated. The present study investigated the effects of PRP on osteoarthritis related inflammatory cytokines expressed in fibroblast-like synoviocytes (FLS) from osteoarthritic knees. The synovial tissues were harvested from eight osteoarthritic patients who had undertaken total knee arthroplasties (TKAs) and cultured in DMEM containing 10% FBS. Platelet-rich plasma releasate (PRPr) was made by clotting or activation of PRP by citrate. The levels of PDGF-AA and VEGF in PRPr and whole blood were measured with ELISA method. The FLS were isolated and cultured from osteoarthritic knees. The IL-1β stimulated FLS were cultivated with three different conditions (none, 1% and 10% of PRP). To determine the expression of IL-6, MMP-3, and MCP-1, we used reverse transcriptase polymerase chain reaction (RT-PCR). The concentrations of platelet count in PRP were about 7 to 9 times higher than those of whole blood. The levels of PDGF-AA in PRPr were approximately 3 to 4 times higher than those of whole blood. The levels of VEGF in PRPr were also significantly 7 to 18 times higher than those of whole blood. Without induction of the FLS with IL-1β, 1% or 10% PRPr did not reduce expressions of inflammatory proteins (MMP-3, MCP-1), except for IL-6. However, with induction of the FLS with IL-1β, both concentrations (1% and 10%) of PRPr reduced significantly all inflammatory protein expressions (IL-6, MMP-3, MCP-1). PRPr diminished inflammatory IL-1β-mediated effects on human osteoarthritic fibroblast-like synoviocytes. These results suggest that platelet-rich plasma can be a good therapeutic option for the treatment of osteoarthritis.
The Effect of Cardiovascular Responses after Aerobic Exercise in Menstrual Cycle  [PDF]

Abstract: Purpose: We investigated the effect of menstrual cycle on the cardiovascular responses during recovery period from exercise. 10 healthy women were participated in the early follicular phase (EP: low estrogen and low progesterone: 1 - 4 days) and the late follicular phase (LP: high estrogen and low progesterone: 10 - 13 days) during menstrual cycle. Methods: All subjects completed a graded cycling exercise testing to determine the relative exercise intensity. All subjects were assessed systolic blood pressure (SBP), diastolic blood pressure (DBP), mean arterial pressure (MAP), heart rate (HR), stroke volume (SV), cardiac output (CO), and total vascular conductance (TVC) at resting and during recovery period from moderated cycling exercise during EP and LP. Results: During EP, compared with LP, resting SBP and MAP were higher and TVC was lower (p < 0.05). During recovery from exercise, SBP and MAP were higher and TVC lower during EP compared with LP (p < 0.05). There were no differences in HR, SV and CO in both phases. Conclusions: This study could be increased the peripheral vasoconstriction in low estrogen levels. And during early follicular phase with low estrogen level, it may maintain an increased blood pressure during recovery period following cycling. We suggest that the timing of the menstrual cycle should be considered when measuring blood pressure during clinical exercise testing.
Incidence and clinicopathologic behavior of uterine cervical carcinoma in renal transplant recipients

Abstract: Among 453 women receiving renal transplantation from January 1990 to December 2008, 5 patients were diagnosed with cervical carcinoma. Medical records of these 5 patients were retrospectively reviewed, and clinicopathologic data were collected and analyzed.The incidence of cervical carcinoma in renal transplant recipients was 58.1 out of 100,000 per year, which is 3.5 times higher than in the general Korean population. The mean interval between the time of renal transplantation and the time of cervical carcinoma diagnosis was 80.7 months. After a median follow-up of 96.2 months, there was no recurrence of the disease or death. In 4 patients who were positive from human papillomavirus in situ hybridization (HPV ISH), high or probably high risk HPV DNA was detected in all. Punctate staining of HPV ISH was detected in 3 out of 4 patients.Higher incidence of cervical carcinoma is expected in renal transplant recipients, so appropriate surveillance is needed to ensure early detection and treatment of cervical carcinoma.Recent advancements in immunosuppressive therapy have reduced cases of acute rejection and improved the long-term survival rate of grafts in organ transplant patients. Long-term use of immunosuppressants has likewise increased the incidence of infectious diseases, autoimmune diseases, and malignancy in organ transplant patients.Particularly, the incidence of malignancy was reported to be 2-31% in patients who had received renal transplants [1-6], and was also reported to be a major cause of one-third of cases of post-organ transplant death [7]. Prevalence of Metabolic Syndrome according to Sasang Constitutional Medicine in Korean Subjects

Abstract: Metabolic syndrome (MS) is a complex disorder defined by a cluster of abdominal obesity, atherogenic dyslipidemia, hyperglycemia, and hypertension; the condition is recognized as a risk factor for diabetes and cardiovascular disease. This study assessed the effects of the Sasang constitution group (SCG) on the risk of MS in Korean subjects. We have analyzed 1,617 outpatients of Korean oriental medicine hospitals who were classified into three SCGs, So-Yang, So-Eum, and Tae-Eum. Significant differences were noted in the prevalence of MS and the frequencies of all MS risk factors among the three SCGs. The odds ratios for MS as determined via multiple logistic regression analysis were 2.004 for So-Yang and 4.521 for Tae-Eum compared with So-Eum. These results indicate that SCG may function as a significant risk factor of MS; comprehensive knowledge of Sasang constitutional medicine may prove helpful in predicting susceptibility and developing preventive care techniques for MS.
