Evaluation of the Add-On Effect of Chinese Patent Medicine for Patients with Stable or Unstable Angina: A Systematic Review and Meta-Analysis

Abstract: Chinese herbal medicine (CHM) has been widely used as an adjunct to western medicine in treating angina in China. We carried out this systematic review to evaluate the effectiveness of CHM on top of western medicine for angina. This meta-analysis included 46 randomized control trials with 4212 patients. For trials that included stable angina patients, the CHM group had significant lower incidence of total heart events (relative risk , 95% confidence interval (CI) 0.33–0.78), myocardial infarction ( , 95% CI 0.14–0.72), heart failure ( , 95% CI 0.15–0.91), and angina ( , 95% CI 0.30–0.71) than that of control group. For trials that included unstable angina patients, CHM led to significantly lower occurrence of total heart events ( , 95% CI 0.32–0.66), myocardial infarction ( , 95% CI 0.26–0.54), and angina ( , 95%CI 0.26–0.51). Likewise, for trials that included stable or unstable angina patients, the rates of myocardial infarction ( , 95% CI 0.17–0.68) and angina ( , 95% CI 0.30–0.70) in CHM group were significantly lower than that in control group. In conclusion, CHM is very likely to be able to improve the survival of angina patients who are already receiving western medicine. 1. Introduction Angina is pain or constricting discomfort that typically occurs in the front of the chest and is brought on by physical exertion or emotional stress [1]. It is the main symptomatic manifestation of myocardial ischemia, caused by an imbalance between myocardial blood supply and oxygen demand [1, 2]. The prevalence of angina in the population appears to increase in the past decades [3]. A meta-analysis of data from 31 countries indicated the population weighted prevalence was 6.7% in woman and 5.7% in man [4]. Angina is a common initial presentation of coronary disease [5], and it may exert a major impact on quality of life, ability to work, and costs to society [6, 7]. Angina is clinically classified into stable angina (SA) and unstable angina (UA), and treatment strategy is different between them. SA is a chronic medical condition and the aim of management for it is to abolish or minimize symptoms, improve quality of life, and decrease long-term morbidity and mortality [1] while UA is an acute coronary syndrome, which should be treated as an emergency [8]. The current antiangina medical management includes pharmacological strategies, revascularization strategies, and lifestyle interventions. Meta-Analysis on the Relationship between HLA-DRBl Gene Polymorphism and Cervical Cancer in Chinese Population

Abstract: Aim To determine the association between HLA-DRB1 haplotypes and risk of cervical cancer in unselected and samples from Chinese ethnicities. Methods A comprehensive search for articles from their inception to April 1st, 2013 was conducted from PubMed, Medline, Elsevier Science, Springer Link, Cochrane Library database, China biology medical literature database (CBM),China National Knowledge Infrastructure (CNKI),VIP,and Chinese literature database(Wang fang). A total of 1596 patients with cervical cancer and 2048 controls from the 12 studies on the relationship between gene polymorphism of HLA-DRB l and cervical cancer were performed and data were analyzed and processed using Review Manager 5.0 and Stata 11.0. Results Among the 13 family alleles, two (DRB1*03 and DRB1*08) were found to be negatively associated with cervical cancer in all studies or in Uighur subgroups, and two (DRB1*10 and DRB1*15) were positively associated with in all studies or in Uighur subgroups. Among the 25 specific alleles, six (DRB1*0301, *0403,*0404, *0803, *1312 and *1502) were associated with an increased risk cervical cancer in all studies. No significant association was established for other HLA-DRB1 family alleles and specific alleles. Ethnicity partially explained the race influence of DRB1*12, DRB1*14, DRB1*0301, DRB1*0403, DRB1*0404, DRB1*0803, DRB1*1312 and DRB1*1502 phenotypes. Conclusion Our results support the hypothesis that the HLA-DRB1 family alleles and specific alleles might influence the susceptibility or resistance to cervical cancer, suggesting that immune regulation may play a key role in this disease, although further investigations are still needed.
Gemcitabine Plus Erlotinib for Advanced Pancreatic Cancer: A Systematic Review with Meta-Analysis

Abstract: Background This study aims to comprehensively summarize the currently available evidences on the efficacy and safety of gemcitabine plus erlotinib for treating advanced pancreatic cancer. Methodology/Principal Findings PubMed, EMBASE, The Cochrane Library and abstracts of recent major conferences were systematically searched to identify relevant publications. Studies that were conducted in advanced pancreatic cancer patients treated with gemcitabine plus erlotinib (with or without comparison with gemcitabine alone) and reporting objective response rate, disease control rate, progression-free survival, time-to-progression, overall survival, 1-year survival rate and/or adverse events were included. Data on objective response rate, disease control rate, 1-year survival rate and adverse events rate, respectively, were combined mainly by using Meta-Analyst software with a random-effects model. Data on progression-free survival, time-to-progression and overall survival were summarized descriptively. Sixteen studies containing 1,308 advanced pancreatic cancer patients treated with gemcitabine plus erlotinib were included. The reported median progression-free survival (or time-to-progression), median overall survival, 1-year survival rates, objective response rates and disease control rates were 2–9.6 months, 5–12.5 months, 20%–51%, 0%–28.6% and 25.0%–83.3%, respectively. The weighted 1-year survival rate, objective response rate and disease control rate based on studies reporting robust results were 27.9%, 9.1% and 57.0%, respectively. According to the studies with relevant data, the incidences of total and severe adverse events were 96.3% and 62.9%, respectively. The most frequently reported adverse events were leucopenia, rash, diarrhea, vomitting, neutropenia, thrombocytopenia, anaemia, stomatitis, drug-induced liver injury, fatigue and fever. Compared with gemcitabine alone, the progression-free survival and overall survival with gemcitabine plus erlotinib were significantly longer, but there were also more deaths and interstitial lung disease-like syndrome related to this treatment. Conclusions/Significance Gemcitabine plus erlotinib represent a new option for the treatment of advanced pancreatic cancer, with mild but clinically meaningful additive efficacy compared with gemcitabine alone. Its safety profile is generally acceptable, although careful management is needed for some specific adverse events.

Abstract: 探讨不同育苗基质下桢楠(Phoebe zhennan)苗木对周期性干旱胁迫的适应能力及生存对策,为筛选抗旱育苗基质提供参考。以耕作土、森林腐殖土、泥炭土、蛭石为原料,按体积比配制成5种育苗基质,T1:耕作土=10(传统桢楠育苗)、T2:森林腐殖土=10、T3:耕作土:森林腐殖土:泥炭土:蛭石=4:3:2:1、T4:耕作土:森林腐殖土:泥炭土:蛭石=3:2:4:1、T5:耕作土:森林腐殖土:泥炭土:蛭石=2:1:6:1,采用随机区组设置干旱胁迫梯度,分析不同胁迫水平对桢楠幼苗生长的影响。结果表明:(1)配比基质的理化性质与苗木生长密切相关,正常浇水时,基质处理苗生长质量均优于传统育苗,但重度胁迫下,仅T3、T4处理苗木生长较好;(2)随着干旱胁迫程度的增加,各基质配比桢楠苗木株高、基径、叶面积、叶面积指数、地上生物量、地下生物量等生长指标均受到抑制,但受影响程度存在差异;(3)各基质处理苗木质量隶属函数值T3 > T4=T1 > T2 > T5,T3基质配比最优,所育苗木生长优良,耐旱抗旱性强,显著提高造林成效。表明适宜的基质配比能够协调植物生长所需的水、肥、气、热,有利于植物在逆境中生存。
Telomere Recombination Accelerates Cellular Aging in Saccharomyces cerevisiae

Abstract: Telomeres are nucleoprotein structures located at the linear ends of eukaryotic chromosomes. Telomere integrity is required for cell proliferation and survival. Although the vast majority of eukaryotic species use telomerase as a primary means for telomere maintenance, a few species can use recombination or retrotransposon-mediated maintenance pathways. Since Saccharomyces cerevisiae can use both telomerase and recombination to replicate telomeres, budding yeast provides a useful system with which to examine the evolutionary advantages of telomerase and recombination in preserving an organism or cell under natural selection. In this study, we examined the life span in telomerase-null, post-senescent type II survivors that have employed homologous recombination to replicate their telomeres. Type II recombination survivors stably maintained chromosomal integrity but exhibited a significantly reduced replicative life span. Normal patterns of cell morphology at the end of a replicative life span and aging-dependent sterility were observed in telomerase-null type II survivors, suggesting the type II survivors aged prematurely in a manner that is phenotypically consistent with that of wild-type senescent cells. The shortened life span of type II survivors was extended by calorie restriction or TOR1 deletion, but not by Fob1p inactivation or Sir2p over-expression. Intriguingly, rDNA recombination was decreased in type II survivors, indicating that the premature aging of type II survivors was not caused by an increase in extra-chromosomal rDNA circle accumulation. Reintroduction of telomerase activity immediately restored the replicative life span of type II survivors despite their heterogeneous telomeres. These results suggest that telomere recombination accelerates cellular aging in telomerase-null type II survivors and that telomerase is likely a superior telomere maintenance pathway in sustaining yeast replicative life span.

Abstract: 根据GenBank上精氨酸脱亚氨酶(arginine deiminase,AD)序列AF546864,设计并合成一对引物,用PCR检测29株猪链球菌和7株马链球菌兽疫亚种的ad基因,发现29株猪链球菌均能检出此基因,而7株马链球菌兽疫亚种均未检出。扩增出的猪链球菌2型(SS2)强毒株HA9801的ad基因片段,定向克隆至pBAD/Myc-HisC严紧型质粒并转化TOP10。阳性重组菌经L-阿拉伯糖诱导,表达出分子量约47000Da的重组蛋白。经镍柱亲和层析,获得纯化的重组酶。活性分析显示,该酶具有巯基酶和金属酶的特征,其最适反应温度为37℃,最适pH值为6.5。Western blot分析显示,该酶能与SS2-HA9801全菌制备的兔抗血清发生特异性反应,表明其具有一定的免疫原性。检测该酶有助于进一步分析猪链球菌可能的毒力因子。

Abstract: Historical records indicate that all the roughskin sculpin Trachidermus fasciatus Hackel 1840 in China distributed along coast north to south from Liaoning to Taiwan. For clarifying issues on population and possible regional difference among populations, as well as the taxonomy, intensive investigations were carried out by us in recent years. Data show that the current distribution in China has been obviously reduced as the natural distribution in geography have been broken with actual formation of barriers in evolution, and limited to a few locations in waters of Yalu River, Qinglong River, and Fuchun River only, representing three isolated populations. In this paper, evolutions and taxonomy of the species are discussed. Samples were collected from the rivers regions in China. 19 morphological statistic indices were used for correlation and comparison. The results show that at present, morphological variation among populations are still within the level of species. Subspecies could not be taxonomized.
Histone H4 Lysine 12 Acetylation Regulates Telomeric Heterochromatin Plasticity in Saccharomyces cerevisiae

Abstract: Recent studies have established that the highly condensed and transcriptionally silent heterochromatic domains in budding yeast are virtually dynamic structures. The underlying mechanisms for heterochromatin dynamics, however, remain obscure. In this study, we show that histones are dynamically acetylated on H4K12 at telomeric heterochromatin, and this acetylation regulates several of the dynamic telomere properties. Using a de novo heterochromatin formation assay, we surprisingly found that acetylated H4K12 survived the formation of telomeric heterochromatin. Consistently, the histone acetyltransferase complex NuA4 bound to silenced telomeric regions and acetylated H4K12. H4K12 acetylation prevented the over-accumulation of Sir proteins at telomeric heterochromatin and elimination of this acetylation caused defects in multiple telomere-related processes, including transcription, telomere replication, and recombination. Together, these data shed light on a potential histone acetylation mark within telomeric heterochromatin that contributes to telomere plasticity.
Oral Cancer Development in Patients with Leukoplakia – Clinicopathological Factors Affecting Outcome

Abstract: Background Oral leukoplakia (OL) is the best-known potentially malignant disorder. The objective of the current study was to evaluate the clinicopathological factors predictive of outcome in a large cohort of patients with OL, and report our experience in the early detection of malignant events. Methods A total of 320 patients with biopsy-proven OL were retrospectively reviewed from the study institution who had a mean follow-up of 5.1 years. Data on patient and lesion at initial diagnosis and patient underwent sequential biopsies were reviewed. Multiple biopsies indicates > = 3 times sequential biopsies. Oral cancer-free survival rate (OCFS) was determined by the Kaplan-Meier method and significant factors were identified by Cox regression analysis. Results The 3-year and 5-year OCFS was 86.6% and 82.0%, respectively. A new binary system of grading oral dysplasia was performed and Kaplan-Meier analysis indicated that high-grade dysplasia had significantly higher malignant incidence than low-grade dysplasia (5-year OCFS, 90.5% vs 59.0%; P<0.001), especially during the first 2–3 years of follow-up. Multivariate analysis revealed that the 4 factors including patient aged >60 years, lesion located at lateral/ventral tongue, non-homogenous lesion, high-grade dysplasia were independent significant indicators for OL malignant transformation. In addition, significant positive correlation between the multiple biopsies and these 4 factors and malignant outcome was established. Conclusions Elderly patients with OL located at lateral/ventral tongue and who had non-homogenous lesion with high-grade dysplasia correlated much higher risk of transformation. This high-risk subpopulation was suggested to undergo sequential biopsies and histologic examination contributing to early detection of malignant event.

Abstract: The response time of a magnetorheological damper used in tracked vehicle is analysied. On the base of Bingham model for the magnetorheological and by using fluid dynamics method, the response time of the damper and some influence factors for the response time are theoretically obtained. Experiments for the working of the damper is made. Useful conclusions are drawn.
