Downregulation of CD147 expression alters cytoskeleton architecture and inhibits gelatinase production and SAPK pathway in human hepatocellular carcinoma cells

Abstract: Flow cytometry and western blot assays were employed to detect the transfection efficiency of si-CD147. Confocal microscopy was used to determine the effects of si-CD147 on SMMC-7721 cells' cytoskeleton. Invasion assay, gelatin zymography and cell adhesion assay were employed to investigate the effects of si-CD147 on SMMC-7721 cells' invasion, gelatinase production and cell adhesive abilities. Western blot assay was utilized to detect the effects of si-CD147 on focal adhesion kinase (FAK), vinculiln and mitogen-activated protein kinase (MAPK) expression in SMMC-7721 cells.Downregulation of CD147 gene induced the alteration of SMMC-7721 cell cytoskeleton including actin, microtubule and vimentin filaments, and inhibited gelatinase production and expression, cells invasion, FAK and vinculin expression. si-CD147 also blocked SMMC-7721 cells adhesion to collagen IV and phosphorylation level of SAPK/JNKs. SAPK/JNKs inhibitor SP600125 inhibited gelatinase production and expression.CD147 is required for normal tumor cell architecture and cell invasion. Downregulation of CD147 affects HCC cell structure and function. Moreover, the alteration of cell behavior may be related to SAPK/JNK Pathway. siRNA against CD147 may be a possible new approach for HCC gene therapy.CD147 is a member of immunoglobulin (Ig) superfamily that takes an essential role in several normal tissues but is particularly enriched on the surface of malignant tumor cells in vitro and in vivo [1]. CD147 stimulates production of several matrix metalloproteinases (MMPs) by fibroblasts and endothelial cells [2,3]. Gelatinase, including MMP-2 and MMP-9, is one of the important MMPs and has been proposed to participate in human tumor invasion and metastasis. Other names for CD147 include human basigin, extracellular matrix metalloproteinase inducer (EMMPRIN), and human leukocyte activation-associated M6 antigen [4]. Ordering policy for substitutable products based on consumer preference bias

Abstract: 摘要: 基于消费者选择偏好,建立了相应零售商利润-订单量的条件风险价值模型。以零售商利润为最优化目标,在市场总需求不确定时,考虑零售商风险偏好、消费者选择偏好及两种产品的可替代性,研究零售商的最优订货策略。研究结果表明,当产品订货量在一定范围时,产品替代性将失去作用:当两种产品总订货量在一定范围时,产品最优订货量随顾客偏好系数及产品替代系数的提高而增大,随替代产品顾客偏好系数和零售商风险规避程度的提高而减小:产品与替代产品间的顾客偏好系数差异越大,两产品总订货量越大。
Abstract: Based on consumer preference bias, a profit-order quantity decision model with stochastic demand under Conditional Value-at-Risk(CVaR)criterion was established. In order to optimize retailer's profit, when the demand is uncertain, the optimal ordering policy with the consideration of retailer's risk preference, consumer preference bias and product substitutability were discussed. The results show that when the order is in certain range, the ordering policy depends on consumer preference bias and not on the product substitutability. When the total order is in certain range, the optimal ordering quantity is positively correlated to the consumer preference bias factor and substitutable factor, and it has a negative correlation to the preference bias of another substitutable product and the retailer's risk averse. If the consumer bias between substitute products is bigger, the total ordering quantity will also be bigger
Theoretical research on cascaded difference frequency generation of terahertz radiation

Abstract: A novel mechanism of cascaded difference frequency generation (DFG) is advanced in this article to solve the problem of low-efficiency in terahertz (THz) DFG. The cascaded DFG process is theoretically analyzed using ZnTe crystal as an example. The optimal pumping condition and crystal length is obtained, and the effects of crystal absorption, wave number mis-match and pumping intensity are investigated by solving the coupling wave equations of cascaded DFG. It is obviously seen from the calculation results that the terahertz conversion efficiency can be greatly enhanced in cascaded DFG, in which the photon conversion efficiency can even break through the Manley-Rowe limit.

Abstract: 研究了太赫兹波在具有有限电导率的金属镀层空芯波导中的传输特性.从其传输的特性方程出发,利用Newton-Raphson迭代方法数值模拟了传输损耗和相位常数随太赫兹波频率、波导内径以及波导中金属镀层的电导率的变化关系.结果表明,采用大芯径波导和高电导率的金属镀层能有效降低太赫兹波的传输损耗.

Abstract: The matrix method is employed to analyze the optical activity of liquid crystal, and the matrix presentation of optical rotation is given. The magneto-optical rotation angle of BL-009 nematic liquid crystal is measured using JG-3 type continuous tunable magnetism device and infrared 1350 nm laser. The effect of magnetic field on optical rotation capability of liquid crystal is analyzed in detailed. The threshold of liquid crystal is discussed through experimental testing, and we reached at the conclusion that the rotation direction in liquid crystal is independent of the direction of magnetic field. The obtained result is useful for the design and manufacturing of liquid crystal device.

Abstract: 通过优化双棒串接直腔结构设计,利用大功率LD侧面抽运、声光Q开关、Ⅱ类相位匹配S-KTP内腔倍频获得高效大功率绿色激光输出.当抽运电流为45 A、重复频率为15 kHz时,激光平均功率为132 W,光—光转换效率为132%,脉宽约为120 ns.在输出130 W时,测得1 h功率不稳定度小于05%,光束质量因子M2为67.对高功率抽运情况下激光介质的热透镜效应以及谐振腔稳定运转工作区域也进行了理论分析和实验研究.

Abstract: Using the plane wave expansion method, a kind of two-dimensional photonic crystal with a hexagonal lattice of GaAs columns in air is optimized by connecting the dielectric rods with veins of proper width. The optimal parameters are calculated in order to obtain the largest complete band gap. The width of the complete band gap is insensitive to the radius of dielectric columns, so this kind of two-dimensional photonic crystal is of great practicability.

Abstract: 通过优化平-凹-平三镜折叠腔结构设计,利用大功率半导体激光器侧面抽运、Ⅱ类相位匹配KTP晶体腔内倍频,获得高效高功率连续绿色激光输出.当抽运电流约为36 A时,得到最高36.6 W的连续绿光激光输出,对应的光—光转换效率为8.71%.在输出功率33 W时测量激光功率稳定性,其功率不稳定度为0.27%.用刀口法测量了激光器高输出功率时的光束质量,光束质量因子小于8.对高功率抽运情况下三镜折叠腔的像散补偿、失调灵敏度和基模在腔内分布情况做了数值模拟.

Abstract: An efficient and high-power diode-side-pumped cw 532nm green laser based on a V-shaped cavity geometry, and capable of generating 22.7 W green radiation with opticM conversion effciency of 8.31%, has been demonstrated. The laser is operated with rms noise amplitude of less than 1% and with M2-parameter of about 6.45 at the top of the output power. This laser has the potential for scaling to much higher output power.

Abstract: A cw diode side-pumped Nd:YAG laser is frequency doubled to 532nm with an intracavity KTP crystal in a V-shaped arrangement, achieving an output power of 40W corresponding to an optical-optical conversion efficiency of 9.7%. The instabilities and the M2-parameters of the laser are measured at different output powers after the beam is filtered.
