Prevalence of Human Leukocyte Antigen HLA-B*5701 in HIV-1 Infected Individuals in Brazil  [PDF]

Abstract:

This study was designed to establish the prevalence of HLA-B*5701 at HIV-1 infected individuals in Brazil. A total of 517 consecutive individuals were followed in this study from February 2009 through July 2011. The presence of HLA-B*5701 was determined by Nested-PCR with HLA-B*57 and HLA-B*5701 sequence-specific primers (PCR-SSP). The expression of HLA-B*57 was negative in the 385 (74.5%) and positive in the 103 (19.9%) of infected individuals. Among these, the expression of HLA-B5701 was positive in the 29 (5.6%) of individuals. No demographic or ethnic differences were found between HLA-B*57/HLA-B*5701 HIV-1 negative patients, with a prevalence of Caucasians (57.1%) individuals. During the period of study, 68 patients were submited to an abacavir contain- ing regimen. The HLA-B*5701 allele was observed in 7 (10.3%) patients, with a significant incidence of Hypersensitivity reactions at 4 of them (p < 0.001). Conclusions: Although Brazilian population consists of a mixture of individuals of Caucasian, African and Native American genetic background, prevalence of HLA-B*5701 in this population is similar to the one found in pure Caucasians.


A software framework for microarray and gene expression object model (MAGE-OM) array design annotation

Abstract: We have developed a graphical Java-based application (Adamant) to assist with submission of Microarray designs to public repositories. Output of the application is fully compliant with the standards prescribed by the various public data repositories.Adamant will allow researchers to annotate and submit their own array designs to public repositories without requiring programming expertise, knowledge of the MAGE-OM or XML. The application has been used to submit a number of ArrayDesigns to the Array Express database.Genome-wide measurement of gene expression and genomic variation via DNA microarrays is a widely used technique in biological and medical research. The quantity and complexity of data generated by such studies represents a challenge in terms of storage, analysis and data transfer. There are a number of different microarray technologies and platforms which generate data in a range of different formats and units. The Microarray Gene Expression Data (MGED) society has proposed a standard known as the Minimum Information About a Microarray Experiment (MIAME) standard to address these issues [1]. A MIAME-compliant data-centric model has been developed: the Microarray Gene Expression Object Model (MAGE-OM); this applies a structure to all components of a microarray-based study, from the sequences attached to the array through to describing equipment and platform used to perform the hybridisation reactions [2]. The MGED Ontology (MO) provides a common terminology and hierarchy for data annotation within the MAGE-OM [3]. A notable aspect of the MO is that it allows terms from existing external ontologies to be referenced within the MO framework. Additionally, an XML data-interchange format, MAGE mark-up language, MAGE-ML, has been derived from the MAGE-OM. Abstract: cerebral vasospasm accounts for an increased morbidity and mortality in patients with spontaneous subarachnoid hemorhage (sah). its early and non invasive detection by transcranial doppler (tcd) may contribute to a better prognosis in such cases. purpose: to discuss the use of transcranial doppler (tcd) in the detection of cerebral vasospasm secondary cerebral to spontaneous subarachnoid hemorrhage (sah). methods: we analyzed the flow velocities registered by tcd in intracranial arteries in 31 patients with sah, performed sequentially, associated with neurological examinations. all patients (except one) had intracranial aneurysms. results: clinical vasospasm was found in 11 patients (36,6% from the total), with correspondent sonographic vasospasm present in 9 cases (82%). in 3 cases (33.3%) the flow velocity alterations preceded the clinical symptoms. between the 20 asymptomatic patients, 15 (75%) had normal flow velocity, with evidence of sonographic vasospasm without clinical repercussion in 5 (25%). the index of false negatives in the correlation between clinical examination and tcd (symptomatic vasospasm with normal flow velocity records) was 18%. however, there was correspondence between clinical and ultrasonographic findings in 26 of the 31 patients (83.8%). the average flow velocity values in the various arteries and its interpretation for each subgroup are discussed with further details. conclusion: tcd is a valuable auxiliary method in the detection of cerebral vasospasm (even pre-symptomatic) secondary to sah, allowing a more precocious therapeutic intervention. our results are similar to those described in the literature.

Abstract: O vasoespasmo cerebral é responsável por aumento de morbidade e mortalidade em pacientes acometidos por hemorragia subaracnóide (HSA). Sua detec o de forma precoce e n o-invasiva pelo método de Doppler transcraniano (DTC) pode contribuir para um melhor prognóstico destes doentes. OBJETIVO: Discutir o uso do método de DTC para diagnóstico do vasoespasmo cerebral secundário à HSA espontanea. MéTODOS: Foram analisados os registros de velocidade de fluxo em artérias intracranianas por DTC em 31 pacientes com diagnóstico tomográfico de HSA, realizados seqüencialmente em conjunto com exame neurológico. Todos os pacientes (à exce o de um) eram portadores de aneurismas intracranianos. RESULTADOS: Vasoespasmo clínico foi constatado em 11 pacientes (36,6% do total), com correspondente vasoespasmo ultra-sonográfico presente em nove casos (82%). Em três casos (33,3%) as altera es de velocidade de fluxo precederam os sinais clínicos. Entre os 20 pacientes assintomáticos, 15 (75%) apresentaram medidas de velocidade normais, havendo evidência de vasoespasmo sonográfico sem repercuss o clínica em cinco (25%). O índice de falso negativo na correla o entre exame clínico e DTC (sinais clínicos de vasoespasmo presentes, com velocidades de fluxo normais) foi de 18%. No entanto, houve correspondência entre os achados clínicos e ultra-sonográficos em 26 dos 31 pacientes (83,8%). Os valores de velocidades médias nas várias artérias e sua interpreta o para cada subgrupo s o discutidos com maiores detalhes. CONCLUS O: O método de DTC é um auxiliar valioso na detec o pré-sintomática do vasoespasmo secundário à HSA, permitindo uma interven o terapêutica mais precoce. Os dados obtidos neste trabalho coadunam-se com os diversos estudos publicados na literatura.

Abstract: this paper reports about several factorial tests carried out in 1960 and with n p k fertilization for wheat on ortho dark red latosol in the south of the state of s?o paulo, occupied with field vegetation. the results showed that these soils with over 0.10%of total nitrogen did not react to nitrogenated fertilizers, whereas 0.10 e.mg of potassium was the limit of soil reaction. phosphorus was the element reacting the most showing an evident correlation between response to fertilization and soluble content of the element in the soil. for soils with amounts equal to 0.18 e.mg. of po4-3 soluble in h2so4 0.05 n, productions of around 1000 kg per hectare of wheat seeds were reached without any addition of posphorous fertilizers. at this level, any addition of p2o5 implies to an increase of yield. the results indicate further that the fertility indexes established by catani and others, for the soils of the state of s?o paulo do not apply to the ortho dark red latosol when grown with wheat, concerning the elements nitrogen and potassium.

The main sources for this study are the official national-level documents, information from the website that manages the registration and selection process of the PARFOR – the Plataforma Freire, and information shared by the institutions involved in the plan and by the permanent Forum of support to teacher training of the state of Mato Grosso do Sul.
