Toxoplasma gondii: I. Avalia??o da virulência de oito amostras

Abstract: in order to evaluate the virulence and pathogenicity of tachyzoites, eight toxoplasma gondii strains isolated from different animal species and humans - liv iv, liv v e s 11 isolated from swine, rh e vps from humans, as 28 from mice, hv iii from dog and cn from cat - were inoculated in mice and rabbits. all the strains were strongly virulent for mice. groups of mice inoculated, intraperitonially, with 104 tachyzoites died in an average of 6.0 to 7.8 days, after inoculation. the strains isolated more recently, liv v and hv iii (ld50 of 7 and 15 tachyzoites, respectively) were the most virulent. the rh strain showed the lowest virulence, with ld50 of 3160 tachyzoites. the liv v strain also seems to be most virulent to rabbits.

Abstract: coproparasitological analyses were performed on 191 daycare children and 434 elementary school children from urban and rural areas in rolandia, parana state, brazil. the overall prevalence of enteroparasites was 15.2 % for daycare children and 52.5% for elementary school children. risk factors are discussed.

Abstract: the occurrence of toxoplasmosis and enteroparasitosis was studied in 434 children from elementary schools in the rural and urban areas of rolandia, paraná state, brazil. sera and fecal samples from all the students were submitted to ifa for toxoplasma gondii and coproparasitological tests, respectively. the children were tested by amsler grid and 72 of them were examined for the presence of lesions compatible with ocular toxoplasmosis. some variables were tested but none showed increased risk for toxoplasmosis. the distribution according to sex and age and also same other variables are presented and discussed. correlations between amsler's grid test, toxoplama rifi, occurrence of eyes lesions and enteroparasitosis are also considered.

Abstract: Toxoplasma gondii is a protozoan parasite recognized as an important public health problem. The objective of the present study was to evaluate the occurrence of anti-T. gondii antibodies in pregnant and non pregnant zebus’ breed beef cows (Bos indicus), their fetuses, killed at an abattoir in northern of Paraná state. In the present study 169 cows were evaluated, 92 pregnant (in different stages of gestation) and 77 non pregnant. Sero-occurrence of anti-T. gondii antibodies was performed by Indirect fluorescent antibody test (IFAT) considering positive animals with titers ? 50 for cows and ? 25 for fetuses. Blood (with EDTA) from pregnant cows and blood and tissue samples (brain, lung, heart, and liver) from their fetuses were collected and used for mouse bioassay. Antibodies against T. gondii were observed in 26.0% of cows and 2.5% of fetuses. There was no statistical difference when prevalence of toxoplasmosis was compared between pregnant (23.9%), and non-pregnant (28.6%) animals, and age of gestation (p > 0.59). However, the occurrence of anti-T.gondii antibodies increased with age of animals (p=0.004). Mouse bioassay showed three fetuses positives (3.2%), however, none T. gondii strain was isolated. The present study showed that transplacental transmission of T. gondii naturally occurs in zebu beef cows from Brazil, however, in low rate (5.4%). The anti-T. gondii antibodies occurrence increase with the age of animals, which could be related to the fact that main transmission in cattle T. gondii is horizontal. Toxoplasma gondii é um protozoário reconhecido como um dos mais importantes parasitas em saúde pública. O objetivo do presente estudo foi avaliar a ocorrência de anticorpos contra T. gondii em vacas de corte zebuínas (Bos indicus) gestantes, e seus fetos, bem como, em vacas n o gestantes abatidas em um matadouro no norte do Paraná. No presente estudo foram avaliadas 169 vacas, 92 prenhas (em diferentes fases de gesta o) e 77 n o prenhas. A ocorrência de anticorpos contra T. gondii foi realizada por meio da rea o de imunofluorescência indireta (IFI) considerando animais positivos aqueles com títulos ? 50 para as vacas e ? 25 para os fetos. Sangue (EDTA) de vacas prenhas e amostras de sangue e tecidos (cérebro, pulm o, cora o, e fígado) de seus fetos foram coletadas e utilizadas para o bioensaio em camundongos. Abstract: Thirty-four bovine abortions (fetuses) from Paraná State were examined histologically and immunohistochemically. Eight fetuses had non-suppurative encephalitis and miocarditis, and Neospora caninum were detected by immunohistochemistry. Four cows were seropositive to N. caninum. It was the first detection of congenital neosporosis in aborted bovines fetuses, in Paraná State, Brazil. Trinta e quatro fetos bovinos abortados, no Estado do Paraná, foram examinados através de análise histológica e imuno-histoquímica. O Neospora caninum foi detectado, pela imuno-histoquímica, em oito fetos que apresentavam miocardite e encefalite n o supurativa. Quatro vacas foram soropositivas para o Neospora caninum. Este é o primeiro relato de neosporose congênita em fetos bovinos abortados, no Estado do Paraná, Brasil.
Encephalomyelitis in a calf associated with the protozoan Neospora caninum in the Parana State, Brazil/ Encefalomielite congênita em bezerro associada ao Neospora caninum no estado do Paraná, Brasil

Abstract: A 21 days old Jersey male calf showing neurological signals was examined by the Pathology Emergency Service of Veterinary Hospital of State University of Londrina, Parana, Brazil. After the euthanasia the calf was submitted to the necropsy and fragments of cerebral cortex, cervical area of the spinal marrow, heart, liver and lung were picked for histological exams and virus isolation. No viral agent was isolated from the processed tissues of the calf. The histological exams revealed several ovoid and spherical cysts of protozoan in the brain and spinal marrow. These cysts showed positive reaction in the immunohistochemical (IHQ) for the N. caninum and also for T. gondii. Sera samples of the calf mother’s also presented positive serology for N. caninum, T. gondii, BHV-1 and VBVD. The results of the maternal serology, IHQ, histopathology and clinical signs that the calf presented suggest the involvement of the N. caninum in the case. This is the first report of congenital neosporose in calf in the state of Paraná-Brazil. Um bezerro da ra a Jersey, macho, de 21 dias de vida, com opistótono, apatia e paresia de membros posteriores, desde o nascimento, foi encaminhado ao Hospital Veterinário da Universidade Estadual de Londrina, Paraná-Brasil, para avalia o. Após a eutanásia o bezerro foi submetido à necropsia e fragmentos de córtex cerebral, da regi o cervical da medula espinhal, cora o, fígado e pulm o foram colhidos para exames histológicos e virológicos. Nenhum agente viral foi isolado nos tecidos do bezerro. No exame histológico foram observados cistos de protozoário ovóides e circulares de diversos tamanhos no cérebro e medula espinhal. Estes cistos mostraram rea o positiva na imunoistoquímica (IHQ) para o N. caninum e também para o T.gondii. Amostras de soros colhidas da m e do bezerro também apresentaram sorologia positiva para N. caninum, T. gondii, BHV-1 e VBVD. Os resultados da sorologia materna, da IHQ, os achados histopatológicos e sinais clínicos que o bezerro apresentava sugerem o envolvimento do Neospora caninum no caso. Este é o primeiro relato de neosporose congênita em bezerro no estado do Paraná-Brasil.

Abstract: eight toxoplasma gondii strains were analyzed using elisa and western blot techniques, in order to demonstrate possible immunological differences. the analyzed strains were: liv iv, liv v and s 11 isolated from swine, rh and vps from a human being, as 28 from a wild mouse, hv iii from a dog and cn from a cat. with the elisa assay the eight strains showed similar reactivity with homologous and heterologous sera. the antigenic suspension, consisting of total cellular extract of tachyzoites, was effective in the indirect elisa assay, with the positive sera reacting strongly and the negative not reacting with the antigens. the western blot analysis showed that the t. gondii strains have similar antigenic profiles with a few variations. three bands were observed in all strains: one of about 33 kda (p33), another of 54 kda (p54) and a third one of 66 kda (p66). the hv iii strain, isolated from a dog, did not show three antigens (50, 70 and 75 kda) that were present in the others. however, this difference was not detected by the elisa assay. only two antigens (62 kda of the cn and 67 kda of the liv iv) were strain-specific antigens.
Comportamento imunológico e antigênico de cinco amostras de Toxoplasma gondii inoculadas em gatos

Abstract: the biology of toxoplasma gondii demonstrate that cats are the complete host responsable for the dissemination of this parasites. two domestic cats were immunised with toxoplasma gondii strains vps (human), liv-iv and liv-v (porcine), cpl (caprine) and cn (feline). live tachyzoites were utilized in intravenous inoculation of 2x107 (first inoculum) and 4x107 (second inoculum - 35 days later), except for strain vps, where one cat died 10 days after the first inoculation and another showed symptoms of acute toxoplasmosis. in all other strains, no clinical signs were detected during 6 months of observation. the antibody response after immunization was monitored by indirect immunofluorescence (if) test by the use of anti-cat igg conjugate. the antibody titers obtained at the 20th day varied from 1:1,024 to 1:4,096 and from 1:1,024 to 1:8,000 at the 40th day. only the vps strain attained titers of 1:16,000 at the 30th day of immunization. homologous and heterologos titers were equivalem without any difference among the strain. when the immune sera were adsorbed with live tachyzoites, a reduction in the antibody titers was demonstrated both in homologous and heterologous levels. these results suggest that although dijferences in virulence for cats are evident among the strains. the surface antigens are commom among the t. gondii strains on the basis of if antibody level. the results also demonstrated that apparently there is no correlation between virulence and serological characteristics of the studied strains of toxoplasma gondii. however the importance of the if test in the laboratorial diagnosis of toxoplasmosis is reinforced.
