Candidate Transcriptomic Sources of Inbreeding Depression in Drosophila melanogaster

Abstract: The genomic causes of inbreeding depression are poorly known. Several studies have found widespread transcriptomic alterations in inbred organisms, but it remains unclear which of these alterations are causes of the depression and which are mere responses to the ensuing physiological stress induced by increased homozygosity due to inbreeding. Attempting to differentiate causes from responses, we made a c-DNA microarray analysis of inbreeding depression in Drosophila melanogaster. The rationale of the experiment was that, while depression is a general phenomenon involving reductions in fitness in different inbred lines, its first genetic causes would be different for each inbred line, as they are expected to be caused by the fixation of rare deleterious genes. We took four sets of inbred sublines, each set descending from a different founding pair obtained from a large outbred stock, and compared the expression of the three most depressed sublines and the three least depressed sublines from each set. Many changes in expression were common to all sets, but fourteen genes, grouped in four expression clusters, showed strong set-specific changes, and were therefore possible candidates to be sources of the inbreeding depression observed.
Insights into the role of differential gene expression on the ecological adaptation of the snail Littorina saxatilis

Abstract: Among the 99 transcripts shared between ecotypes, 12.12% showed significant differential expression. At least 4% of these transcripts still displayed significant differences after correction for multiple tests, highlighting that gene expression can differ considerably between subpopulations adapted to alternative habitats in the face of gene flow. One of the transcripts identified was Cytochrome c Oxidase subunit I (COI). In addition, 6 possible reference genes were validated to normalize and confirm this result using qPCR. α-Tubulin and histone H3.3 showed the more stable expression levels, being therefore chosen as the best option for normalization. The qPCR analysis confirmed a higher COI expression in SU individuals.At least 4% of the transcriptome studied is being differentially expressed between ecotypes living in alternative habitats, even when gene flow is still substantial between ecotypes. We could identify a candidate transcript of such ecotype differentiation: Cytochrome c Oxidase Subunit I (COI), a mitochondrial gene involved in energy metabolism. Quantitative PCR was used to confirm the differences found in COI and its over-expression in the SU ecotype. Interestingly, COI is involved in the oxidative phosphorylation, suggesting an enhanced mitochondrial gene expression (or increased number of mitochondria) to improve energy supply in the ecotype subjected to the strongest wave action.Unravelling processes that underlie population divergence is a crucial step towards elucidating the origin and maintenance of biodiversity [1], and towards understanding the genetic basis of speciation, which is one of the most fundamental goals in evolutionary genetics [2]. Abstract: alcohol drinking is one of the main causes of motor vehicle crasches and has an impact on the happening of traffic accidents, particularly those with high number of victims. a literature review was made using services as infomed, medline, national highway traffic safety administration (usa) and mothers against drug driving (canada), with the objective of describing the effects of drinking on the quality of motor vehicle driving and its implications in the legislation of different countries. certain blood alcohol concentration has negative effects on psychomotor capacity, sight, behaviour and other abilities necessary for safe driving, which gradually deteriorate as driver's blood alcohol concentration increases. the synergistic effect of alcohol drinking plus other medications or drug taking should be borne mind as well as the deterioration of driving capabilities associated to the driver's diseases. on the basis of alcohol drinking effects on driving, the law generally sets blood alcohol concentration limits from 0.03 to 0.05 g/dl for non-professional drivers whereas it does not allow professional or very young and unexperienced drivers to have a single drop of alcohol in their blood. these alcohol concentration figures are recommended to be set in cuba.
Costos de las infecciones nosocomiales en pacientes atendidos en una unidad de cuidados intermedios

Abstract: Se estudiaron 129 pacientes egresados de la unidad de cuidados intermedios del hospital "Joaquín Albarrán" durante el primer trimestre de 2002. Se registró la edad, sexo, estadía hospitalaria, procederes de riesgo realizados, complicación con infecciones nosocomiales, la localización de las mismas y costo por estadía. Se utilizaron las pruebas estadísticas siguientes: t de homogeneidad y de Wilcoxon y en una segunda etapa, regresión lineal múltiple. Sin controlar la edad, la estadía global en la unidad y otras salas hospitalarias, resultó superior en los pacientes con infección nosocomial. Igual resultado se obtuvo para los costos por estadía y utilización de antimicrobianos (p = 0,00). Al ajustarse por edad, los resultados del estudio no cambiaron lo que sugirió que la infección nosocomial es un predictor independiente del incremento de los costos por estadía y utilización de antimicrobianos.

Abstract: Se realizó un estudio descriptivo de pacientes ingresados para determinar la justificación de los ingresos en el servicio de medicina interna como elemento para elaborar intervenciones que eleven la calidad y eficiencia de los servicios de salud. Se aplicó a 454 pacientes el Protocolo de Evaluación de la Adecuación (AEP). Se utilizó la técnica estadística de análisis de distribución de frecuencia, comparación de medias y pruebas de independencia para demostrar diferencias entre los grupos según la adecuación del ingreso. Se halló que el 18,7 % de los ingresos fueron considerados no adecuados (INA), los que tuvieron edades (53,5 + 18,2 a os) inferiores que aquellos con ingresos adecuados(62,1 + 18,9 a os) y estadías ligeramente superiores (9,8 vs. 8,5 d). Los pacientes con INA utilizaron el 20 % del total de días/pacientes del período (833 días/pacientes), fueron ingresados básicamente los lunes, martes, miércoles y viernes, y provenían, en general, de áreas de salud no asignadas al hospital. Se concluyó que el frecuente ingreso de pacientes de forma inadecuada requiere de la revisión de aspectos organizativos del servicio y la institución, que conduzcan a una mejor eficiencia de la gestión hospitalaria. A descriptive study of patients admitted was conducted to determine the justification of admissions in the Internal Medicine service as an element to make interventions that increase the quality and efficiency of the health services. The Adapting Evaluation Protocol was applied to 454 patients. The statistical technique of frequency distribution analysis, mean comparison and independence tests was used to show the differences between the groups according to the admission adapting .It was found that 18.7% of the admissions were considered inadequate (INA). The individuals that were inadequately admitted were younger (53.5 ± 18.2 years old) than those with adequate admissions (62.1 ± 18.9 years old), and their lengths of stay were slightly higher (9.8 vs 8.5 days). Patients with inadequate admissions used 20 % of the total of days/patients of the period (833 days/patients), they were admitted basically on Monday, Tuesday, Wednesday and Friday, and they generally came from health areas that were not assigned to the hospital. It was concluded that the frequent inadequate admission of patients requires a review of organizational aspects of the service and institution leading to a better efficiency of hospital management.

Abstract: La ingestión de bebidas alcohólicas constituye una de las principales causas de accidentes de tráfico, influye en la ocurrencia de ellos especialmente en aquellos en los que se producen más victimas. Se realizó una revisión de la literatura, utilizando como fuentes el sitio de salud en Cuba (Infomed), MEDLINE, National Highway Traffic Safety Administration (EE.UU.) y Mothers Against Drugs Driving (Canada), con el propósito de describir los efectos de la ingestión de alcohol en la calidad de la conducción de vehículos automotores y su implicación en las regulaciones legales de diferentes países. Determinada concentración de alcohol en sangre tiene efectos desfavorables sobre la capacidad psicomotora, la visión, el comportamiento y otras conductas necesarias para conducir, las cuales se deterioran de forma progresiva a medida que se incrementa esta concentración en la sangre del conductor. Se debe considerar el efecto sinérgico del consumo de alcohol con otros medicamentos o drogas, así como el deterioro de las capacidades para conducir relacionadas con enfermedades del conductor. Basado en los efectos de la ingestión de alcohol en la conducción, las regulaciones legales autorizan en general a conductores no profesionales a tener concentraciones menores de 0,03 a 0,05 g/dL, mientras que para conductores profesionales o conductores jóvenes e inexpertos no se permite la presencia de alcohol en sangre.
