A Phytoanticipin Derivative, Sodium Houttuyfonate, Induces in Vitro Synergistic Effects with Levofloxacin against Biofilm Formation by Pseudomonas aeruginosa

Abstract: Antibiotic resistance has become the main deadly factor in infections, as bacteria can protect themselves by hiding in a self-constructed biofilm. Consequently, more attention is being paid to the search for “non-antibiotic drugs” to solve this problem. Phytoanticipins, the natural antibiotics from plants, could be a suitable alternative, but few works on this aspect have been reported. In this study, a preliminary study on the synergy between sodium houttuyfonate (SH) and levofloxacin (LFX) against the biofilm formation of Pseudomonas aeruginosa was performed. The minimal inhibitory concentrations (MIC) of LFX and SH, anti-biofilm formation and synergistic effect on Pseudomonas aeruginosa, and quantification of alginate were determined by the microdilution method, crystal violet (CV) assay, checkerboard method, and hydroxybiphenyl colorimetry. The biofilm morphology of Pseudomonas aeruginosa was observed by fluorescence microscope and scanning electric microscope (SEM). The results showed that: (i) LFX and SH had an obvious synergistic effect against Pseudomonas aeruginosa with MIC values of 0.25 μg/mL and 128 μg/mL, respectively; (ii) ? × MIC SH combined with 2 × MIC LFX could suppress the biofilm formation of Pseudomonas aeruginosa effectively, with up to 73% inhibition; (iii) the concentration of alginate decreased dramatically by a maximum of 92% after treatment with the combination of antibiotics; and (iv) more dead cells by fluorescence microscope and more removal of extracellular polymeric structure (EPS) by SEM were observed after the combined treatment of LFX and SH. Our experiments demonstrate the promising future of this potent antimicrobial agent against biofilm-associated infections.

Abstract: Semi-supervised Support Vector Machines is an appealing method for using unlabeled data in classification. Smoothing homotopy method is one of feasible method for solving semi-supervised support vector machines. In this paper, an inexact implementation of the smoothing homotopy method is considered. The numerical implementation is based on a truncated smoothing technique. By the new technique, many “non-active” data can be filtered during the computation of every iteration so that the computation cost is reduced greatly. Besides this, the global convergence can make better local minima and then result in lower test errors. Final numerical results verify the efficiency of the method.
Dynamics and Inequalities in Energy Efficiency in China  [PDF]

Abstract: As rapid economic growth in China in recent decades, the quality of economic growth through improvement of energy efficiency has attracted great attention. This paper evaluated energy efficiency of 29 provinces in China between 2000 and 2016 based on a global non-radial directional distance function. Moreover, the dynamics of energy efficiency were investigated using the non-radial global Malmquist-type efficiency index. The paper also sheds light on the evolution of inequalities in energy efficiency by decomposing interprovincial inequality into its within-region and between-region components. The findings of the study are as follows. First, the national energy efficiency was 0.49 in 2016, which indicated that 51% improvement could be made to reach the global technology frontier. Tianjin, Shanghai, Jiangsu, Shandong and Guangdong had the best energy efficiency in 2016, while Ningxia and Xinjiang had the lowest performance. Second, the national annual growth rate of energy efficiency was 3.4% between 2011 and 2016, which was a positive sign of energy efficiency improvement. Shandong made the biggest improvement in energy efficiency from 2011 to 2016, with 26.2% annual growth rate. Lastly, within-region inequality saw a decreasing trend after 2010 and was overtaken by between-region inequality in 2016.
NMDA Receptors of Gastric-Projecting Neurons in the Dorsal Motor Nucleus of the Vagus Mediate the Regulation of Gastric Emptying by EA at Weishu (BL21)

Abstract: A large number of studies have been conducted to explore the efficacy of electroacupuncture (EA) for the treatment of gastrointestinal motility. While several lines of evidence addressed the basic mechanism of EA on gastrointestinal motility regarding effects of limb and abdomen points, the mechanism for effects of the back points on gastric motility still remains unclear. Here we report that the NMDA receptor (NMDAR) antagonist kynurenic acid inhibited the gastric emptying increase induced by high-intensity EA at BL21 and agonist NMDA enhanced the effect of the same treatment. EA at BL21 enhanced NMDAR, but not AMPA receptor (AMPAR) component of miniature excitatory postsynaptic current (mEPSC) in gastric-projecting neurons of the dorsal motor nucleus of the vagus (DMV). In sum, our data demonstrate an important role of NMDAR-mediated synaptic transmission of gastric-projecting DMV neurons in mediating EA at BL21-induced enhancement of gastric emptying.
Aβ Damages Learning and Memory in Alzheimer’s Disease Rats with Kidney-Yang Deficiency

Abstract: Previous studies demonstrated that Alzheimer’s disease was considered as the consequence produced by deficiency of Kidney essence. However, the mechanism underlying the symptoms also remains elusive. Here we report that spatial learning and memory, escape, and swimming capacities were damaged significantly in Kidney-yang deficiency rats. Indeed, both hippocampal Aβ40 and 42 increases in Kidney-yang deficiency contribute to the learning and memory impairments. Specifically, damage of synaptic plasticity is involved in the learning and memory impairment of Kidney-yang deficiency rats. We determined that the learning and memory damage in Kidney-yang deficiency due to synaptic plasticity impairment and increases of Aβ40 and 42 was not caused via NMDA receptor internalization induced by Aβ increase. β-Adrenergic receptor agonist can rescue the impaired long-term potential (LTP) in Kidney-yang rats. Taken together, our results suggest that spatial learning and memory inhibited in Kidney-yang deficiency might be induced by Aβ increase and the decrease of β2 receptor function in glia.
Grp78 promotes the invasion of hepatocellular carcinoma

Abstract: The Grp78 and FAK expression levels in 44 patients with hepatocellular carcinoma were examined using immunohistochemistry. Grp78 overexpressing SMMC7721 cells were established by pcDNA3.1 (+)-Grp78 transfection and screened by G418. Grp78 and FAK levels in Grp78 overexpressing cells were down-regulated by siRNA transfection. The invasion status of tumor cells was evaluated by transwell assay in vitro, and chick embryo metastasis model in vivo. Cell spreading was determined by cell spreading assay, and quantitatively measured by Orisis software HUG. Grp78, pY397 FAK, pY576/577 FAK and FAK levels were detected by western blot. RhoA activity was detected by GST pulldown assay. The distribution of actin cytoskeleton was observed by fluorescent staining.Grp78 expression levels in 44 patients with hepatocellular carcinoma were negatively correlated with tumor grading, and positively correlated with portal invasion and intra-hepatic invasion. Overexpression of Grp78 in SMMC7721 cells promoted the invasion of cancer cells in vitro and in vivo, and this increase in tumor cell invasion was blocked by Grp78 siRNA knockdown. Our results also revealed that overexpression of Grp78 in SMMC7721 cells accelerated the process of cell spreading and promoted lamellipodia formation. Further analysis showed that overexpression of Grp78 in SMMC7721 cells increased pY397 and pY576/577 levels of FAK. Grp78 siRNA knockdown decreased FAK activation and activity. Our results also revealed that Grp78 overexpression in SMMC7721 cells decreased RhoA-GTP level, and Grp78 siRNA knockdown rescued RhoA-GTP level in Grp78 overexpressing cells, indicating Grp78 inhibited RhoA activity in hepatocellular carcinoma cells. Furthermore, overexpression of Grp78 in SMMC7721 cells increased phospho-p190RhoGAP level. FAK siRNA knockdown in Grp78 overexpressing cells reversed phospho-p190RhoGAP level. CBCT Evaluation of the Upper Airway Morphological Changes in Growing Patients of Class II Division 1 Malocclusion with Mandibular Retrusion Using Twin Block Appliance: A Comparative Research

Abstract: Objective The purpose of this study was to evaluate the morphological changes of upper airway after Twin Block (TB) treatment in growing patients with Class II division 1 malocclusion and mandibular retrusion compared with untreated Class II patients by cone beam computed tomography (CBCT). Materials and Methods Thirty growing patients who have completed TB treatment were recruited into TB group. The control group (n = 30) was selected from the patients with the same diagnosis and without TB treatment. CBCT scans of the pre-treatment (T1) and post-treatment (T2) data of TB group and control data were collected. After three-dimensional (3D) reconstruction and registration of T1 and T2 data, the morphological changes of upper airway during TB treatment were measured. The statistical differences between T1 and T2 data of TB group as well as T2 and control data were accessed by t-test. Results During the TB treatment, the mandible moved advanced by 3.52±2.14 mm in the horizontal direction and 3.77±2.10 mm in the vertical direction. The hyoid bone was in a more forward and inferior place. The upper airway showed a significant enlargement in nasopharynx, oropharynx and hypopharynx. In addition, the nasopharynx turned more circular, and the oropharynx became more elliptic in transverse shape. However, the transverse shape of the hypopharynx showed no significant difference. After comparison between T2 and control data, only the horizontal movement of the hyoid bone, the volumetric expansion of the oropharynx and hypopharynx, and changes of the oropharyngeal transverse shape showed significant difference. Conclusion Compared to the untreated Class II patients, the upper airway of growing patients with Class II division 1 malocclusion and mandibular retrusion showed a significant enlargement in the oropharynx and hypopharynx as well as a more elliptic transverse shape in the oropharynx, and the hyoid bone moved to an anterior position after TB treatment.
A New Equation to Estimate Glomerular Filtration Rate in Chinese Elderly Population

Abstract: Background We sought to develop a new equation to estimate glomerular filtration rate (GFR) in Chinese elderly population. Methods A total of 668 Chinese elderly participants, including the development cohort (n = 433), the validation cohort (n = 235) were enrolled. The new equation using the generalized additive model, and age, gender, serum creatinine as predictor variables was developed and the performances was compared with the CKD-EPI equation. Results In the validation data set, both bias and precision were improved with the new equation, as compared with the CKD-EPI equation (median difference, ?1.5 ml/min/1.73 m2 vs. 7.4 ml/min/1.73 m2 for the new equation and the CKD-EPI equation, [P<0.001]; interquartile range [IQR] for the difference, 16.2 ml/min/1.73 m2 vs. 19.0 ml/min/1.73 m2 [P<0.001]), as were accuracies (15% accuracy, 40.4% vs. 30.6% [P = 0.02]; 30% accuracy, 71.1% vs. 47.2%, [P<0.001]; 50% accuracy, 90.2% vs. 75.7%, [P<0.001]), allowing improvement in GFR categorization (GFR category misclassification rate, 37.4% vs. 53.2% [P = <0.001]). Conclusions A new equation was developed in Chinese elderly population. In the validation data set, the new equation performed better than the original CKD-EPI equation. The new equation needs further external validations. Calibration of the GFR referent standard to a more accurate one should be an useful way to improve the performance of GFR estimating equations.
False Positives Reduction in Top-down Protein Informatics using Support Vector Machines

Abstract: The scarce but consistent chance of getting falsepositive matches [1], [2] in protein database search [3] hasalways casted a shadow over the reliability of results. Thesituation can be helped by viewing the protein data froma descriptive and the probabilistic framework, together.Using the conventional approach as the first stage, top downprotein data is descriptively searched for proteins and theresults are scored and ranked, using a top down proteinsearch engine. We then suggest applying Support VectorMachine, (SVM) as a second stage probabilistic scoringsystem, to the first stage protein database search results so asto further enhance protein classification. For SVM scoring,features are extracted from the top down data and a featuretable is constructed. An SVM using Radial Basis Functionis trained with this feature table. Later classification isperformed on the test data using this SVM. The classificationcan then be viewed together with the previously calculatedsearch engine score and a reordering of top ranked proteinsmay be done.

Abstract: Arbuscular mycorrhizal(AM) fungi play an important role in maintaining the ecosystem function and plant biodiversity in arid lands,and serving as the indispensible factor in the restoration and reestablishment of vegetation.AM fungi are widely distributed in global terrestrial ecosystems and play a bridge role in the cycle of nutrient material between plant and soil.Many studies on the survey of AM fungi resources,ecological distribution,symbiotic relationship and AM fungi related with different environmental factors had been conducted now.Caragana korshinskii Kom.was widely used for vegetation rehabilitation for its high ecological and economic values in China.In addition,it has certain features such as the ability that easy to sprout and forest,rooted deep and the strong resistance to stress.And it also has nodule formation so that can help restore degraded land by atmospheric nitrogen fixation and improve the soil structure and ecological environment.In order to elucidate the diversity and distribution of AM fungi associated with the major desert plant Caragana korshinskii Kom.in desert zone.In July 2010,the soil samples were collected from a depth of 50cm in the rhizosphere of Caragana korshinskii Kom.and were divided into five depth intervals: 0—10,10—20,20—30,30—40,40—50cm in three sampling sites in the desert zone of Hebei and Inner Mongolia province north China including Duolun Lake,Hei Chengzi and Er Yangdian.The annual precipitation,average altitude and coordinate of Duolun Lake were 384.5mm,1312m and N42°11′601″E116°36′870″respectively;of Hei Chengzi were 250-350mm,1321m and N42°09′817″,E115°56′107″;of Er Yangdian were 426mm,1386m and N41°51′095″,E115°47′657″.In this study,24 AM fungal species from four genera were isolated in three sampling sites.In these species,16 species belonged to Glomus,five to Acaulospora,two to Scutellospora and one to Diversispora.Glomus was the dominant genus and Acaulospora was the subdominant genus in the three sampling sites.G.reticulatum was the dominate species in Duolun Lake and Hei Chengzi,and was the subdominant species in Er Yangdian;G.melanosporum was the dominate species in Hei Chengzi and Er Yangdian,and was the subdominant species in Duolun Lake.Spore density of AM fungi in Er Yangdian was the highest,and had no obvious differences between Duolun Lake and Hei Chengzi.Spore density had a significant positive correlation with the available N,soil organic carbon and available P,and the available N was the highest.The results showed that the diversity of AM fungal species was rich in the rhizosphere of Caragana korshinskii Kom.,and they had significant spatial distribution dynamics.The paper described the diversity and distribution of AM fungi in the rhizosphere of Caragana korshinskii Kom.,so that it will provide great help to ecosystem reestablishment,isolation of dominate species and sustainable economic development in desert zone.
