Impaired inflammatory pain and thermal hyperalgesia in mice expressing neuron-specific dominant negative mitogen activated protein kinase kinase (MEK)

Abstract: Formalin-induced second phase spontaneous pain behaviors as well as thermal hyperalgesia measured 1 – 3 hours post-formalin were significantly reduced in the DN MEK mice when compared to their wild type littermate controls. In addition, spinal ERK phosphorylation following formalin injection was significantly reduced in the DN MEK mice. This was not due to a reduction of the number of unmyelinated fibers in the periphery, since these were almost double the number observed in wild type controls. Further examination of the effects of suppression of MEK function on a downstream target of ERK phosphorylation, the A-type potassium channel, showed that the ERK-dependent modulation of the A-type currents is significantly reduced in neurons from DN MEK mice compared to littermate wild type controls.Our results demonstrate that the neuronal MEK-ERK pathway is indeed an important intracellular cascade that is associated with formalin-induced inflammatory pain and thermal hyperalgesia.Extracellular signal-regulated kinases (ERKs) belong to a cascade that is part of a phosphorelay system composed of three sequentially activated kinases regulated by phosphorylation. Initiation of this cascade occurs via multiple mechanisms which ultimately activate raf kinases. Activated raf phosphorylates MEK which phosphorylates ERK1 and ERK2 on tyrosine and threonine residues. Extracellular signal-regulated kinases are involved in the regulation of meiosis and mitosis, and in differentiated cells, ERKs integrate a wide variety of postmitotic functions [1-3]. Within the past decade, numerous studies in rodents have elucidated the role of ERKs in nociceptive plasticity. ERK activation is activity-dependent, and occurs following noxious stimulation [4,5]. Abstract: In the title compound, {[Cu(C18H12N6)]NO3·H2O}n, the CuI ion is coordinated by three N atoms [Cu—N 1.962 (3)–2.019 (3) ] from three 2,4,6-tri-4-pyridyl-1,3,5-triazine (L) ligands. Each L ligand bridges three CuI atoms, generating a positively charged three-dimensional polymeric network with voids propagated along the b axis. These voids are filled with NO3 anions with a shortest Cu...O distance of 2.645 (3) and water molecules, which are linked into negatively charged helical chains via intermolecular O—H...O hydrogen bonds.

Abstract: In the title compound, [Ni(CHO2)2(C18H12N6)(H2O)2]n, the NiII ion, lying on a crystallographic inversion center, has a distorted octahedral coordination comprising two water ligands, two O-atom donors from formate ligands and two N-atom donors from the 2,4,6-tris(4-pyridyl)-1,3,5-triazine ligands. These ligands bridge the NiII complex units, forming zigzag chains along the c axis. Adjacent chains are linked by O—H...O hydrogen bonds, forming a three-dimensional supramolecular network.

Abstract: Persistent infection with high-risk types of human papillomavirus(HPV) is known to cause cervical cancer; however, additional genetic and epigenetic alterations are required for progression from precancerous disease to invasive cancer. DNA methylation is an early and frequent molecular alteration in cervical carcinogenesis. In this review, we summarize DNA methylation within the HPV genome and human genome and identify its clinical implications. Methylation of the HPV long control region (LCR) and L1 gene is common during cervical carcinogenesis and increases with the severity of the cervical neoplasm. The L1 gene of HPV16 and HPV18 is consistently hypermethylated in invasive cervical cancers and can potentially be used as a clinical marker of cancer progression. Moreover, promoters of tumor suppressor genes (TSGs) involved in many cellular pathways are methylated in cervical precursors and invasive cancers. Some are associated with squamous cell carcinomas, and others are associated with adenocarcinomas. Identification of methylated TSGs in Pap smear could be an adjuvant test in cervical cancer screening for triage of women with high-risk HPV, atypical squamous cells of undetermined significance, or low grade squamous intraepithelial lesion (LSIL). However, consistent panels must be validated for this approach to be translated to the clinic. Furthermore, reversion of methylated TSGs using demethylating drugs may be an alternative anticancer treatment, but demethylating drugs without toxic carcinogenic and mutagenic properties must be identified and validated.
Preparation and Characterization of a Novel Adsorbent Made by Cellulose Acetate

Abstract: A novel adsorbent made by cellulose acetate was developed. Preparation method, structure, and adsorbing characterization of the adsorbent were discussed. SEM results showed that the surface of round adsorbent was stable membrane of cellulose acetate, free from obvious cracks, holes, or other defects; while the cross section of the adsorbent was meshy and a lot of cavities were found. The adsorption results of 4 organochlorinated pesticides, such as Dieldrin, Aldrin, Endrin and Heptachlor, show that the adsorbent has higher efficient for organic pollutants, the adsorption rate is about 85% after 12 h. The adsorption rate is faster with the higher lgKow,and the removal efficiency of Heptachlor and Aldrin is up to 99% after 0.5h. The adsorbent can be used to remove persistent organic pollutants effectively.

Abstract: Coagulation behavior of Al13 species was examined in synthetic water with high alkalinity and high humic acid concentration from viewpoint of the transformation of Al hydrolysis products during the coagulation process.The results indicated that coagulation efficiency of Al coagulants positively correlated with the content of Al13 in the coagulation process.Aluminum chloride(AlCl3) was more effective than polyaluminum chloride(PACl) in removing turbidity and dissolved organic matter in the synthetic water because AlCl3 could not only generate Al13 species but also function as pH control agent in the coagulation process. During coagulation process pH control can improve coagulation process through regulating Al speciation,and AlCl3 benefited most from pH control.

Abstract: This paper investigates the absorptive spectral lines of four-level atomic system driven by a coupling, probe and microwave fields. Due to the perturbation of the microwave field, the original electromagnetically induced transparency is changed to electromagnetically induced absorption and the absorptive spectral line can be very narrow. This ultranarrow spectral line has potential applications to the microwave atomic frequency standard and the measurement of very weak magnetic field.
