A highly efficient β-glucosidase from the buffalo rumen fungus Neocallimastix patriciarum W5

Abstract: In this study, a cDNA encoding β-glucosidase was isolated from the buffalo rumen fungus Neocallimastix patriciarum W5 and is named NpaBGS. It has a length of 2,331 bp with an open reading frame coding for a protein of 776 amino acid residues, corresponding to a theoretical molecular mass of 85.1 kDa and isoelectric point of 4.4. Two GH3 catalytic domains were found at the N and C terminals of NpaBGS by sequence analysis. The cDNA was expressed in Pichia pastoris and after protein purification, the enzyme displayed a specific activity of 34.5 U/mg against cellobiose as the substrate. Enzymatic assays showed that NpaBGS was active on short cello-oligosaccharides from various substrates. A weak activity in carboxymethyl cellulose (CMC) digestion indicated that the enzyme might also have the function of an endoglucanase. The optimal activity was detected at 40°C and pH 5?~?6, showing that the enzyme prefers a weak acid condition. Moreover, its activity could be enhanced at 50°C by adding Mg2+ or Mn2+ ions. Interestingly, in simultaneous saccharification and fermentation (SSF) experiments using Saccharomyces cerevisiae BY4741 or Kluyveromyces marxianus KY3 as the fermentation yeast, NpaBGS showed advantages in cell growth, glucose production, and ethanol production over the commercial enzyme Novo 188. Moreover, we showed that the KY3 strain engineered with the NpaNGS gene can utilize 2 % dry napiergrass as the sole carbon source to produce 3.32 mg/ml ethanol when Celluclast 1.5 L was added to the SSF system.Our characterizations of the novel β-glucosidase NpaBGS revealed that it has a preference of weak acidity for optimal yeast fermentation and an optimal temperature of ~40°C. Since NpaBGS performs better than Novo 188 under the living conditions of fermentation yeasts, it has the potential to be a suitable enzyme for SSF.

Abstract: We have developed an efficient platform that uses a combination of transcriptomic and proteomic approaches to N. patriciarum to accelerate gene identification, enzyme classification and application in rice straw degradation. By conducting complementary studies of transcriptome (Roche 454 GS and Illumina GA IIx) and secretome (ESI-Trap LC-MS/MS), we identified 219 putative GH contigs and classified them into 25 GH families. The secretome analysis identified four major enzymes involved in rice straw degradation: β-glucosidase, endo-1,4-β-xylanase, xylanase B and Cel48A exoglucanase. From the sequences of assembled contigs, we cloned 19 putative cellulase genes, including the GH1, GH3, GH5, GH6, GH9, GH18, GH43 and GH48 gene families, which were highly expressed in N. patriciarum cultures grown on different feedstocks.These GH genes were expressed in Pichia pastoris and/or Saccharomyces cerevisiae for functional characterization. At least five novel cellulases displayed cellulytic activity for glucose production. One β-glucosidase (W5-16143) and one exocellulase (W5-CAT26) showed strong activities and could potentially be developed into commercial enzymes.Cellulosic ethanol produced by microbial fermentation from feedstocks has been proposed to replace fossil fuels in transportation. A key step in cellulosic ethanol production is to break down cellulose into glucose and hemicellulose into xylose, which can subsequently be converted into ethanol by fermentative microbes. Therefore, finding efficient cellulases is important to bioethanol production, as well as for hydrolyzing feedstocks into sugars in general. Neocallimastix species is one of the major anaerobic fungi in the rumen of water buffalo capable of efficiently digesting cellulosic biomass [1-4]. Haunting Native Speakerism? Students’ Perceptions toward Native Speaking English Teachers

Abstract: This paper intends to explore how Taiwanese university students perceive their native-speaking English teachers (NESTs). Mutual expectations between the NESTs and students are also investigated. Collected data include questionnaires from 107 students and interviews with three NESTs and 19 students who have filled out the questionnaire. The result shows that students expect more encouragement and interaction with the NESTs, and more relaxed activities with less assignment and test. A third of the students expect NEST with a standard accent, while a quarter do not care about accent at all. The NESTs reveal their dissatisfaction toward the students’ passiveness and irresponsiveness. While students expect their NESTs to be interactive, they themselves seem to give the NESTs an impression of an unwillingness to participate. The discussion centers on this dilemma and offer some suggestions for English teachers.
Regulation of Ubiquitination-Mediated Protein Degradation by Survival Kinases in Cancer

Abstract: The ubiquitin–proteasome system is essential for multiple physiological processes via selective degradation of target proteins and has been shown to plays a critical role in human cancer. Activation of oncogenic factors and inhibition of tumor suppressors have been shown to be essential for cancer development, and protein ubiquitination has been linked to the regulation of oncogenic factors and tumor suppressors. Three kinases, AKT, extracellular signal-regulated kinase, and IκB kinase, we refer to as oncokinases, are activated in multiple human cancers. We and others have identified several key downstream targets that are commonly regulated by these oncokinases, some of which are regulated directly or indirectly via ubiquitin-mediated proteasome degradation, including FOXO3, β-catenin, myeloid cell leukemia-1, and Snail. In this review, we summarize these findings from our and other groups and discuss potential future studies and applications in the clinic.

Abstract: A powerful method, termed flow relaxography, allowing the practical and accurate determination of the water proton MR signal relaxation time distributions has been recently introduced with a phantom, having both flowing and non-flowing water. It was demonstrated that this method can discriminate the signals from the flowing and non-flowing spins, since flow affects the apparent T1 (T1*) value. In this report, it is demonstrated that this technique is effective also in the human leg in vivo. Flow relaxographic angiography is presented for the first time. This technique promises to accomplish the twin goals of flow studies in medical MRI, to display vascular anatomy and measure its blood flow rate, in the same image.
Does Immateriality Matter? Evidence from Accountants Concerning the Acceptability of Questionable Accounting Practices  [PDF]

Abstract: Questionable accounting practices, sometimes, cause from misusing the concept of materiality to manage their reported earnings. The purpose of this study is to examine the attitudes of accountants concerning the acceptability of 14 questionable accounting practices from the amount of material and immaterial perspectives. The results show that, for all 14 questionable accounting practices, the differences in acceptability between the amount of materiality and immateriality were all statistically significant. Regarding the amount of immateriality, especially for questionable, but still legal accounting practices, accountants appear to have the higher average level of acceptability under the extreme pressure from the top management. While questionable accounting practices are clearly illegal, no matter what the amount is material or immaterial, accountants appear to have the lowest average level of acceptability. This finding may be helpful for businesses and professional associations as they develop their own policies, standards and educational programs regarding dealing with questionable accounting practices.

Abstract: Recent studies have shown interest in flame retardants containing phosphorus, nitrogen and sulfur a combination small molecule with a promising new approach in preparing an important class of flame retardant materials. Tetraethyl piperazine-1,4-diyldiphosphonate (TEPP) and O,O,O’,O’- tetramethyl piperazine-1,4-diyldiphosphonothioate (TMPT), based on Piperazine derivatives, were prepared successfully and their structures were proved by means of 1H, 13C and 31P NMR. Cotton twill fabric was treated with both compounds to provide different add-on levels. Thermogravimetric Analysis (TGA), microscale combustion calorimeter (MCC), vertical and 45° flame test and limiting oxygen index (LOI) were performed on the treated cotton fabrics and showed promising results. When the treated twill fabrics (5 wt% - 7 wt% add-ons) were tested using the vertical flammability test (ASTMD6413-11), we observed that the ignited fabrics self extinguished and left behind a streak of char. Limiting oxygen index (LOI, ASTM 2863-09) was utilized to determine the effectiveness of the flame retardant on the treated fabrics. LOI values increased from 18 vol% oxygen in nitrogen for untreated twill fabric to a maximum of 30 vol% for the highest add-on of twill. Furthermore, Scanning Electron Microscope (SEM), Attenuated Total Reflection-Infrared (ATR-IR), and Thermogravimetric Analysis-Fourier Transform Infrared (TGA-FTIR) spectroscopy were employed to characterize the chemical structure on the treated fabrics, as well as, the surface morphology of char areas of treated and untreated fabrics. Additionally, analysis of the release gas products by TGA-FTIR shows some distinctive detail in the degradation of the treated fabrics during the burning process.
