Functional characterization of cellulases identified from the cow rumen fungus Neocallimastix patriciarum W5 by transcriptomic and secretomic analyses

Abstract: We have developed an efficient platform that uses a combination of transcriptomic and proteomic approaches to N. patriciarum to accelerate gene identification, enzyme classification and application in rice straw degradation. By conducting complementary studies of transcriptome (Roche 454 GS and Illumina GA IIx) and secretome (ESI-Trap LC-MS/MS), we identified 219 putative GH contigs and classified them into 25 GH families. The secretome analysis identified four major enzymes involved in rice straw degradation: β-glucosidase, endo-1,4-β-xylanase, xylanase B and Cel48A exoglucanase. From the sequences of assembled contigs, we cloned 19 putative cellulase genes, including the GH1, GH3, GH5, GH6, GH9, GH18, GH43 and GH48 gene families, which were highly expressed in N. patriciarum cultures grown on different feedstocks.These GH genes were expressed in Pichia pastoris and/or Saccharomyces cerevisiae for functional characterization. At least five novel cellulases displayed cellulytic activity for glucose production. One β-glucosidase (W5-16143) and one exocellulase (W5-CAT26) showed strong activities and could potentially be developed into commercial enzymes.Cellulosic ethanol produced by microbial fermentation from feedstocks has been proposed to replace fossil fuels in transportation. A key step in cellulosic ethanol production is to break down cellulose into glucose and hemicellulose into xylose, which can subsequently be converted into ethanol by fermentative microbes. Therefore, finding efficient cellulases is important to bioethanol production, as well as for hydrolyzing feedstocks into sugars in general. Neocallimastix species is one of the major anaerobic fungi in the rumen of water buffalo capable of efficiently digesting cellulosic biomass [1-4]. Abstract: A quantitative analysis method of molybdenum in FeMo alloys by X-ray spectrometry using borate fusion technique was compared with that with pressed pellet. The complete pre-oxidation of FeMo alloys for the preparation of homogeneous fused discs was achieved by employing an automated fusion machine equipped with specially designed O2-blowing nozzles, which used lithium tetra-borate as flux with the addition of lithium nitrate (LiNO3) as oxidizer. The calibration curves of Mo and Fe were used in the quantitative analysis of standard materials and unknown plant samples with satisfactory accuracy and precision, utilizing the corrections of the matrix effects and line overlap. It was confirmed that the newly proposed method of preparing fused glass discs of FeMo alloys can replace the conventional wet chemical analyses requiring the labor intensive and time consuming procedure.

Abstract: The present socioeconomic order tries to legitimate itself saying that there is no alternative for the development except the one proposed by contemporary globalized capitalism. The author analyzes exactly this presupposition. The main purpose of the paper is to indicate the kinds of challenge that this sort of legitimation brings to ethics and theology. It takes into consideration two concepts that are only rarely used by critics of the new liberalism: self-organizing systems and evolution. It is a new interdisciplinary form to approach the complex questions involved in the legitimation of the present world order. The author works in the interface of economy, sociology, biology, psychology and ethics. The article is so a tentative synthesis to bring new light to some well known questions in ethics and theology, which are needing in need of being rethought.
Biomechanical analysis of the lumbar spine on facet joint force and intradiscal pressure - a finite element study

Abstract: A lumbar spine model was constructed from processes including smoothing the bony outline of each scan image, stacking the boundary lines into a smooth surface model, and subsequent further processing in order to conform with the purpose of effective finite element analysis performance. For simplicity, most spinal components were modelled as isotropic and linear materials with the exception of spinal ligaments (bilinear). The contact behaviour of the facet joints and changes of the intradiscal pressure with different postures were analyzed.The results revealed that asymmetric responses of the facet joint forces exist in various postures and that such effect is amplified with larger loadings. In axial rotation, the facet joint forces were relatively larger in the contralateral facet joints than in the ipsilateral ones at the same level. Although the effect of the preloads on facet joint forces was not apparent, intradiscal pressure did increase with preload, and its magnitude increased more markedly in flexion than in extension and axial rotation.Disc pressures showed a significant increase with preload and changed more noticeably in flexion than in extension or in axial rotation. Compared with the applied preloads, the postures played a more important role, especially in axial rotation; the facet joint forces were increased in the contralateral facet joints as compared to the ipsilateral ones at the same level of the lumbar spine.The lumbar spine is a part of the human body that is frequently activated during daily life. This consequently leads to a high incidence of disc problems, such as herniated disc, sciatica, and low back pain. Dose Perturbations of Gold Fiducial Markers in the Prostate Cancer Intensity Modulated Proton Radiation Therapy (IMPT)  [PDF]

Abstract: The objective of this study is to investigate the dose perturbations introduced by the implanted gold fiducial markers in the prostate cancer intensity modulated proton therapy (IMPT) and the impacts of different plan designs on the pertur-bations. Five proton plans: a single lateral field 3D-modulation (3D-mod) plan, 2 fields laterally opposing 3D-mod plan, 6-, 9-, and 18-field distal edge tracking (DET) plans were designed on the CT images of a prostate patient. The dose distributions were first generated for the plans free of fiducial markers with 78 Gy prescribed to 95% of the PTV. To derive the dose perturbations of the gold fiducial markers, three cylindrical shaped gold fiducial markers (3 mm long and 1 mm in diameter) were artificially inserted into the prostate, and the dose distributions were re-computed. Monte Carlo method was used for dose computation. It was found that the gold fiducial markers perturbed the dose distribu-tions, especially along the beam paths. The markers caused a shadowing effect reducing the doses in the areas beyond the markers. Overall, due to the presence of the fiducial markers, D99% of prostate were reduced by 2.96 Gy, 4.21 Gy, 0.16 Gy, 0.34 Gy, 0.15 Gy for the plans of single field 3D-mod, 2-field parallel opposed 3D-mod, 6-, 9-, and 18-field DET respectively. Our study showed these dose perturbation effects decreased with the increase of number of beam angles. Up to 6 beam angles may be required to reduce the dose perturbations from the gold fiducial markers to a clini- cally acceptable level in IMPT.
Joint Power Control and Scheduling for Two-Cell Energy Efficient Broadcasting with Network Coding  [PDF]

Abstract: We consider the energy minimization problem for a two-cell broadcasting system, where the focus is devising energy efficient joint power control and scheduling algorithms. To improve the retransmission efficiency, linear network coding is applied to broadcast packets. Combined with network coding, an optimal algorithm is proposed, which is based on dynamic programming. To reduce computational complexity, two sub-optimal algorithms are also proposed for large networks. Simulation results show that the proposed schemes can reduce energy consumption up to 57% compared with the traditional Automatic Repeat-reQuest (ARQ).

Abstract: Objective: To investigate the application of WPBL (Web Problem-Based Learning) teaching model in surgical teaching so as to better explore and research surgical teaching model. Methods: Questionnaire survey was given to the objects selected from students and teachers of Youjiang Medical University for Nationalities through random table number. Results: Most of the teachers knew about WPBL teaching model and agreed with the introduction of WPBL. As for the students’ satisfaction degree for teaching methods, almost half of the students believed that the teachers could apply flexible and various teaching methods in the classroom teaching, 18% of the students thought that only a small number of teachers met the requirement, but there were still 2% of the students who held that teachers didn’t meet above-mentioned requirement. Conclusion: Teachers have a certain understanding about WPBL teaching model, but the understanding is not deep enough, so the standardized teaching training for teachers is very necessary. At the same time, the students’ high satisfaction degree for teaching and teachers approval indicated that WPBL teaching mode can be recognized by the teachers and students consistently.

Abstract: In the title compound, C22H25NO3, the molecule has a pseudo-mirror plane. The structure is a positional isomer of 2,4-bis(4-methoxyphenyl)-3-azabicyclo[3.3.1]nonan-9-one [Cox, McCabe, Milne & Sim (1985). J. Chem. Soc. Chem. Commun. pp. 626–628]. The 3-azabicyclo[3.3.1]nonan-9-one moiety adopts a double chair conformation with equatorial orientations of both 2-methoxyphenyl substituents on either side of the secondary amino group. The benzene rings are oriented at an angle of 33.86 (4)° with respect to each other and the methoxy groups point towards the carbonyl group. The crystal structure is stabilized by intermolecular N—H...π interactions.

Abstract: In the molecular structure of the title compound, C20H19Br2NO, the fused six-membered heterocyclic and cyclohexane rings adopt a twin-chair conformation with equatorial orientations of all the substituents. Both the ortho-bromo substituents of the benzene rings are oriented towards the carbonyl group; the dihedral angle between the ring planes is 29.13 (3)°. In the crystal structure, the N—H group does not participate in any hydrogen bonds.
