Glycosaminoglycans in Human and Bovine Serum: Detection of Twenty-Four Heparan Sulfate and Chondroitin Sulfate Motifs Including a Novel Sialic Acid-Modified Chondroitin Sulfate Linkage Hexasaccharide

Abstract: Heterogeneous heparan sulfate and chondroitin sulfate glycosaminoglycan (GAG) polysaccharides are important compo- nents of blood circulation. Changes in GAG quantity and structure in blood have been indicated in cancers and other human diseases. However, GAG quantities and structures have not been fully characterized due to lack of robust and sensitive analytical tools. To develop such tools, we isolated GAGs from serum and plasma. We employed liquid chromatography (LC) for GAG quantification and LC/mass spectrometry (MS) for GAG structural analysis. Twenty-four heparan and chondroitin sulfate motifs were identified, including linkage hexasaccharides, repeating disaccharide compositions, reducing, and non-reducing end mono-, di-, tri-, and tetrasaccharide structures. Disaccharides were detectable at picomolar level without radiolabeling or derivitization, so only a few ml of human and fetal bovine serum was required for this study. The detection of different reducing end structures distinct from GAG linkage hexasaccharides revealed that free GAG chains generated by GAG degradation enzymes co-existed with proteoglycans in serum. In addition, a novel sialic acid-modified linkage hexasaccharide was found conjugated to bikunin, the most abundant serum proteoglycan.
Identification of Chemically Sulfated/desulfated Glycosaminoglycans in Contaminated Heparins and Development of a Simple Assay for the Detection of Most Contaminants in Heparin

Abstract: Contaminated heparin was linked to at least 149 deaths and hundreds of adverse reactions. Published report indicates that heparin contaminants were a natural impurity, dermatan sulfate, and a contaminant, oversulfated chondroitin sulfate (OSCS). OSCS was assumed to derive from animal cartilage. By analyzing 26 contaminated heparin lots from different sources, our data indicate that the heparin contaminants were chemically sulfated or chemically sulfated/desulfated glycosaminoglycans (GAGs) consisting of heparan sulfate, chondroitin sulfate, and dermatan sulfate based on monosaccharide quantification, CE, heparin lyase digestion, and liquid chromatography/mass spectrometry analysis. Since currently recommended heparin quality control assays had failed to detect certain heparin contaminants, a simple method that detects most contaminants in heparin was developed. This assay detects specific heparin structures that most contaminants cannot mimic and can be performed in any laboratory equipped with an UV spectrometer.

Abstract:

In the present study, we developed silicosis of rat model by bronchial perfusion SiO2 dust, and intervenes with AcSDKP, immunohisto chemistry was used to detect NF-κb and MCP-1 expression in lung tissue, and positive cells were counted. We found that compared with silicotic model group, the positive cells of NF-κb and MCP-1 were decreased significantly in anti-fibrosis treatment of AcSDKP group. The findings suggest that AcSDKP could inhibit the expression of NF-κb and MCP-1 in lung tissue of silicosos, this may be related to AcSDKP inhibit of macrophage infiltration in lung tissue and reduced the degree of dust alveolitis.

Rapid, Simple and Sensitive Detection of Q Fever by Loop-Mediated Isothermal Amplification of the htpAB Gene

Abstract: Background Q fever is the most widespread zoonosis, and domestic animals are the most common sources of transmission. It is not only difficult to distinguish from other febrile diseases because of the lack of specific clinical manifestations in humans, but it is also difficult to identify the disease in C. burnetii-carrying animals because of the lack of identifiable features. Conventional serodiagnosis requires sera from the acute and convalescent stages of infection, which are unavailable at early diagnosis. Nested PCR and real-time PCR require equipment. In this study, we developed a Loop-Mediated Isothermal Amplification (LAMP) assay to identify C. burnetii rapidly and sensitively. Methods A universal LAMP primer set was designed to detect the repeated sequence IS1111a of the htpAB gene of C. burnetii using PrimerExplorer V4 software. The sensitivity of the LAMP assay was evaluated using known quantities of recombined reference plasmids containing the targeted genes. The specificity of the developed LAMP assay was determined using 26 members of order Rickettsiae and 18 other common pathogens. The utility of the LAMP assay was further compared with real time PCR by the examination 24 blood samples including 6 confirmed and 18 probable Q fever cases, which diagnosed by IFA serological assessment and real time PCR. In addition, 126 animal samples from 4 provinces including 97 goats, 7 cattle, 18 horses, 3 marmots and 1 deer were compared by these two methods. Results The limits of detection of the LAMP assay for the htpAB gene were 1 copy per reaction. The specificity of the LAMP assay was 100%, and no cross-reaction was observed among the bacteria used in the study. The positive rate of unknown febrile patients was 33.3%(95%CI 30.2%–36.4%) for the LAMP assay and 8.3%(95%CI 7.4%–9.2%) for the real time PCR(P<0.05). Similarly, the total positive rate of animals was 7.9%(95%CI 7.1%–8.7%) for the LAMP assay and 0.8%(95%CI 0.7%–0.9%)for the real time PCR(P<0.01). Using the developed LAMP assay, Q fever in the Yi Li area, Xinjiang Province, was confirmed. Conclusions The LAMP assay is a potential tool to support the diagnosis of Q fever in humans and domestic animals in the field, especially in the rural areas of China, because of its rapid and sensitive detection without the aid of sophisticated equipment or a complicated protocol.
Impact of Gender and Working Environment on the Level of State Anxiety among Primary and Middle school Teachers in China  [PDF]

Abstract: State anxiety has become serious among primary and middle school teachers in China. By using stratified random sampling, we selected 2790 primary and middle school teachers as participants. They were from seven regions of China, among them 2278 were valid subjects. Constructed on the basis of anxiety content, the Teachers’ State Anxiety Questionnaire (TSAQ) was used in the investigation. The result of the survey shows that primary and middle school teachers are generally affected by state anxiety; further analysis indicates that the prevalence of state anxiety is relatively higher among male teachers, teachers from key middle schools, teachers working more than 10 hours a day, and teachers loaded with additional task as head teachers. This is directly connected with China’s national situation, teaching envi-ronment, discrepancies in teachers themselves, and their own ability of self-regulation.

Abstract: Anxiety has become one of the major psychological problems which harass people in modern times; this problem is particularly obvious with primary and middle school teachers. Based on the contents of anxiety, the self-compiled Teachers’ State Anxiety Questionnaire (TSAQ) was used to investigate the effective of 1930 subjects in seven regions of China through stratified random sampling methodology. The results show that that the sense of anxiety for satisfaction and control is higher in male teachers; and the sense of teachers’ anxiety for satisfaction and conflict is severe in the key middle school; and the sense of conflict anxiety in the married teachers is the highest; the anxiety in all are high in teachers with poor physical health and main teacher as well as the teachers with poor economic situation in their family. The feasibility of measures to improve the teachers’ state anxiety in primary and middle school should be considered to enhance the level of teachers’ mental health, work efficiency and quality of life.
The integrated risk assessment and zoning of meteorological disaster in Heilongjiang Province of China

Abstract: This paper describes studies on the integrated risk assessment and zoning of meteorological disaster in Heilongjiang Province, in northeastern China, by using information-diffusion theory and cluster method with 35 years of summer temperature and precipitation data from 74 meteorological stations from 1971 through 2005. The information-diffusion theory has been used extensively in risk assessment, yet almost no one has done research about risk assessment by information-diffusion theory based on meteorological disaster standards. Some research results are as follows: the risk probability of low temperature and cold damage in the northern region is higher than that in the southern region; the risk probability of general low temperature and cold damage in the southwestern region is the highest; the risk probability of serious low temperature and cold damage in the northern region is the highest, followed by the central and southeast region; the high-risk region of arid disaster in Heilongjiang Province was primarily located in the southwestern, central, and southern parts of the province; the high-intensity arid disaster was located in the southeastern region; the high-risk region of flood in Heilongjiang Province was primarily located from the southwest and then across the central part to the western part of Heilongjiang Province; the high-intensity flood disasters were located in almost every part of Heilongjiang Province. We can conclude from the integrated meteorological disaster risk zoning that the high-risk region of meteorological disaster is primarily located in the southern and northern part of the province, the moderate-risk region is distributed in the central southern region and western region, the low-risk region is located in the eastern part, and the light-risk region is located in the central western part of Heilongjiang Province.

Abstract: In this paper, we will investigate the boundedness of the bi-parameter Fourier integral operators (or FIOs for short) of the following form: $$T(f)(x)=\frac{1}{(2\pi)^{2n}}\int_{\mathbb{R}^{2n}}e^{i\varphi(x,\xi,\eta)}\cdot a(x,\xi,\eta)\cdot\widehat{f}(\xi,\eta)d\xi d\eta,$$ where for $x=(x_1,x_2)\in \mathbb{R}^{n}\times \mathbb{R}^{n}$ and $\xi,\eta \in \mathbb{R}^{n}\setminus\{0\}$, the amplitude $a(x,\xi,\eta)\in L^\infty BS^m_\rho$ and the phase function is of the form $ \varphi(x,\xi,\eta)=\varphi_1(x_1,\xi)+\varphi_2(x_2,\eta)$ with $\quad \varphi_1,\varphi_2 \in L^\infty \Phi^2 (\mathbb{R}^{n}\times\mathbb{R}^{n}\setminus\{0\})$ and $\varphi(x, \xi, \eta)$ satisfies a certain rough non-degeneracy condition. The study of these operators are motivated by the $L^p$ estimates for one-parameter FIOs and bi-parameter Fourier multipliers and pseudo-differential operators. We will first define the bi-parameter FIOs and then study the $L^p$ boundedness of such operators when their phase functions have compact support in frequency variables with certain necessary non-degeneracy conditions. We will then establish the $L^p$ boundedness of the more general FIOs with amplitude $a(x,\xi,\eta)\in L^\infty BS^m_\rho$ and non-smooth phase function $\varphi(x,\xi,\eta)$ on $x$ satisfying a rough non-degeneracy condition.

Abstract: Marek s Disease (MD) is one of the main infectious diseases that threaten the healthy development of the Chinese poultry industry. The evolution of epidemic strains of the MD Virus (MDV) in China is different from the evolution of strains overseas. The meq gene sequence of pathogenic strains is different from that of the attenuated vaccine strain (MDV-1). The meq and L-meq gene sequences of the MDV in the lymphoid tissue of chickens in latency period are homologous. The meq gene was only found in the lymphoid tissue of chickens during the onset period.

The PEComa family of tumors includes angiomyolipoma (AML), clear cell sugar tumor of the lung (CCST), lymphangioleiomyomatosis (LAM), and rare tumors in other locations [2]. To date, non-AML/non-LAM/non-CCST PEComas have progressively been documented in a variety of anatomical sites, such as visceral organs, soft tissue and bone [3-10]. However, only 12 cases of PEComas of the urinary bladder have been documented in the English-language literature worldwide [11-20]. Herein, we present a urinary bladder lesion occurring in a Chinese adolescent. To the best of our knowledge, the current case is the first published example of bladder PEComa occurring in pediatric population.A 16-year-old girl was referred to a peripheral hospital with a 3-year history of vague abdominal discomfort. A 2.0-cm solid mass was identified in the urinary bladder by ultrasonography. However, her parents preferred to undergo regular follow-up examinations instead of mass resection despite a recommendation. Two years later, at the age of 18, the girl was admitted to our hospital with 1-month history of frequent micturition without hematuria or dysuria. Her family history was unremarkable, and no stigmata of tuberous sclerosis were detected. Ultrasound and computed tomography (CT) scan demonstrated a solid mass arising from the left posterior wall of the bladder. Subsequent magnetic resonance imaging (MRI) of the pelvis revealed a 2.5 × 2.2 cm2 sharply circumscribed soft tissue mass, with a wide base.
