Nuclear expression of Rac1 in cervical premalignant lesions and cervical cancer cells

Abstract: Protein expression was analyzed by immunochemistry on 102 cervical paraffin-embedded biopsies: 20 without Squamous Intraepithelial Lesions (SIL), 51 Low- grade SIL, and 31 High-grade SIL; and in cervical cancer cell lines C33A and SiHa, and non-tumorigenic HaCat cells. Nuclear localization of Rac1 in HaCat, C33A and SiHa cells was assessed by cellular fractionation and Western blotting, in the presence or not of a chemical Rac1 inhibitor (NSC23766).Immunoreacivity for Rac1, RhoA, Tiam1 and beta-Pix was stronger in L-SIL and H-SIL, compared to samples without SIL, and it was significantly associated with the histological diagnosis. Nuclear expression of Rac1 was observed in 52.9% L-SIL and 48.4% H-SIL, but not in samples without SIL. Rac1 was found in the nucleus of C33A and SiHa cells but not in HaCat cells. Chemical inhibition of Rac1 resulted in reduced cell proliferation in HaCat, C33A and SiHa cells.Rac1 is expressed in the nucleus of epithelial cells in SILs and cervical cancer cell lines, and chemical inhibition of Rac1 reduces cellular proliferation. Further studies are needed to better understand the role of Rho-GTPases in cervical cancer progression.Cervical cancer is the second most common malignant neoplasia affecting woman worldwide. Infection with High-Risk Human Papillomavirus (HR-HPV) is considered the main risk factor for developing cervical cancer and its precursor lesions [1-3]. Development of cervical Low-grade Squamous Intraepithelial Lesions (L- SIL) and High-grade Squamous Intraepithelial Lesions (H-SIL), and progression to invasive carcinoma, are associated with alterations in the regulation of several cellular processes such as cell cycle progression, apoptosis, and DNA repair [2,4,5]. The HR-HPV oncoproteins E6 and E7 are responsible for many of these alterations, they act by binding to, and/or modifying the expression/activity of a growing number of cellular proteins [6], including p53 [7], pRb [8], p21 [9,10], and p27 [11,12]. Abstract: purpose: the endothelial function is the cornerstone of several cardiovascular disease. in this trial we compared how the nitric oxide (no) and oxidative stress (os) serum levels, as surrogate markers of endothelial function, change in patients who received (or not) rosuvastatin during the first seven days of an acute coronary syndrome (acs). methods: twenty-two patients with acs (age:66 ± 9 years, gender: ten female and 12 male) were randomized in two groups. patients in the first group (g1) received the conventional treatment for an acs, plus placebo. the other group (g2) additionally received a daily oral dose of 40 mg of rosuvastatin. we measured the blood levels of nitrates and os in both groups twice: at baseline (admission to intensive care unit) and seven days after. the statistical analysis was performed using the paired t-test orthe chi2 test depending of the variables. statistical significance was considered with a p < 0.05. results: groups (g1 and g2) differed statistically on age (g1 = 71 years ± 10 vs g2 63 ± 9 years, p = 0.04). after 7 days of the acs onset, on levels diminished on 21% (p = 0.17) in g1, but raised on 24% in the group who received rosuvastatin (p = 0.005), with statistically difference between groups (p = 0.005). on the other hand, the os, augmented statistically on both groups: g1 (17%, p < 0.001) and g2 (13%, p < 0.001), without any difference between groups (p = 0.77). conclusion: the endothelial dysfunction in the first days of an acs is accentuated, but with the use of rosuvastatina, the endothelial function improves. in contrast, the os increase in both groups, without differences between groups.

Abstract: "syndrome of recommended patient" is manifested as the presence of numerous unexpected and unusual complications in patients that the treating physician is trying to give a better assistance. even assuming that a few complications may appear by chance, there are several factors from daily clinical practice that facilitate the presence of such a syndrome, and some of them can be corrected in order to reduce its incidence. all of them come from the change on daily clinical practice on these patients, as if they do not fit for the attention provided for other people. these factors favouring the presence of this syndrome come from: patients' attitude, inefficient use of health resources, absence of an adequate register of clinical data and change in usual clinical practice on interpretation of diagnostic tests as well as in the indication of treatment of these patients. the best way to prevent this "syndrome of recommended patient" is to maintain, even within these patients, an attitude based on solid clinical knowledge and to follow up the same clinical rules accepted for other patients.

Abstract: In the last decades of the 17th century a small group of merchants from Mexico City guaranteed production of most silver produced in New Spain. This article presents the business networks established by these merchants to purchase silver, the role of family bonds and the importance of trust relationships to finance their ventures. It also analyzes how credit provided by silver merchants funded the mercury supply, specially after they took control of the Mexico City merchant guild which became responsible to collect alcabala taxes. Lastly, this study also examines how venality in royal positions allowed the same merchants to control the only mint in all of New Spain, as well as to illicitly adulterate coinage. En las últimas décadas del siglo XVII un peque o grupo de mercaderes de la ciudad de México garantizaron la producción de la mayor parte de la plata que se producía en Nueva Espa a. En el artículo se exponen las redes que articularon dichos mercaderes para adquirir la plata, el papel de los vínculos familiares y la importancia de las relaciones de confianza en el financiamiento de sus negocios. Se analiza la forma en que el crédito de los tratantes de plata garantizó el abasto de azogue, en particular cuando tomaron el control del Consulado de México y éste se hizo cargo de administrar la renta de alcabalas. Por último, se examina cómo la venalidad de los oficios reales permitió a dichos actores dominar la única Casa de Moneda de Nueva Espa a, así como las prácticas ilícitas que llevaban a cabo en relación con la acu ación.

Abstract: El artículo parte de una toma de posición respecto a la actividad docente: se trata más de un arte complejo que de un método. La clase, en cualquiera de sus formas, es siempre un acontecimiento especial; es una situación presente y personal para docentes y alumnos. En ella se deben sincronizar las expectativas académicas con las expectativas personales de docentes y alumnos para que se genere un espacio de aprendizaje. Tal logro requiere de una especial práctica (o arte): aquella que está abierta a lo inesperado. Es por esta razón que el artículo se concentra en el enfoque de buenas prácticas docentes y luego se propone vincularlo con una concepción humanista de los estudios generales.

Abstract: introduction since three years from now, in mexico there has been several surveys and specific research regarding drug use. over these years the national surveillance systems databases (sisvea, cij) have shown that cocaine use has stabilized and marijuana use is still increasing, although there are some regional differences. also, the results of the 2003 mexico city students' survey revealed a similar inclination: an increase on marijuana use, a slight diminish on cocaine use and as a new stage: a rebound on inhalant use. however, these changes and trends on drug use are not the same all over the country. the results found in these databases suggest that the northern border behaves differently from the rest of the country, even all along the northern border itself. while drug use in matamoros is lower than in other border cities, nuevo laredo, reynosa and ciudad juarez registered high cocaine and heroin use. on the other hand, even though the border cities of sonora and baja california have these same drug use problems, they also show high rates of methamphetamine use. another important issue on border cities is that ciudad juarez has reached tijuana's use levels, mainly among teens between 12 and 1 7 years old. these two cities have the highest substance use levels in the country, even above mexico city. on the central region of the country, the drug use rates are halfway between. even though mexico city's drug use levels are among the central region and the two border cities mentioned before, in all the states that have been studied, there's been a general significant increase in drug use. in southern states, especially yucatan (where there's a household and a students' survey), the drug rates are the lowest in the country. however, the women's drug use rates keeps increasing. specifically the 2003 students' survey showed that in some mexico city's counties women have higher alcohol and tobacco prevalence, and their illegal drug use trends begin to be alike to men

Abstract: The authors carried out a steady and unsteady mass transfer studies to simulate both the release of proteins in physiologic environments and proteins transport through a tissue or organ from polymeric capsules by using a substance, the rhodamine B isothiocyanate dextran (RBID) that mimics the behaviour of glycoproteins such as vascular endothelial growth factor (VEFG). These studies highlighted the importance of electrostatic interactions between alginate and proteins in the release processes. Thereby, this fact has opened new perspectives in order to use these kind of capsules in protein recognition processes. The electrostatic interactions between alginate and RBID allow pH-dependent controlled release systems that simulate the behaviour of glycoproteins.

Abstract: The fractures of the odontoid process of the Axis has been a polemic topic for many years. The alternative technique of placing a fixation screw directly across the fracture site, reattaching the odontoid to the body of C2 had been gained defensors in the actuality in the world. The topic was reviewed, and the seventh case knowned was been operated in Cuba by this technique was presented. This intervention was realized in the Department of Neurosurgery of Cienfuegos. Current considerations are emitted. Las fracturas del proceso odontoides del axis han sido un tema polémico durante muchos a os. La alternativa técnica de colocar una fijación con tornillo directamente a través del sitio de la fractura, reatando la odontoides y el cuerpo de C2, ha ido ganando defensores en la actualidad mundialmente. Se revisa el tema y se reporta el séptimo caso conocido operado en Cuba mediante esta técnica, realizado en el Servicio de Neurocirugía de Cienfuegos. Se emiten consideraciones al respecto.

Abstract: PARP inhibition can induce anti-neoplastic effects when used as monotherapy or in combination with chemo- or radiotherapy in various tumor settings; however, the basis for the anti-metastasic activities resulting from PARP inhibition remains unknown. PARP inhibitors may also act as modulators of tumor angiogenesis. Proteomic analysis of endothelial cells revealed that vimentin, an intermediary filament involved in angiogenesis and a specific hallmark of EndoMT (endothelial to mesenchymal transition) transformation, was down-regulated following loss of PARP-1 function in endothelial cells. VE-cadherin, an endothelial marker of vascular normalization, was up-regulated in HUVEC treated with PARP inhibitors or following PARP-1 silencing; vimentin over-expression was sufficient to drive to an EndoMT phenotype. In melanoma cells, PARP inhibition reduced pro-metastatic markers, including vasculogenic mimicry. We also demonstrated that vimentin expression was sufficient to induce increased mesenchymal/pro-metastasic phenotypic changes in melanoma cells, including ILK/GSK3-β-dependent E-cadherin down-regulation, Snail1 activation and increased cell motility and migration. In a murine model of metastatic melanoma, PARP inhibition counteracted the ability of melanoma cells to metastasize to the lung. These results suggest that inhibition of PARP interferes with key metastasis-promoting processes, leading to suppression of invasion and colonization of distal organs by aggressive metastatic cells.
