Expression of Human Globular Adiponectin-Glucagon-Like Peptide-1 Analog Fusion Protein and Its Assay of Glucose-Lowering Effect In Vivo

Abstract: In this study, human globular adiponectin-glucagon-like peptide-1 analog (gAd-GLP-1-A) fusion protein was expressed and its glucose-lowering effect was measured in vivo. We constructed a prokaryotic expression vector PET28a-gAd-GLP-1-A and transformed the vector into Escherichia coli BL21 (DE3). A recombinant fusion protein of about 25KD was expressed from BL21 (DE3) cells after isopropylthio-β-D-galactoside induction. This protein was N-terminal His-tagged gAd-GLP-1-A fusion protein. Most of the protein was expressed in inclusion body. The fusion protein in inclusion body was purified by using High-Affinity Nickel Iminodiacetic Acid Resin and refolded in urea gradient refolding buffer. The refolded protein was incubated with enterokinase to remove the N-terminal His-tag. The fusion protein without His-tag is gAd-GLP-1-A fusion protein, which exhibited significant glucose-lowering effect in diabetic mice.
Cardioprotective Effect of Sodium Ferulate in Diabetic Rats

Abstract: Reactive oxygen species (ROS) play important roles in the occurrence and development in diabetic cardiomyopathy (DC). Ferulic acid is one of the ubiquitous compounds in diet. Sodium ferulate (SF) is its sodium salt. SF has potent free radical scavenging activity and can effectively scavenge ROS. The study investigated the effect of SF on cardioprotection in diabetic rats. The diabetic rats induced by streptozotocin (STZ) were treated with SF (110mg/kg) by gavage per day for 12 weeks. Results showed that the levels of nitric oxide (NO) and superoxide dismutase (SOD) activity in plasma and myocardium in SF-treated group were significantly higher than those in diabetic control group. The levels of malondialdehyde (MDA) in plasma and myocardium in SF-treated group were significantly lower than those in diabetic control group. Expression of connective tissue growth factor (CTGF) in myocardium in SF-treated group was apparently lower than that in diabetic control group. Compared with normal control group, electron micrographs of myocardium in diabetic control group showed apparently abnormality, while that was significantly ameliorated in SF-treated group. The study demonstrated that SF has a cardioprotective effect via increasing SOD activity and NO levels in plasma and myocardium, inhibiting oxidative stress in plasma and myocardium, and inhibiting the expression of CTGF in myocardium in diabetes rats.
NMR Studies of the C-Terminus of alpha4 Reveal Possible Mechanism of Its Interaction with MID1 and Protein Phosphatase 2A

Abstract: Alpha4 is a regulatory subunit of the protein phosphatase family of enzymes and plays an essential role in regulating the catalytic subunit of PP2A (PP2Ac) within the rapamycin-sensitive signaling pathway. Alpha4 also interacts with MID1, a microtubule-associated ubiquitin E3 ligase that appears to regulate the function of PP2A. The C-terminal region of alpha4 plays a key role in the binding interaction of PP2Ac and MID1. Here we report on the solution structure of a 45-amino acid region derived from the C-terminus of alpha4 (alpha45) that binds tightly to MID1. In aqueous solution, alpha45 has properties of an intrinsically unstructured peptide although chemical shift index and dihedral angle estimation based on chemical shifts of backbone atoms indicate the presence of a transient α-helix. Alpha45 adopts a helix-turn-helix HEAT-like structure in 1% SDS micelles, which may mimic a negatively charged surface for which alpha45 could bind. Alpha45 binds tightly to the Bbox1 domain of MID1 in aqueous solution and adopts a structure consistent with the helix-turn-helix structure observed in 1% SDS. The structure of alpha45 reveals two distinct surfaces, one that can interact with a negatively charged surface, which is present on PP2A, and one that interacts with the Bbox1 domain of MID1.
Removing Mixture of Gaussian and Impulse Noise by Patch-Based Weighted Means

Abstract: We first establish a law of large numbers and a convergence theorem in distribution to show the rate of convergence of the non-local means filter for removing Gaussian noise. We then introduce the notion of degree of similarity to measure the role of similarity for the non-local means filter. Based on the convergence theorems, we propose a patch-based weighted means filter for removing impulse noise and its mixture with Gaussian noise by combining the essential idea of the trilateral filter and that of the non-local means filter. Our experiments show that our filter is competitive compared to recently proposed methods.
Patch-Based Low-Rank Minimization for Image Denoising

Abstract: Patch-based sparse representation and low-rank approximation for image processing attract much attention in recent years. The minimization of the matrix rank coupled with the Frobenius norm data fidelity can be solved by the hard thresholding filter with principle component analysis (PCA) or singular value decomposition (SVD). Based on this idea, we propose a patch-based low-rank minimization method for image denoising, which learns compact dictionaries from similar patches with PCA or SVD, and applies simple hard thresholding filters to shrink the representation coefficients. Compared to recent patch-based sparse representation methods, experiments demonstrate that the proposed method is not only rather rapid, but also effective for a variety of natural images, especially for texture parts in images.
Antioxidant Properties of Cap and Stipe from Coprinus comatus

Abstract: Coprinus comatus, also called chicken drumstick mushroom, is currently commercially available in China. Hot water and ethanolic extracts were prepared from cap and stipe of C. comatus fruit bodies and their antioxidant properties were studied. Ethanolic extract from stipe showed high antioxidant activity (80.6%) at 1 mg/mL. Reducing power of hot water extracts from cap was 1.653 at 10 mg/mL. Extracts from cap showed better scavenging ability on DPPH (57.9% at 1 mg/mL) than stipe ones. Ethanolic extracts were more effective in scavenging ability on hydroxyl radicals (57.4–61.3% at 5 mg/mL) than hot water extracts. Ethanolic extracts showed moderate scavenging ability on superoxide radicals (46.3–47.0% at 20 mg/mL). Naturally occurring antioxidant components including total phenols (3.60–20.00 mg/g), tocopherols (0.58–11.93 mg/g), flavonoids (0.19–3.52 mg/g) and polysaccharides (58.52–547.86 mg/g) were found in the extracts. Overall, extracts from cap were more effective for the antioxidant properties assayed.
Isolation and Structural Characterisation of Okara Polysaccharides

Abstract: Okara is a byproduct generated during tofu or soymilk production processes. Crude polysaccharide (yield 56.8%) was isolated by removing fat, protein and low molecular weight carbohydrates from initial okara. Crude okara polysaccharide was further divided into four soluble fractions and an insoluble residue fraction by extracting with 0.05 M EDTA + NH4 oxalate, 0.05 M NaOH, 1 M NaOH and 4 M NaOH, with yields of 7.7%, 3.6%, 20.7%, 16.0% and 27.9%, respectively. Arabinose, galactose, galacturonic acid, xylose and glucose (only for the insoluble fraction) were the major constituent sugars. The primary sugar residues of okara polysaccharides were 1,4-linked β-galactopyranose, 1,5- and 1,3-linked α-arabinofuranose, 1,5-linked α-xylofuranose, 1,2-linked, 1,2,4-linked and terminal α-rhamnopyranose (or fucopyranose), and 1,4-linked β-glucopyranose (only for the insoluble fraction), indicating okara polysaccharides might contain galactan, arabinan, arabinogalactan, xylogalacturonan, rhamnogalacturonan, xylan, xyloglucan and cellulose.
High power, longevity gallium arsenide photoconductive semiconductor switches

Abstract: A GaAs Photoconductive Semiconductor Switch (PCSS) with a 3-mm gap between the two opposed contact electrodes was developed with carefully chosen GaAs material, mechanical structure design, contact fabrication techniques and its insulation protection. It is charged by a pulse power supply under a bias of 15 kV, illuminated by laser pulses of 1064 nm in wavelength, 56.12 μJ in optical energy per shot and 1 kHz pulse repetition rate (PRR). The GaAs PCSS can last for more than 3.6×106 shots and produce output pulses of 2 MW in peak power, 2 ns pulse duration and 65 ps time jitter from a 50-Ω load of the oscilloscope. When an electric field of 100 kV/cm bias was applied, the peak power of the load was measured at 10 MW. A series of measurements on the voltage conversion rates (VCR) and time jitters have been carried out as the bias voltage increases. In particular, taking into account the dependence of optical absorption coefficient on the bias voltages, the curve of the VCR changes with the bias voltages were analyzed quantitatively.
Propagation characteristics of arrayed transient electromagnetic pulses

Abstract: Starting from the uniform disk current model, an analytical solution for the electromagnetic pulse axial energy radiation is derived and a physical meaning then given to the axial energy propagation characteristics of a unit antenna. From this solution, a point-source approximation model in the time domain is introduced. Parameters such as the energy propagation characteristics and beamwidth of the electromagnetic pulse beam radiating from arrayed antenna are analyzed theoretically. An arrayed transient electromagnetic pulse generator is developed incorporating a 23×16 ultra-wideband tapered slot antenna integrated with a Blumlein formation line and photoconductive semiconductor switch. Experiments performed over a 2.5 km range confirm that the arrayed transient electromagnetic pulse can propagate in free space with the specified waveform. Moreover, the field components can be superposed in phase, and the radiation beam can be focused into a single pulse. Results from calculations are in agreement with experimental results within the measurement uncertainties.
Identification of Phenol-Degrading Nocardia Sp. Strain C-14-1 and Characterization of Its Ring-Cleavage 2,3-Dioxygenase

Abstract: An aerobic bacterial strain C-14-1 was isolated from an acrylic fiber wastewater. The strain was found to belong to Nocardia sp. according to morphological, physiological and its 16S rRNA gene sequence. This strain was able to degrade both alkanes and succinonitrile such as phenol. Catechol 2,3-dioxygenase (C23O) gene was found and amplified with the designed primers from the total DNA of C-14-1. The result of Southern blot indicated that there is only one C23O gene in the genome of C-14-1.
