Regulatory effect of miR-424 on the osteogenic differentiation of human adipose derived mesenchymal stem cells

Abstract: 目的 探讨miR-424在人脂肪来源间充质干细胞(hASC)成骨分化中的表达及miR-424对hASC体外成骨分化的生物学作用。方法 qPCR检测miR-424在hASC成骨分化不同阶段的表达水平。通过慢病毒颗粒在体外感染hASC,使miR-424过表达或抑制其表达,进行Western blotting、细胞免疫荧光试验及碱性磷酸酶(ALP)和茜素红(ARS)染色试验,研究miR-424对骨形态发生蛋白2(BMP-2)及成骨诱导液诱导hASC成骨分化过程的影响。预测miR-424的靶基因,并利用qPCR及Western blotting验证。结果 qPCR结果显示miR-424在hASC的表达量随着成骨诱导时间延长逐渐下降(P<0.05)。在BMP-2诱导下,过表达组的碱性磷酸酶(ALP)和骨桥蛋白(OPN)蛋白水平均下降,而抑制组的表达增加(P<0.05)。细胞免疫荧光显示,过表达miR-424的细胞骨唾液酸蛋白(BSP)阳性率显著下降(P<0.05)。ALP和ARS染色显示,过表达组ALP活性降低,钙盐沉积下降,而抑制组ALP活性升高,矿化能力增强。过表达miR-424能降低hASC中Smad5的mRNA和蛋白水平,而抑制miR-424则使其水平升高(P<0.05)。结论 miR-424在hASC成骨分化中表达下调,其在hASC体外成骨分化过程中起抑制作用。
: Objective To explore the expression and biological effect of miR-424 during osteogenic differentiation of human adipose derived mesenchymal stem cells (hASC). Methods The expressions of miR-424 at different stages of osteogenic differentiation of hASC were detected with real-time PCR. miR-424 was over-expressed or miR-424 expression was inhibited via infection of hASCs with lentivirus particles in vitro. Western blotting, immunocytochemistry, and alkaline phosphatase (ALP) staining and alizarin red (ARS) staining were performed. The effects of miR-424 on osteogenic differentiation of hASCs induced with BMP-2 and osteogenic differentiation medium were investigated. The target gene of miR-424 was predicted and was verified with qPCR and Western blotting. Results The results of qPCR revealed that the expression of miR-424 declined with the time of osteogenic differentiation (P<0.05). The protein levels of ALP and osteopontin (OPN) decreased in the over-expression group and increased in the inhibition group with the induction of bone morphogenetic protein 2(BMP-2) (P<0.05). The results of immunocytochemistry revealed that over-expression of miR-424 significantly reduced the cell positive rate of bone sialoprotein (BSP)(P<0.05). ALP and ARS staining showed that the activity of ALP and capacity of mineralization in the over-expression group decreased, while the activity of ALP and capacity of mineralization increased in the inhibition group. Over-expression of miR-424 could decrease the mRNA and protein expression levels of Smad5 in hASCs and inhibition of miR-424 could increase the mRNA and protein expression levels of Smad5 (P<0.05). Conclusion The expression of miR-424 declines during the osteogenic differentiation of hASCs and miR-424 inhibits the osteogenic differentiation of hASCs
Clinicopathological significance of LRP16 protein in 336 gastric carcinoma patients

Abstract: AIM: To investigate the expression of leukemia related protein 16 (LRP16), and the possible relationship between LRP16 expression and clinicopathological indices in 336 gastric carcinoma patients.METHODS: Immunohistochemistry was used to detect LRP16 expression in 336 cases of paraffin-embedded gastric carcinoma tissues and 60 cases of distal normal mucosa. The relationships between LRP16 expression and patients’ age, tumor size, histological grade, clinical stage, metastatic status and prognosis were analysed.RESULTS: The expression of LRP16 was 58.6% (197/336) in gastric carcinoma and 31.7% (19/60) in distal normal gastric mucosa. The expression of LRP16 in carcinoma was significantly higher than that in normal mucosa tissues (χ2 = 14.929, P = 0.001). LRP16 protein expression was found in 44.1% (63/143) carcinomas at stage I and II, and 69.4% (134/193) carcinomas at stage III and IV (χ2 = 21.804, P = 0.001), and in 56.9% (182/320) of cancers without metastasis but 93.8% (15/16) of those with metastasis (χ2 = 8.543, P = 0.003). The expression of LRP16 was correlated with tumor size, infiltrative depth, clinical stage, lymphatic invasion and distant metastasis (all P < 0.05). Follow-up data showed that there was a significant difference in median survival time between cancer patients with expression of LRP16 (27.0 mo) and those without (48.0 mo, Log rank =31.644, P = 0.001).CONCLUSION: The expression of LRP16 may be associated with invasion, metastasis and prognosis of gastric cancer.
Conversion of entanglement between continuous variable and qubit systems

Abstract: We investigate how entanglement can be transferred between continuous variable and qubit systems. We find that a two-mode squeezed vacuum state and a continuous variable Werner state can be converted to the product states of infinitive number of two-qubit states while keeping the entanglement. The reverse process is also possible.

Abstract: We retrospectively reviewed the records of 73 consecutive patients with SSC who were treated between June 2002 and September 2009. Twenty-two patients were treated with VC and 51 with VCC. Outcome was assessed by clinical examination and magnetic resonance imaging.The patients were divided into five groups based on age at presentation: age less than 1 year (n = 6), 1-5 years (n = 36), 6-10 years (n = 15), 11-20 years (n = 11), and 21-53 years (n = 5). The main clinical presentations were macrocrania (100%), motor deficits (50%), and gaze disturbance (33.3%) in the age less than 1 year group; macrocrania (75%), motor deficits (63.9%), and gaze disturbance (27.8%) in the 1-5 years group; macrocrania (46.7%), symptoms of raised intracranial pressure (ICP) (40.0%), endocrine dysfunction (40%), and seizures (33.3%) in the 6-10 years group; symptoms of raised ICP (54.5%), endocrine dysfunction (54.5%), and reduced visual field or acuity (36.4%) in the 11-20 years group; and symptoms of raised ICP (80.0%) and reduced visual field or acuity (40.0%) in the 21-53 years group. The overall success rate of endoscopic fenestration was 90.4%. A Kaplan-Meier curve for long-term efficacy of the two treatment modalities showed better results for VCC than for VC (p = 0.008).Different age groups with SSCs have different main clinical presentations. Abstract: This paper proposed a matrix-based iceberg construction method called MICC, in order to reduce the space Consumption during the full materialization for a data cube. Users' interests arc considered as the standard to construct an iceberg cube. On the basis of the result of MICC, the paper proposed an incremental update method to dynamically update the iceberg cube while the users' interests are changing. An experiment proves the efficiency and accuracy of these two methods MICC and ICIU.
Microstructure and Phase Behavior of Cationic Gemini/Anionic Polyelectrolyte/Water Ternary System

Abstract: The microstructure of cationic gemini surfactant 1,6-bis(dodecyldimethylammonium) hexane dibromide C12H25(CH3)2N-(CH2)6-N(CH3)2C12H25·2Br] (12-6-12·2Br-) and oppositely charged polyelectrolyte poly(acrylic acid,sodium salt) (NaPA) in aqueous solution has been studied by using fluorescence, conductivity measurement, freeze-etching and TEM. The data obtained from fluorescence and conductivity measurement show that micelle-like or complex can form between the gemini surfactant (12-6-12.2Br-) and polyelectrolyte NaPA due to the static electric interaction and hydrophobic forces. Through freeze-etching and TEM, the microstructure of the mixture solution has been studied, which is consistent with the result from micropolarity. Comparing the fluorescence spectrum of system of dodecyltrimethylammonium bromide (DTAB) and NaPA with that of system of gemini surfactant (12-6-12.2Br-) and NaPA, it can be found that the interaction of (12-6-12.2Br-) and NaPA in aqueous solution has also been detected. It can be shown that the precipitate will transform into gel in higher NaPA concentration.
Deep-Sea Cobalt Crusts Water Jet Cutting Ability  [PDF]

Abstract: Deep-sea cobalt crusts water jet cutting method is proposed to avoid cutter impact load. With simulation calculations and experimental tests, water jet system parameters and its cutting ability were studied. Simula-tion results show that working pressure, ejection range and ejection angle of water jet system are main parameters of its cutting ability. Its important degree is in turn the working pressure, ejection range and ejection angle. Increasing water jet system working pressure is the most effective way to improve its cutting ability. When water jet working pressure is constant, in order to improve its cutting ability, the ejection range should be less than 4mm (four times of nozzle diameter) and the ejection angle should be about 13o. Experimental results show that there is a threshold pressure during water jet cutting cobalt crusts simulation material. With the increase of water jet working pressure, its cutting ability increases dramatically. With the increasing of water jet ejection range, its cutting ability decreases sharply. Genetic Polymorphisms of the TYMS Gene Are Not Associated with Congenital Cardiac Septal Defects in a Han Chinese Population

Abstract: Background Clinical research indicates that periconceptional administration of folic acid can reduce the occurrence of congenital cardiac septal defects (CCSDs). The vital roles of folate exhibits in three ways: the unique methyl donor for DNA expression regulation, the de novo biosynthesis of purine and pyrimidine for DNA construction, and the serum homocysteine removal. Thymidylate synthase (TYMS) is the solo catalysis enzyme for the de novo synthesis of dTMP, which is the essential precursor of DNA biosynthesis and repair process. To examine the role of TYMS in Congenital Cardiac Septal Defects (CCSDs) risk, we investigated whether genetic polymorphisms in the TYMS gene associated with the CCSDs in a Han Chinese population. Method Polymorphisms in the noncoding region of TYMS were identified via direct sequencing in 32 unrelated individuals composed of half CCSDs and half control subjects. Nine SNPs and two insertion/deletion polymorphisms were genotyped from two independent case-control studies involving a total of 529 CCSDs patients and 876 healthy control participants. The associations were examined by both single polymorphism and haplotype tests using logistic regression. Result We found that TYMS polymorphisms were not related to the altered CCSDs risk, and even to the changed risk of VSDs subgroup, when tested in both studied groups separately or in combination. In the haplotype analysis, there were no haplotypes significantly associated with risks for CCSDs either. Conclusion Our results show no association between common genetic polymorphisms of the regulatory region of the TYMS gene and CCSDs in the Han Chinese population.
