Estudo da frequência dos alelos de HLA-DRB1 em pacientes brasileiros com artrite reumatoide

Abstract: the hla-drb1 alleles encoding an amino acid sequence (qkraa/qrraa/rrraa) at position 70 74 of the third hypervariable region of the β1 chain of the hla-drb1 gene, called shared epitope (se), are associated with increased susceptibility to and severity of rheumatoid arthritis (ra) in different populations. objective: to determine the frequency of hla-drb1 alleles in brazilian patients with ra and their association with rheumatoid factor (rf) and anti-citrullinated peptide antibodies (acpa). methods: four hundred and twelve patients with ra (acr 1987) and 215 controls were included. hla-drb1 typing was performed by use of polymerase chain reaction (pcr) with specific primers and hybridization with sequence-specific oligonucleotide probe (ssop). acpa was measured by use of the elisa technique and rf by nephelometry. the statistical analysis comprised the chi-square and student t tests and logistic regression. results: hla-drb1*04:01, *04:04, *04:05 alleles were associated with ra (p < 0.05); despite the wide confidence interval, it is worth noting the association between the drb1*09:01 allele and ra (p < 0.05). hla-drb1 se+ alleles were observed in 62.8% of the patients and in 31.1% of controls (or 3.62; p < 0.001) and were associated with acpa (or 2.03; p < 0.001). drb1-deraa alleles showed a protective effect against ra (or 0.42; p < 0.001). conclusion: in a sample of brazilian patients with ra, most of whom of mixed heritage, hla-drb1 se+ alleles were associated with susceptibility to disease and presence of acpa.

Abstract: thoracic hyperkyphosis is one of the most common postural changes, which is defined as an increase of thoracic spine curvature in the sagittal plane. the alterations that occur in the postural control system with age influence functional mobility and result in a lack of balance in older adults. the objective of this study was to compare thoracic kyphosis between physically active and sedentary older adults in order to determine the influence of physical activity on dorsal kyphosis in the elderly. in a cross-sectional study, 40 healthy volunteers of both genders, ranging in age from 60 to 69 years, were randomly divided into a physically active group and a sedentary group and evaluated by the flexicurve method. descriptive statistics (mean and standard deviation) and the student t-test for independent samples were used for analysis. the results showed that physically active older adults presented fewer changes in the curvature of the thoracic spine than sedentary subjects, but the difference was not significant. in conclusion, active and sedentary older adults present similar characteristics in terms of thoracic kyphosis.

Evaluation of two regional anesthetic methods on the front limb of dogs using hyperbaric bupivacaine

Abstract: purpose: to evaluate the effects of bupivacaine 0.5 and 0.25% in intravenous regional anesthesia (ivra) and brachial plexus block (bpb), respectively, on anesthesia, motor block and cardiovascular parameters in dogs. methods: fourteen healthy adult dogs averaging 10 kilograms (kg) of body weight. animals were randomly assigned to receive one of the two treatments ivra (n=7) or bpb (n=7). all the animals were sedated with acepromazine (0.1 mg/kg intramuscular). to execute the bpb was used an electrical nerve stimulation. anesthesia, motor block, sedation, cardiovascular and respiratory effects were measured as effect of the treatment. results: bpa showed superior efficiency and duration of anesthesia (bpb - 456 ± 94 minutes vs ivra - 138 ± 44) as well as motor block. there only physiologic parameter which change were the systolic pressure in bpb and respiratory rate for both treatments. conclusion: in dogs the 0.25 % hyperbaric bupivacaine in bpb produces a front limb anesthesia about three times more than the 0.5 % in ivra, with ptosis of the limb blocked and little interference in the cardiovascular system but with decrease in respiratory rate.

Abstract: the paper aims at analyzing the article by gerson lima on the manner by which fiscal deficit should be covered. it presents a more general dynamic model, where the principle of effective demand is explicitly used. by doing that, it is possible to treat as endogenous variables the national income and the government entries, what brings the result that the public debt must not follow an explosive path unless the very restrictive conditions of lima's paper prevail. it also evaluates lima's implicit inflation theory, and argues against his approximation to friedman's framework.

Abstract: allelopathy is an interaction among two organisms, where one of that is affected and the other stays stable. it can be source for discoveries of new natural phytotoxic compounds with low toxicity to the organisms that are not target of control. most of the allelochemicals are secondary metabolites like terpenoids, phenolic compounds, organic cyanides and longchain fatty acids. the performance of the allelochemicals can be different according the situations, and it affects a great number of biochemical reactions, resulting in different physiologic modifications in the plants. allelochemicals could affect different pathways, like, the enzymatic activity, division and structure of cells, permeability of the membranes and ions reception, culminating in reduction or inactivation of the germination and plant growth. effects of the allelochemicals on the photosynthesis and breathing has been better characterized, however, several researches had demonstrated the performance of those compounds in oxidative stress, resulting in an increase of oxygen reactive species production which in high concentrations are harmful to cells. in that way, the knowledge of mechanisms of allelochemicals performance is necessary for the development of sustainable techniques in agriculture.
LTB-R1: an alternative to swine mycoplasmal pneumoniae control

Abstract: mycoplasmal pneumoniae is the main respiratory disease in swine. the most efficient way to control it is through the use of vaccines (bacterins), whose production cost is high. the objective of this work was to develop a new alternative for controlling swine mycoplasmal pneumoniae, based on a recombinant subunit vaccine containing the r1 region of p97 adhesin of mycoplasma hyopneumoniae fused to the b subunit of the heat-labile enterotoxin of escherichia coli (rltb-r1). in this work we report the amplification of the genes, genetic fusion between ltb and r1 coding sequences, cloning, construction of the expression vector, as well as expression and purification of rltb-r1 in e. coli.
Tolerance to water deficiency between two soybean cultivars: transgenic versus conventional

Abstract: the main goal of the study was to evaluate the effects on the development and physiological aspects of soybean plants grown under water deficiency at greenhouse conditions, comparing the levels of tolerance to water deficiency between one transgenic and one conventional cultivar, enabling a systematic way to carried out physiological comparisons between soybean cultivars under drought. the study was divided into completely randomized using a factorial 2×2 design, with five replicates. the experimental design included two replenishment levels of daily irrigation (100% and 40%), applied to the leaves during the v4 developmental stage (fourth trifoliate fully expanded) and two cultivars, 'cd 202' conventional and 'cd 226rr' transgenic. the results showed that both cultivars had similar effects, caused by water deficiency, on dry mass production, but the transgenic cultivar tended to maintain higher biomass allocation in pods, as well as, higher efficiency of leaves to support dry mass production than conventional cultivar in both water conditions. moreover, the higher maximum co2 assimilation values and lower membranes damages in the transgenic cultivar under water deficiency showed that the expected higher drought tolerance of the conventional cultivar was not supported by a more accurate physiological investigation.

Abstract: background: although the occurrence of spleen alterations in the various forms of dyslipidemia, such as gaucher's disease, has been well established, the relation between the spleen and lipid metabolism has not been properly studied. in order to contribute to the filling of this gap, in the present investigation we assessed experimentally the lipidogram of rats in the presence of the spleen, in asplenia, and after conservative spleen surgeries. methods: forty female wistar rats of similar weight and age were divided into 4 groups submitted to the following procedures: group 1- control, with an intact spleen; group 2 - total splenectomy; group 3 - subtotal splenectomy, and group 4 - total splenectomy complemented with autogenous spleen tissue implants. four months after the interventions, serum triglycerides, total cholesterol and fractions (vldl, ldl, hdl) were determined. the results for the four groups were compared by analysis of variance followed by the tukey-kramer test, with the level of significance set at p < 0.05. results: there were no differences between groups 1, 3 and 4. in the animals submitted to total splenectomy, total cholesterol (p = 0.0093) and ldl fraction concentrations (p < 0.0001) were higher, whereas hdl fraction concentrations were lower (p = 0.05) than those detected in the other groups. there was no difference in triglycerides (p = 0.1882) or vldl (p = 0.2030) between groups. conclusions: the spleen probably plays an important role in the lipid metabolism of female rats and total splenectomy may be related to changes in cholesterol control. it is possible that preservation of spleen tissue prevents such metabolic disorders.

Abstract: OBJETIVO: Apesar de serem bem estabelecidas as altera es esplênicas nas dislipidemias, como a doen a de Gaucher, ainda n o se estudou adequadamente a rela o do ba o com o metabolismo lipídico. Com vista a contribuir para preencher esse hiato, a presente investiga o avaliou experimentalmente o lipidograma na presen a do ba o, em asplenia e após opera es conservadoras desse órg o. MéTODOS: Foram utilizadas 50 ratas Wistar de pesos e idades semelhantes distribuídas em quatro grupos: Grupo 1- controle, com ba o íntegro; Grupo 2 - laparotomia e laparorrafia, Grupo 3 - esplenectomia total; Grupo 4 - esplenectomia subtotal e Grupo 5 - esplenectomia total complementada por implantes de tecido esplênico autógeno. Após quatro meses, foram dosados os níveis séricos de triglicérides, colesterol total e suas fra es VLDL, LDL, HDL. Os resultados dos quatro grupos foram comparados entre si pela análise de variancia, seguida pelo teste de Tukey-Kramer, com significancia para p < 0,05. RESULTADOS: N o houve diferen a entre os grupos 1, 2, 3 e 4. Nos animais submetidos a esplenectomia total, as concentra es de colesterol total (p = 0,0151) e de sua fra o LDL (p < 0,0001) foram maiores, enquanto a fra o HDL foi menor (p = 0,0026) do que as encontradas nos demais grupos. N o houve diferen a entre os grupos com rela o aos triglicérides (p = 0,1571) e VLDL (p = 0,2527). CONCLUS ES: é provável que o ba o desempenhe um papel de destaque no metabolismo lipídico de ratas e que a esplenectomia total se relacione com altera es no controle do colesterol. é possível que a preserva o de tecido esplênico previna tais distúrbios metabólicos.
