This study investigated the effects of different concentrations of fullerenol on cultured rat hippocampal neurons. Methods and results: Fullerenol at low concentrations significantly enhanced hippocampal neuron viability as tested by MTT assay and Hoechst 33342/propidium iodide double stain detection. At high concentrations, fullerenol induced apoptosis confirmed by Comet assay and assessment of caspase proteins. Conclusion: These findings suggest that fullerenol promotes cell death and protects against cell damage, depending on the concentration present. The concentration-dependent effects of fullerenol were mainly due to its influence on the reduction-oxidation pathway.

Multistep hybridization was applied to promote the hybridization efficiency of each section of sandwich structure. The results showed that both multistep and temperature-controlling hybridization techniques were both especially made to fabricate the sensor for the tendency of internal hybridization of target gene sequences. This strategy provides significantly enhanced hybridization efficiency and sequence specificity of electrochemical detection.

Abstract: On the basis of studies of major elements, trace elements and strontium, neodymium and lead isotopic compositions of Maanshan, Dayingshan and Heikongshan volcanic rocks, it is thought that the magma source of Tengchong volcanoes is the mixture of mid-ocean ridge basalts (MORB) and enriched mantle. It is deduced that the Neotethys subducted ocean crust refused; hence volcanoes were activated with high potassium calc-alkaline magma. This explains why volcanic rocks with geochemical characteristics which suggest tectonic settings of island arcs or active continental margins appeared in the intra-plate background. The magma of Maanshan, Dayingshan and Heikongshan endure crystal fractionation of pyroxene and ilmenite at the chamber stage and plagioclase fractionation at the ascending stage, which induced the magma evolving, from intermediate-basic magma to intermediate-acidic magma, and lava evolving by the course of basaltic trachyandesite- trachyandesite-trachydacite.
High resolution genomic analysis of sporadic breast cancer using array-based comparative genomic hybridization

Abstract: We employed high-resolution array comparative genomic hybridization with 4,134 bacterial artificial chromosomes that cover the genome at 0.9 megabase resolution to analyze 47 primary breast tumors and 18 breast cancer cell lines.Common amplicons included 8q24.3 (amplified in 79% of tumors, with 5/47 exhibiting high level amplification), 1q32.1 and 16p13.3 (amplified in 66% and 57% of tumors, respectively). Moreover, we found several positive correlations between specific amplicons from different chromosomes, suggesting the existence of cooperating genetic loci. Queried by gene, the most frequently amplified kinase was PTK2 (79% of tumors), whereas the most frequently lost kinase was PTK2B (hemizygous loss in 34% of tumors). Amplification of ERBB2 as measured by comparative genomic hybridization (CGH) correlated closely with ERBB2 DNA and RNA levels measured by quantitative PCR as well as with ERBB2 protein levels. The overall frequency of recurrent losses was lower, with no region lost in more than 50% of tumors; the most frequently lost tumor suppressor gene was RB1 (hemizygous loss in 26% of tumors). Finally, we find that specific copy number changes in cell lines closely mimicked those in primary tumors, with an overall Pearson correlation coefficient of 0.843 for gains and 0.734 for losses.High resolution CGH analysis of breast cancer reveals several regions where DNA copy number is commonly gained or lost, that non-random correlations between specific amplicons exist, and that specific genetic alterations are maintained in breast cancer cell lines despite repeat passage in tissue culture. These observations suggest that genes within these regions are critical to the malignant phenotype and may thus serve as future therapeutic targets.Genomic instability is a hallmark of cancer, and specific subchromosomal copy number changes are thought to play a driving role in the transformation of normal cells to malignant clones. Some chemical multidrug resistance inhibitors, such as ciclosporin and verapamil, have been reported to reverse resistance in tumor cells. However, the accompanying side effects have limited their clinical application. In this study, we have developed a novel drug delivery system, ie, a polyethyleneglycol-polycaprolactone (PEG-PCL) copolymer micelle encapsulating doxorubicin, in order to circumvent drug resistance in adriamycin-resistant K562 tumor cells. Methods: Doxorubicin-loaded diblock copolymer PEG-PCL micelles were developed, and the physicochemical properties of these micelles, and accumulation and cytotoxicity of doxorubicin in adriamycin-resistant K562 tumor cells were studied. Results: Doxorubicin-loaded micelles were prepared using a solvent evaporation method with a diameter of 36 nm and a zeta potential of +13.8 mV. The entrapment efficiency of doxorubicin was 48.6% ± 2.3%. The micelles showed sustained release, increased uptake, and cellular cytotoxicity, as well as decreased efflux of doxorubicin in adriamycin-resistant K562 tumor cells. Conclusion: This study suggests that PEG-PCL micelles have the potential to reverse multidrug resistance in tumor cells.

Abstract: Baded on the study of nickel electroplating technology at room temperature, the plated sheet containing rapidly solidified Al-Fe-Cu- V-Si-Ni-Ce-Zr aluminum alloy powders is constructed successfully. The powders and nickel matrix are combined well in the sheet. It can be used to prepare the observation specimen for TEM and determine the hardness of single powder particle as solidified and after heat-treated. The advantages of this method are the realization of heat treatment of powders and the TEM observation of non-interfered microstructure of powders in the size of several microns.

Methods: A novel composite doxorubicin-loaded micelle consisting of polyethylene glycol-polycaprolactone/Pluronic P105 was developed, and carrier-mediated doxorubicin accumulation and release from multicellular spheroids was evaluated. We used confocal laser scanning microscopy and flow cytometry to study the accumulation and efflux of doxorubicin from A549 multicellular spheroids. Doxorubicin radiosensitization and the combined effects of irradiation and doxorubicin on cell migration and proliferation were compared for the different doxorubicin delivery systems. Results: Confocal laser scanning microscopy and quantitative flow cytometry studies both verified that, for equivalent doxorubicin concentrations, composite doxorubicin-loaded micelles significantly enhanced cellular doxorubicin accumulation and inhibited doxorubicin release. Colony-forming assays demonstrated that composite doxorubicin-loaded micelles are radiosensitive, as shown by significantly reduced survival of cells treated by radiation + composite micelles compared with those treated with radiation + free doxorubicin or radiation alone. The multicellular spheroid migration area and growth ability verified higher radiosensitivity for the composite micelles loaded with doxorubicin than for free doxorubicin. Conclusion: Our composite doxorubicin-loaded micelle was demonstrated to have radiosensitization. Doxorubicin loading in the composite micelles significantly increased its cellular uptake, improved drug retention, and enhanced its antitumor effect relative to free doxorubicin, thereby providing a novel approach for treatment of cancer.
Distribution and tectonic setting of Upper Paleozoic volcanic rock in northern Xinjiang

Abstract: Carboniferous volcanic rock developed in the translating period from ocean basin to continental basin in Carboniferous-Early Permian. There are two types of tectonic setting volcanic rock: Collision and post-collision extending stage volcanic rock, and they are distributed in western Junggar, eastern Junggar and southern Junggar around the orogenic belt. Lower Carboniferous volcanic rock was oceanic basaltic, representing continental margin tectonic setting in collision stage. Upper Carboniferous was marine-continental transitional facies intermediate-acid volcanic rock, representing passive continental margin tectonic setting. Lower Permian was continental basaltic and intermediate-acid volcanic rock. Carboniferous volcanic rock in western Junggar was composited by basaltic and intermediate-acid volcanic rock, showing the character of convergence island arc transitional crust. Carboniferous volcanic rock in eastern Junggar was an assemblage of basic and intermediate-acid, representing early island arc extrusion stage and late intraplate extension stage. Carboniferous volcanic rock in southern Junggar in the front of the Bogda Mountain displays a typical rift environment. The Permian volcanic rock mainly distributed in western Junggar, having the characteristics of post-collision stage intraplate extension tectonic setting. It showed the tectonic evolution sequence was: Kelameili remnant ocean basin closed and uplifted in the first in eastern Junggar, Daerbute remnant ocean basin in the next western Junggar, northern Tianshan Ocean closed lastly. The reservoir formation of Carboniferous and Permian volcanic rock was mainly controlled by Carboniferous hydrocarbon source rock. Favorable pool-forming area was surrounded effective source rock. Late Carboniferous extension rift was a potential of hydrocarbon accumulation.

Abstract: The reaction process and mechanistn of reaction synthesis of TiC in Al-Ti-C system have been investigated in detail by observing the microstructure of the water quenched preform with SEM and EDX. The results showed that the reaction processing can be divided into three stages: aluminium powder is melted firstly, then titanium dissolves in the melted aluminium and Al3Ti is formed around titanium powder, titanium concentrates around carbon powder and a Ti-rich layer is observed around carbon pariicles,in which titanium atom reacts with carbon atom to synthesize TiC;TiC precipitates out of the melt, and diffuses out of the layer. A solution-precipitation mechanism and a reaction model were set up based on the experimental results.

Abstract: Compared with conventional cast Cr12Mo V steel and forged Cr12Mo V steels, carbide morphology and properties of modified cast Cr12MoV steels were investigated by performing hardness test, impact test, fracture toughness test, fatigue crack propagation test, metallographic examination and quantitively metallographic analysis. The results indicate that the carbide morphology in modified cast steel is characterized by granular carbide uniformed- distributed, which is different from that of conventional cast steel, but similar to that of forged steel. The impact toughness of modified cast steel is distinctly increased to 12.75J/cm2 from 6.8J/cm2 in the conventional cast steel with networked carbide, because of carbide granulation. Furthermore, The fracture toughness and fatigue - cock-propagation threshoid of modified cast steel are not only higher than that of the conventional cast steel, but also higher than that of the forged steels.
