Cadernos de Saúde Pública , 2006, DOI: 10.1590/S0102-311X2006001200019
Abstract: the objectives of this study were to investigate the molecular basis for plasmodium falciparum resistance to chloroquine in isolates from the brazilian amazon and to identify polymorphisms in the pfmdr1 gene, codons 184, 1042, and 1246, the kappa and gamma regions of the cg2 gene, and the k76t mutation of the pfcrt gene, in order to calculate the distribution of polymorphism within each target gene, comparing samples from distinct geographic areas, using allele-specific polymerase chain reaction (pcr) for the pfmdr gene and pcr plus restriction fragment length polymorphism (rflp) for the cg2 and pfcrt genes. the sample consisted of 40 human blood isolates, already collected and morphologically diagnosed as carriers of p. falciparum parasites, from four localities: porto velho in rondonia state and maraba, itaituba, and tailandia in pará state. distribution of p. falciparum in vitro chloroquine resistance in the isolates was 100% for pfmdr1, cg2 gamma region, and pfcrt, except for the polymorphism in the cg2 kappa region, which was not found.

Abstract: the species uncaria tomentosa (willd.) dc. and u. guianensis gmel. (rubiaceae), known as cat's claw, are large woody vines occurring in the amazon rain forest and other tropical areas of south and central america. it has been used medicinally by indigenous peoples for at least 2,000 years for several diseases. tetra- and pentacyclic oxindole alkaloids, triterpenoid glycosides, sterols and flavonoids are found in these plants. among these metabolites, six pentacyclic oxindole alkaloids, speciophylline, mitraphylline, pteropodine, uncarine f, isopteropodine and isomitraphylline, are considered to be the biochemical markers and are used to standardize commercial herbal medicines. the present study describes the development of an analytical methodology to determine the profile of these alkaloid markers through thin layer chromatography (tlc). this development has also included a comparison among the use of the crude methanol extract and fractions obtained through the classical acid-base partition or by using the basic resin polyamide 6. silica gel was used as stationary phase with the variation of some parameters such as solvent systems, sample concentration, distance of development and detection method. the tlc method developed was shown to be reliable, reproducible and selective for the target alkaloids. it has been applied to the analysis of leaves and stems from both species as well as phytopharmaceutical derivatives based on u. tomentosa.

Abstract: the synthesis of onion (allium cepa) hybrids relies upon the use of two genetic-cytoplasmic male-sterility systems, cms-s and cms-t, in association to the normal male-fertile (n) cytoplasm. however, the molecular phenotyping of male-sterility-inducing and normal cytoplasms of many onion accessions adapted for cultivation under tropical conditions is not available. some of these accessions were reported as presenting tolerance to diseases and adaptation to tropical and subtropical regions. therefore, these accessions are potential sources of parental lines for the brazilian onion hybrid breeding programs. in the present work, polymerase chain reaction (pcr) assays were conducted using mitochondria-derived markers in order to characterize the cytoplasm type present in different morpho-agronomic onion types cultivated in brazil. the s cytoplasm appeared to be the most frequent one among the 66 evaluated accessions with a frequency of 56%. the t male-sterility-inducing cytoplasm was observed in 25.8% of the accessions. of the samples, 18.2% displayed exclusively the cytoplasm n. this information could be useful to guide the onion breeding programs in the selection of parental lines aiming to develop hybrid cultivars adapted to tropical and subtropical areas of the world.

Abstract: parasitism is an important mechanism affecting populations and communities. the purpose of this study was to investigate the ectoparasites fauna living on the body of the white-necked trush, turdus albicollis vieillot, 1818, and to evaluate if the host body mass is affected by these parasites. turdus albicollis were monthly captured at ilha grande, rio de janeiro, from july 1999 to june 2000 in an area of atlantic forest. the birds were individualy marked, weighed and carefully checked to record and quantify the presence of ectoparasites. parasite abundance and location on the bird's body were recorded. in 54 individuals of t. albicollis sampled, two ectoparasite species were found. the prevalence of the feather mite pterodectes turdinus berla, 1959, and the tick amblyomma longirostre koch, 1844 was 72,2% and 27,8%, respectively. the monthly abundance of p. turdinus was related to the dry and wet months, and it was significantly higher in the dry months than in the wet months. there was no significant relationship between hosts body mass (g) and total abundance of feather mites and ticks.

Abstract: in this article we discuss the propagation of small amplitude waves in a liquid of constant depth based on a wave equation. the general solution shows that propagation speed depends on the wavelength and consequently there is waves dispersion. solutions for shallow and the deep water are shown.

Abstract: Ferulic acid (FA) and its derivatives (FADs) are known for a variety of biological activities, such as photo-protective agent, antioxidant, antiatherogenic and antiplasmodial activities. During structural definition of a FAD isolated from Croton pullei, the possibility of a heterologous series made this definition difficult. In this regard, computational simulations were performed using theoretical calculations at DFT level to predict Infrared (IR) and Nuclear Magnetic Resonance (NMR) data. The IR and NMR 13C and 1H data were compared with the theoretical calculations performed for three structural possibilities of a heterologous series. The theoretical results were compared with the experimental data through linear regression in order to define the most probable structure and showed satisfactory values.

Abstract: Chemical investigation of Croton pullei (Euphorbiaceae) collected in the Brazilian Amazon region was revisited. The chemical composition of the essential oils of leaves and stems was analyzed by GC/MS. It was found that both the oils comprise mainly terpenes, among which linalool was the major one (24.90 and 39.72%, respectively). Phytochemical investigation of the stem methanol extract led to the isolation of a new natural product from the glutarimide alkaloid group named N-[2,6-dioxo-1-(2-phenylethyl)-3-piperidinyl]-acetamide, confirming that C. pullei is a rich source of this class of alkaloids. The hexane and methanol extracts of the stems of C. pullei showed moderate antibacterial and antifungal activity and the highest inhibition was observed when the methanol extract was tested against Staphylococcus aureus CCMB 262 and CCMB 263.

Abstract: this study is based on the health belief model, using a quanti-qualitative approach by means of percentage calculations as quantitative data, while presenting qualitative data through content analysis. this study aims to analyze significant beliefs among nursing workers who were victims of occupational accidents while exposed to biological material, either adhering to chemoprophylactic treatment or not with antiretroviral medication. this study evidenced and reveals the need to consider physical, personal, and emotional aspects of worker injuries when providing them care and to implement actions that minimize the occurrence of such injures.
