Inhibition of cell growth by EGR-1 in human primary cultures from malignant glioma

Abstract: Background The aim of this work was to investigate in vitro the putative role of EGR-1 in the growth of glioma cells. EGR-1 expression was examined during the early passages in vitro of 17 primary cell lines grown from 3 grade III and from 14 grade IV malignant astrocytoma explants. The explanted tumors were genetically characterized at the p53, MDM2 and INK4a/ARF loci, and fibronectin expression and growth characteristics were examined. A recombinant adenovirus overexpressing EGR-1 was tested in the primary cell lines. Results Low levels of EGR-1 protein were found in all primary cultures examined, with lower values present in grade IV tumors and in cultures carrying wild-type copies of p53 gene. The levels of EGR-1 protein were significantly correlated to the amount of intracellular fibronectin, but only in tumors carrying wild-type copies of the p53 gene (R = 0,78, p = 0.0082). Duplication time, plating efficiency, colony formation in agarose, and contact inhibition were also altered in the p53 mutated tumor cultures compared to those carrying wild-type p53. Growth arrest was achieved in both types of tumor within 1–2 weeks following infection with a recombinant adenovirus overexpressing EGR-1 but not with the control adenovirus. Conclusions Suppression of EGR-1 is a common event in gliomas and in most cases this is achieved through down-regulation of gene expression. Expression of EGR-1 by recombinant adenovirus infection almost completely abolishes the growth of tumor cells in vitro, regardless of the mutational status of the p53 gene.

EGR-1 is involved in the regulation of cell responses to a wide array of stimuli such as mitogens, growth factors and stress stimuli [5-7]. Recent studies have shown that EGR-1 expression is altered in several types of neoplasia, compared to normal tissue [1,8,9]. Gene deletion or EGR-1 mutations have been reported in sporadic hematological malignancies [10]. EGR-1 expression has been found to be either decreased or undetectable in human breast cancer tissue and small cell lung carcinoma [11,12]. EGR-1 is altered in a different manner in prostate cancer, where higher levels of EGR-1 expression are found correlated to more advanced stages of malignancy [13]. Abstract: the current global recession has cast doubts about the work of macroeconomists, the usefulness of their theories and their capacity to anticipate and deal with crises. this paper discusses that, while incapable of dealing with all the complexities of the real world, models do help to interpret the economic reality and propose economic policy recommendations. it also claims that, although crises are unpredictable, it is necessary to continue searching for early warnings and policies to try to prevent them and mitigate their propagation and amplification. finally, it emphasizes that, although the initial shock of the current crisis was similar to the one of the great depression, its subsequent development has been essentially different, in particular in emerging economies. this proves that the economies are much stronger, and the policies applied have gone in the proper direction, which has benefited from important practical lessons and advances in economic research.

Abstract: this document analyzes the rationality of keeping international reserves, which are used as a type of insurance in the event of external financing problems and to mediate in the type of exchange rate. here it is argued that it is not possible to separate both objectives. this explains why the use of other forms of insurance, although potentially cheaper, is note generalized. on the other hand, the fact that in practice the use of the international reserves is very low, even during the crisis periods, is evidence of the dissuasive role they have - just the fact of keeping them reduces financial vulnerability. the evidence for the subprime crisis is studied, finding that the financial strains were fewer in economies with high reserve level. finally, diverse forms of measuring the adequate reserve level are discussed and the variation of the reserves and the type of exchange rate are examined during the asian and subprime crisis in emerging economies.

Abstract: Credit Markets and Stagnation in an Endogenous Growth Model This paper studies the effects that the inability of individuals to borrow against future income has on economic growth. The model assumes that human capitals, which is accumulated through education, is the onlv factor of production. It is shown that liquidity contraints reduce growth. Further, in the presence of externalities that may induce two equilibria, it is shown that liquidity constraints not only reduce the rate of growth in the highgrowth equilibrium, but can also make the low-growth equilibrium more likely to occur.

Abstract: Competencia Imperfecta y Políticas Comerciales This paper surveys the recent developments on trade policy under imperfect competition. It is shown that in an imperfectly competitive industry trade policy may be anoeffective tool to divert rents towards domestic firms and consumers. The robustness of the results to the specification of the oligopoly and the rest of the economy is also reviewed. Small changes in the assumptions about the characteristics of the industry can change drastically the policy implications. Some empirical evidence to measure the effects of trade policy in oligopolistic industries are also presented. Finally there is a discussion about the relevance of these new developments as policy guidelines.

Abstract: La actual recesión mundial ha puesto en tela de juicio la labor de los macroeco-nomistas, la utilidad de sus teorías, y su capacidad de anticipar y manejar las crisis. Este artículo discute que pese a que los modelos son limitados para abordar todas las complejidades de la realidad, nos ayudan a interpretarla y a tener recomendaciones de política económica. También argumenta que aunque las crisis son impredecibles, es necesario continuar en la búsqueda de alertas tempranas y políticas para intentar prevenirlas y mitigar su propagación y amplificación. Finalmente, se destaca que a pesar que esta crisis tuvo un shock inicial similar al de la Gran Depresión, su evolución posterior ha sido fundamentalmente distinta, en particular en economías emergentes. Esto demuestra que las economías son más sólidas y las políticas aplicadas han ido en la dirección correcta, lo que se ha nutrido de importantes lecciones de la práctica de la política económica y avances en la investigación.
