Gli2 Acetylation at Lysine 757 Regulates Hedgehog-Dependent Transcriptional Output by Preventing Its Promoter Occupancy

Abstract: The morphogenic Hedgehog (Hh) signaling regulates postnatal cerebellar development and its aberrant activation leads to medulloblastoma. The transcription factors Gli1 and Gli2 are the activators of Hh pathway and their function is finely controlled by different covalent modifications, such as phosphorylation and ubiquitination. We show here that Gli2 is endogenously acetylated and that this modification represents a key regulatory step for Hedgehog signaling. The histone acetyltransferase (HAT) coactivator p300, but not other HATs, acetylates Gli2 at the conserved lysine K757 thus inhibiting Hh target gene expression. By generating a specific anti acetyl-Gli2(Lys757) antisera we demonstrated that Gli2 acetylation is readily detectable at endogenous levels and is attenuated by Hh agonists. Moreover, Gli2 K757R mutant activity is higher than wild type Gli2 and is no longer enhanced by Hh agonists, indicating that acetylation represents an additional level of control for signal dependent activation. Consistently, in sections of developing mouse cerebella Gli2 acetylation correlates with the activation status of Hedgehog signaling. Mechanistically, acetylation at K757 prevents Gli2 entry into chromatin. Together, these data illustrate a novel mechanism of regulation of the Hh signaling whereby, in concert with Gli1, Gli2 acetylation functions as a key transcriptional checkpoint in the control of morphogen-dependent processes.

Abstract: a hemicellulose fraction was obtained after acid hydrolysis of eucalyptus chips soaked in sulfuric acid. the hydrolysate was used as the substrate to grow candida guilliermondii fti 20037. the influence of the nutrients (ammonium sulfate, calcium chloride, rice bran), ph, xylose concentration in the eucalyptus chips hemicellulosic hydrolisate and fermentation time were studied indicating that the xylitol production is mainly influenced by nutrients: ammonium sulfate and rice bran; initial ph of fermentation and by the xylose concentration in the eucalyptus chips hemicellulosic hydrolisate.
Dental Treatment with 30% Trichloroacetic Acid in a Patient with Moderate Hemophilia A  [PDF]

We present the case of a 51-year-old moderate hemophiliac with gingival bleeding due to papillary inflammation and adherence in the molar area 47. After dental diagnosis a calculus was removed and 30% trichloroacetic acid was placed by a pressurized applicator for five seconds; the procedure was repeated until the bleeding stopped, then tranexamic acid was placed by a gauze. The patient chewed the gauze for 30 minutes and was discharged after repeating the same procedure for 30 minutes more. We observed how local treatment with trichloroacetic acid combined to tranexamic acid placed on the hemorrhagic site was able to stop the bleeding.

Abstract: In their works Jünger and Céline – a part from their meetings in Paris – have always dealt with the theme of the catastrophe, even in terms of apocalypse. This catastrophe has been considered both as an historical fact – in the experience of the First and Second World War – and a cultural phenomenon represented by nihilism – the philosophy through which the twentieth century showed its nature. For the French writer the catastrophe coincides with life itself, rather, it proves the unreasonableness of life, so that the apocalypse ultimately comes to represent a sort of universal catharsis. In Jünger, both the catastrophe and the apocalypse are means to a higher revelation, through which man can recover an original and poetic way of being in the world.

Abstract: This paper will focus on some specific aspects of Arnaut Daniel’s “D’autra guiza e d’autra razo” which, despite its success in its day and age (some verses were reused by Petrarch in the opening sonnet of the “RVF”), has received little critical attention in modern times. In particular, I will examine the incipit (whose alterity with respect to the rest of the Occitan lyric tradition points to an explicit declaration of intent on the part of the author), the lack of a tornada and, finally, the possibility that Arnaut Daniel was influenced by the section in the “Rhetorica ad Herennium” devoted to mnemonic technique. Moreover, the hypothesis that material concerning the art of memory has been used here helps to explain some particularly problematic textual loci, such as ll. 5-7 and 34.

Abstract: - The Others Ethics. Violence and Recognition in Healths relationships - The theme of recognition is central in the development of the educational relationship. It involves those social and health practices which – based on normativity, keep the bond between the individual and collectivity going. Starting from Hegel, intersubjectivity is unveiled in its double adaptive and conflictual function, setting out the problematic relation – already delineated by Kant, between autonomy and heteronomy.The essay referring to Hegel as well as to experts in contemporaneity such as Kojève, Foucault, Habermas and Honneth, analyses the different facets through which, in the health practices, the fabric of educational dialectic is woven.
