Probing the Cell Cycle with Flow Cytometry  [PDF]

Abstract: Flow cytometry is a versatile technique to study different aspects of the cell cycle from subpopulations of cells to detailed cell kinetic information. In this paper a basic review of cell kinetic parameters is presented followed by detailed descriptions of the different flow cytometric methodologies that can be used to extract pertinent information for a particular study. The methodologies range from simple DNA profile analysis, the use of bromodeoxyuridine to cell cycle-associated proteins such as the cyclins.

Abstract: background: total colectomy with ileorectal anastomosis (ira) is an alternative to the ileoanal pouch for the surgical treatment of ulcerative colitis in a selected group of patients. this technique leaves rectal mucosa hable to develop persistent proctitis, dysplasia and cancer aim: to describe short and long-term results of ira and to assess the presence of dysplasia. material and methods: descriptive study of patients treated with ira. the data were obtained from the clinical records, and the present status was evaluated with an interview. a proctoscopy and biopsy was offered free of cost to the contacted patients. results: between 1978 and 2005, 26 patients were operated. one patient presented an anastomotic leakage that was treated with a loop ileostomy there was no operative mortality. twenty-three patients were followed for a períod of 1 to 23 years. three patients evolved as crohn 's disease and two of them needed a proctectomy. three patients died of non-related diseases. in the remaining 17, the average evacuation rate was 3.7/24 h and all were continent. none developed a rectal cancer only two patients had their planned annual endoscopic surveillance. in 2 of the 11 patients who accepted endoscopy and biopsy, a low-grade dysplasia was found. conclusions: ira has low morbidity and acceptable functional results in this selected group of patients. no patient present high-grade dysplasia or cancer; however, the adherence to the endoscopic follow-up ispoor.

Abstract: to evaluate the association between cardiovascular risk factors (cvrf) during postmenopausal years and previous menstrual irregularity during reproductive years, we performed a case-control study in 414 postmenopausal women (mean age 60.4 ± 5.5 years; bmi 25.3 ± 4.7 kg/m2). the variables assessed were: menstrual cycle characteristics at age 20-35y (independent) and records of arterial hypertension, dyslipidemia, diabetes mellitus, and coronary heart disease (dependent). statistical analysis used the chi-square test and logistic regression, adjusting for potential confounders for cardiovascular risk, with significance set at 5%. women reporting previous menstrual irregularity were associated with increased risk for some cvrf [adjusted odds ratio (or) 2.14; ci-95%= 1.02-4.48], when compared with those reporting regular menstrual cycles. stratified analysis demonstrated significant associations of previous menstrual irregularity with: arterial hypertension [or= 2.74; ci-95%= 1.39-5.41), hypercholesterolemia (or= 2.32; ci-95%= 1.17-4.59), hypertriglyceridemia (or= 2.09; ci-95%=1.10-4.33), and coronary angioplasty (or= 6.82; ci-95%= 1.44-32.18). these data suggest that a prior history of menstrual irregularity, as indicative of polycystic ovary syndrome, may be related to increased risk for cvd during postmenopausal years.

Abstract: background: appendiceal tumors correspond to 0.5% of all gastrointestinal tumors and their prognosis depends on the type of lesion. aim: to analyze the clinical and pathological features of appendiceal tumors and their prognosis. material and methods: review of medical records of patients in whom an appendiceal tumor was detected during the pathological study in a clinical hospital, between 1981 and 2006. results: sixty seven patients were identified (mean age 45.3+ 19.6 years, 39 females). fifty nine were operated due to the suspicion of an acute appendicitis, that was confirmed pathologically in 37. thirty six patients had a neuroendocrine tumor, 18 had a benign tumor, seven an adenocarcinoma, five an adenocarcinoid and one a neuroendocrine plus a mucinous cystadenoma. a hemicolectomy was performed in a second operation in four patients with a carcinoid tumor, three with an adenocarcinoma, one with an adenocarcinoid and in one with a peritoneal pseudomyxoma. five years survival was 100% in patients with benign tumors, 97% in those with neuroendocrine tumors, 75% in those with adenocarcinoid tumors and 62% in those with adenocarcinoma. conclusions: most appendiceal tumors were discovered during the pathological study of the surgical piece of appendicectomies performed due to the clinical suspicion of an acute appendicitis. survival of patients was dependent on the type of tumor

Abstract: this article is a bibliographical analysis of the problem of learning environments. it does not intend to be thorough, but to contribute to the conceptual delimitation of this problem. although the transformations of contemporary cultures have questioned the monopoly that the school has traditionally had over educational issues; currently, the school still one of the most important social learning environments. it is therefore, necessary to rethink learning environments such as the classroom, from diverse and complex perspectives that approach the learning environment from ludic activities, aesthetics and the new technological mediations, in order to highlight the axis on which we could base a much deeper reflection on contemporary education beyond the instrumentalist, transmissionist and disciplinary principles.

A psicanálise como linguagem social: o caso argentino

Abstract: the recent book by mariano plotkin about the "psychoanalytical culture" in argentina brings forth ample discussions about the role of psychoanalysis in the dynamics of different national societies within the scope of modern western culture. the comparison between argentina and brazil is particularly rich, as also in the latter a significant diffusion of psychoanalysis took place during an homologous period. plotkin presents a very complex and minute analysis of the phenomena in focus. his analysis reveals the main constitutive tensions of argentine history during the xxth century, approaching the relationships with psychiatry, medicine, psychology, as well as with "peronism" and lef-wing militancy. i emphasize the importance of his demonstration that "psychoanalytical culture" has become a "social language" in argentine society, as well as the importance of his description of the diffusion of psychoanalysis by mass communication (which led to a wider social range of reception than in brazil). i also emphasize the analytical relevance of his demonstration of the association between this diffusion and a generalized "scientism" of argentine society, which is connected to the particular characteristics of that country's cultural "modernization".

O fen meno em quest o mereceu de Plotkin uma análise muito complexa e minuciosa, reveladora das principais tens es constitutivas da história argentina ao longo do século XX, envolvendo as rela es com a psiquiatria e a medicina, com a psicologia, com o "peronismo" e com a militancia de esquerda.
