Cytochrome C Biosensor—A Model for Gas Sensing

Abstract: This work is about gas biosensing with a cytochrome c biosensor. Emphasis is put on the analysis of the sensing process and a mathematical model to make predictions about the biosensor response. Reliable predictions about biosensor responses can provide valuable information and facilitate biosensor development, particularly at an early development stage. The sensing process comprises several individual steps, such as phase partition equilibrium, intermediate reactions, mass-transport, and reaction kinetics, which take place in and between the gas and liquid phases. A quantitative description of each step was worked out and finally combined into a mathematical model. The applicability of the model was demonstrated for a particular example of methanethiol gas detection by a cytochrome c biosensor. The model allowed us to predict the optical readout response of the biosensor from tabulated data and data obtained in simple liquid phase experiments. The prediction was experimentally verified with a planar three-electrode electro-optical cytochrome c biosensor in contact with methanethiol gas in a gas tight spectroelectrochemical measurement cell.

Abstract: Here, we systematically investigated the expression of TRPM5 in rat and mouse tissues. Apart from taste buds, where we found TRPM5 to be predominantly localized on the basolateral surface of taste receptor cells, TRPM5 immunoreactivity was seen in other chemosensory organs – the main olfactory epithelium and the vomeronasal organ. Most strikingly, we found solitary TRPM5-enriched epithelial cells in all parts of the respiratory and gastrointestinal tract. Based on their tissue distribution, the low cell density, morphological features and co-immunostaining with different epithelial markers, we identified these cells as brush cells (also known as tuft, fibrillovesicular, multivesicular or caveolated cells). In terms of morphological characteristics, brush cells resemble taste receptor cells, while their origin and biological role are still under intensive debate.We consider TRPM5 to be an intrinsic signaling component of mammalian chemosensory organs, and provide evidence for brush cells being an important cellular correlate in the periphery.Transient receptor potential (TRP) proteins form a large gene family of ion channels characterized by distinct activation mechanisms and biophysical properties. By sequence homology, members of the family fall into six subfamilies (TRPC, TRPV, TRPM, TRPML, TRPP, and TRPA). There is mounting evidence that TRP channels are involved in thermosensation, mechanosensation, smell and taste. A subset of TRP channels, called 'thermo-TRPs' (TRPV1-TRPV4, TRPA1 and TRPM8), have been found to be highly temperature dependent and are directly involved in heat and cold sensation in the peripheral nervous system [1]. Several TRP channels are mechanosensitive or activated by hypotonic challenge (TRPV4, TRPA1, TRPM3, PKD1 and TRPP2) [2]. Abstract: L’article analyse le paradoxe de la conception communiste du citoyen engagé : au niveau idéologique, citoyenneté et engagement sont intrinsèquement liés, au niveau politique ils sont souvent radicalement dissociés. Le citoyen communiste est plus engagé que citoyen. La citoyenneté était dépourvue de plusieurs de ses attributs : de la jouissance de la loi qui établit l’égalité arithmétique et minorise les ma tres ; de la citoyenneté active – civile et politique – pour ne jouir que la protection sociale ; de la participation au profit de l’appartenance. L’individu communiste était engagé, non pas parce qu’il l’avait choisi dans la liberté, mais parce qu’il n’avait pas la liberté de se désengager. L’article compare l’engagement communiste et postcommuniste en analysant le second au croisement de l’individualité et de la citoyenneté et en distinguant trois formes de désengagement – contestataire, bonapartiste et individualiste et trois formes d’engagement – mimétique, héro que, civique. The communist citizen is more engaged than citizen. The citizenship is deprived of several of its attributes: of the law which establishes "the arithmetic equality" and minimises the masters; of the active citizenship, civic and political, enjoying just the social one; of the participation for emphasizing on the belonging. The communist individual is engaged not because he has chosen freely to be such, but because he had not the freedom to disengage him/herself. The article compares the communist and the post communist participation analysing the latter at the intersection of individuality and citizenship and distinguishing three forms of disengagement – contesting, "bonapartist", and individualist, as well as three forms of engagement: mimetic, heroic, civic. Fast Algorithm for Vectorcardiogram and Interbeat Intervals Analysis: Application for Premature Ventricular Contractions Classification

Abstract: In this study we investigated the adequacy of two non-orthogonal ECG leads from Holter recordings to provide reliable vectorcardiogram (VCG) parameters. The VCG loop was constructed using the QRS samples in a fixed-size window around the fiducial point. We developed an algorithm for fast approximation of the VCG loop, estimation of its area and calculation of relative VCG characteristics, which are expected to be minimally dependent on the patient individuality and the ECG recording conditions. Moreover, in order to obtain independent from the heart rate temporal QRS characteristics, we introduced a parameter for estimation of the differences of the interbeat RR intervals. The statistical assessment of the proposed VCG and RR interval parameters showed distinguishing distributions for N and PVC beats. The reliability for PVC detection of the extracted parameter set was estimated independently with two classification methods - a stepwise discriminant analysis and a decision-tree-like classification algorithm, using the publicly available MIT-BIH arrhythmia database. The accuracy achieved with the stepwise discriminant analysis presented sensitivity of 91% and specificity of 95.6%, while the decision-tree-like technique assured sensitivity of 93.3% and specificity of 94.6%. We suggested possibilities for accuracy improvement with adequate electrodes placement of the Holter leads, supplementary analysis of the type of the predominant beats in the reference VCG matrix and smaller step for VCG loop approximation.
