Molecular characteristics and chromatin texture features in acute promyelocytic leukemia

Abstract: In a prospective study, we analyzed peripheral (PB) leukocyte count, immunophenotype, methylation status of CDKN2B, CDKN2A and TP73; FLT3 and NPM1 mutations besides nuclear chromatin texture characteristics of the leukemic cells. We also examined the relation of these features with patient’s outcome.Among 19 cases, 4 had a microgranular morphology, 7 presented PB leukocytes >10x109/l, 2 had FLT3-ITD and 3 had FLT3-TKD (all three presenting a methylated CDKN2B). NPM1 mutation was not observed. PB leukocyte count showed an inverse relation with standard deviation of gray levels, contrast, cluster prominence, and chromatin fractal dimension (FD). Cases with FLT3-ITD presented a microgranular morphology, PB leukocytosis and expression of HLA-DR, CD34 and CD11b. Concerning nuclear chromatin texture variables, these cases had a lower entropy, contrast, cluster prominence and FD, but higher local homogeneity, and R245, in keeping with more homogeneously distributed chromatin. In the univariate Cox analysis, a higher leukocyte count, FLT3-ITD mutation, microgranular morphology, methylation of CDKN2B, besides a higher local homogeneity of nuclear chromatin, a lower chromatin entropy and FD were associated to a worse outcome. All these features lost significance when the cases were stratified for FLT3-ITD mutation. Methylation status of CDNK2A and TP73 showed no relation to patient’s survival.in APL, patients with FLT3-ITD mutation show different clinical characteristics and have blasts with a more homogeneous chromatin texture. Texture analysis demonstrated that FLTD-ITD was accompanied not only by different cytoplasmic features, but also by a change in chromatin structure in routine cytologic preparations. Abstract: the objective of this study was to evaluate, under field conditions, in colina-sp, brazil, the response of two corn cultivars to six soil compaction levels of an oxisol regarding the root system development, and to characterize soil compaction by the least limiting water range (llwr). the experiment was arranged in a randomized complete block design, with strip plots and four replications. the corn hybrids dkb 390 and das 2b710 were used. after corn sowing, undisturbed soil samples were collected to determine physical properties and llwr. at the tasseling stage of the corn plants, three soil samples per plot were taken from the crop inter-row, from the layers 0-10, 10-20 and 20-30 cm, to measure mean root diameter, length density and dry mass. the llwr was equal to zero when soil density reached the critical level of 1.46 t m-3, corresponding to 92 % of the maximum soil density. greater soil compaction resulted in greater root dry mass, length density, and diameter. in the 0-10 cm layer an increase in root dry mass and density was possible up to a penetration resistance of 1.23 mpa and 1.43 mpa, respectively. root dry mass, density and diameter were negatively correlated with the yield of irrigated corn, and therefore represent good soil compaction indicators.
Comparative study between the effects of hyaluronic acid and acid galactan purified from eggs of the mollusk Pomacea sp in wound healing

Abstract: purpose: to compare the effect of hyaluronic acid (ha) and of ag on the healing of intestine wounds. methods: the semi-purified extract of the eggs of the mollusc was obtained by fractionation with ammonium sulfate and purification for ion-exchange chromatography. the obtained galactans were eluted in water (neutral galactan) and in 0.1 and 0.2m nacl (acidic galactans). the in vivo study was performed with 45 "wistar" rats, separated in three groups (n=15). solutions containing ha 1%, ga 1% or saline solution 0,9%, was placed topically on the sutures of wounds in the small intestine of the rats. after 05, 10 and 21 days the animals were sacrificed and biopsy of the healing tissue was done. results: the hystologic grading was more significant for ha and ag groups when compared to the group c. ag stimulated the appearance of macrophages, giant cells and increase in the concentration of collagen in the area of the wound when compared to ha. conclusion: the topical use of ga in intestinal wounds promoted the anticipation of events that are important in the wound healing.

Abstract: As escalas desenvolvidas para a avalia o da dor em idosos n o comunicantes est o insuficientemente estudadas para garantir a sua validade e fiabilidade. O objetivo deste estudo foi adaptar culturalmente para o Português a PAINAD e avaliar as suas propriedades psicométricas em doentes idosos (com ou sem demência) internados em servi os hospitalares com patologia médica e/ou cirúrgica. Estudo descritivo e transversal com o desenvolvimento de painéis e aplica o da escala de forma simultanea e independente a 99 doentes seleccionados de forma acidental. O processo de valida o cultural foi fácil de obter, apesar da necessidade de gerar consenso em rela o aos termos: frigtened , facial grimacing , Knees pulled up e moan . A análise dos componentes principais revelou um factor com variancia explicada de 61,09 %. A consistência interna calculada pelo a de Cronbach foi de 0,84. O nível de concordancia variou entre um valor de Kappa de 0,49 e 0,78. O coeficiente de correla o intraclasse foi de 0,89. A vers o Portuguesa da escala PAINAD quando utilizada em doentes idosos (com ou sem demência) e com patologia médica e/ou cirúrgica internados em servi os hospitalares revela ser válida e fiável. Sugere-se o seu uso por profissionais de saúde com forma o específica na aplica o da escala. Las escalas desarrolladas para la evaluación del dolor en ancianos con problemas comunicativos no han sido aún suficientemente estudiadas para garantizar su validad y fiabilidad. El objeto de este estudio fue adaptar culturalmente al portugués la PAINAD y evaluar sus propiedades psicométricas en pacientes ancianos (con o sin demencia) ingresados en servicios hospitalarios con patología médica y/o quirúrgica. Estudio descriptivo y transversal con el desarrollo de paneles y la aplicación de la escala de forma simultánea e independiente a 99 pacientes seleccionados de forma accidental. El proceso de validación cultural fue fácil de obtener, a pesar de la necesidad de generar consenso con respecto a los términos: asustado/a , muecas faciales , rodillas levantadas y gemidos . El análisis de los componentes principales reveló un factor con una varianza explicada de 61,09 %. La consistencia interna calculada por el a de Cronbach fue de 0,84. El nivel de concordancia varió entre un valor de Kappa de 0,49 y 0,78. El coeficiente de correlación intraclase fue de 0,89. La versión portuguesa de la escala PAINAD, cuando se utiliza en pacientes adultos mayores (con o sin demencia) y con patología médica y/o quirúrgica, ingresados en servicios hospitalarios, resulta ser válida y fiable

Abstract: the scales developed for pain assessment in non-communicative elderly persons have not been sufficiently tested to ensure their validity and reliability. the aim of this study was to culturally adapt to portuguese the painad scale and to assess its psychometric properties with elderly patients (with or without dementia) who are hospitalized with medical and/orsurgical conditions. this descriptive and cross-sectional study was carried out with panels and a simultaneous and independent application of the scale with 99 patients who were both accidentally selected. the process of cultural validation was easily obtained, in spite of the need to generate a consensus regarding the terms: ?frightened?, ?facial grimacing?, ?knees pulled up? and ?moaning?. analysis of the main components revealed a factor with an explained variance of 61,09%. internal consistency calculated by cronbach’s alpha equaled 0,84. the level of concordance varied between a value of kappa ranged from 0,49 to 0,78. the interclass correlation coefficient was 0,89. the portuguese version of the painad scale, when used with hospitalized elderly patients (with or without dementia) and with a medical and/or surgical condition reveals validity and reliability. however, this scale should be used by healthcare professionals with specific training in its application.

Abstract: this research investigated social representations of science teachers working at the 5th-8th grades in the scientific method and the effects of those representations on their educational practices. the methodological theoretical reference used was that of the social representations, in the moscovician perspective. the instruments used for the research were questionnaires and interviews. the analysis, although with some incoherence, points to a prevalence of constructivist representations. in relation to the effects that those representations have on the subjects' practices, we concluded that this happens in part. in general the teachers evaluate as advantageous the use of that methodology in the teaching of sciences, justifying the fact that they don't use it as due to some operational difficulties and even due to educational pressures.

Abstract: As reconstru es identitárias que vem ocorrendo nas Américas est o interligadas com o fen meno dos estados-nac o, uma vez que estes s o conseqüências dos processos de coloniza o que dominaram povos e desintegrou culturas. A quest o nacional iniciada no mundo colonial americano desde o século XVIII com os movimentos deindependência, para a América Latina se configura de modo contraditório posto que tais estados-na o passaram a ser a configura o do poder daqueles que deram continuidade aos interesses e prestígios do poder colonial. O que significou para os povos dominados - índios e negros - a permanência da colonialidade. Neste artigo pretende-se estudar como esses povos que apresentam suas reconstru es identitárias nesse movimento contra-hegemonico na regi o fronteiri a do Brasil com a Guiana. Pretende-se aqui estudar com base nas categorias de identidade, cultura e nacioanalidades. Abstract The reconstruction of identities that is taking place in the Americas is interconnected with the phenomenon of the Nation-State, for they are the consequences of colonization processes that have dominated peoples and disintegrated cultures. The national question - which began in the American colonial world as of the 1800’sindependence movements in Latin America – has been articulated with contradictions. The resulting Nation-States took on the configuration of power of those who continued to pursue the same set of interests and privileges of the colonial power. For those under domination – native and black people – this meant the continuation of colonialism.This paper will investigate the ways through which these peoples have reconstructed their identities within a counter-hegemonic movement in the Brazilian and Guiana frontier. My goal is to study culture and nationalities based on categories of identity.
Preventive but Not Curative Efficacy of Celecoxib on Bladder Carcinogenesis in a Rat Model

Abstract: To evaluate the effect of a cyclooxygenase 2 inhibitor, celecoxib (CEL), on bladder cancer inhibition in a rat model, when used as preventive versus as curative treatment. The study comprised 52 male Wistar rats, divided in 5 groups, during a 20-week protocol: control: vehicle, carcinogen: 0.05% of N-butyl-N-(4-hydroxybutyl) nitrosamine (BBN), CEL: 10?mg/kg/day of the selective COX-2 inhibitor Celebrex, preventive CEL (CEL+BBN-P), and curative CEL (BBN+CEL-C) groups. Although tumor growth was markedly inhibited by the preventive application of CEL, it was even aggravated by the curative treatment. The incidence of gross bladder carcinoma was: control 0/8(0%), BBN 13/20(65%), CEL 0/8(0%), CEL+BBN-P 1/8(12.5%), and BBN+CEL-C 6/8(75%). The number and volume of carcinomas were significantly lower in the CEL+BBN-P versus BBN, accompanied by an ample reduction in hyperplasia, dysplasia, and papillary tumors as well as COX-2 immunostaining. In spite of the reduction of tumor volumes in the curative BBN+CEL-C group, tumor malignancy was augmented. An anti-inflammatory and antioxidant profile was encountered only in the group under preventive treatment. In conclusion, preventive, but not curative, celecoxib treatment promoted a striking inhibitory effect on bladder cancer development, reinforcing the potential role of chemopreventive strategies based on cyclooxygenase 2 inhibition. 1. Introduction Bladder cancer is a prevalent tumor, accounting for 5%–10% of all malignancies in Western countries [1, 2]. It has a high recurrence and progression rate and the prognosis, except for superficial forms, is poor [3]. Furthermore, it has high mortality rates and socioeconomic costs [4, 5]. The conventional surgical techniques and therapeutic options might cause discomfort to patients, especially in invasive and aggressive forms of cancer [6, 7]. Therefore, the improvement of bladder cancer management and treatment could rely on better preventive strategies. The identification of promising drugs remains dependent on a better elucidation of the molecular/cellular mechanisms underlying cancer appearance and progression [8, 9]. Apart from the genetic features and markers already characterized [10, 11], the cellular and molecular mechanisms for development and/or progression might involve inflammatory, proliferative, and oxidative stress phenomena that should be better elucidated.
