Trace glucose and lipid metabolism in high androgen and high-fat diet induced polycystic ovary syndrome rats

Abstract: Female Sprague-Dawley rats were divided into 3 groups: the control group(C), n = 10; the andronate-treated group (Andronate), n = 10 (treated with andronate, 1 mg/100 g body weight/day for 8 weeks); and the andronate-treated and high-fat diet group (Andronate+HFD), n = 10. The rate of glucose appearance (Ra of glucose), gluconeogenesis (GNG), and the rate of glycerol appearance (Ra of glycerol) were assessed with a stable isotope tracer. The serum sex hormone levels, insulin levels, glucose concentration, and the lipid profile were also measured.Compared with control group, both andronate-treated groups exhibited obesity with higher insulin concentrations (P < 0.05) but similar blood glucose concentrations. Of the two andronate-treated groups, the andronate+HFD group had the most serious insulin resistance (IR). Estrus cycles were completely acyclic, with polycystic ovaries and elevated serum lipid profiles in the andronate+HFD group (P < 0.05). Ra of glucose and GNG increased significantly in the andronate+HFD rats. However, the Ra of glycerol was similar in the three groups.Andronate with HFD rat model showed ovarian and metabolic features of PCOS, significant increase in glucose Ra, GNG, and lipid profiles, as well as normal blood glucose levels. Therefore, aberrant IR, increased glucose Ra, GNG, and lipid metabolism may represent the early-stage of glucose and lipid kinetics disorder, thereby might be used as potential early-stage treatment targets for PCOS.Polycystic ovary syndrome (PCOS) is one of the most common endocrine disorders in women of reproductive age [1] and is the most frequent cause of hyperandrogenism and anovulation [2]. Abstract: Studies were identified by searching MEDLINE and PubMed databases for articles from January 1990 to December 2009, using the keywords "cholangiocarcinoma," "perineural invasion," "nerve growth factor"(NGF), "neural cell adhesion molecule" (NCAM), "matrix metalloproteinase"(MMP), "neurotransmitter," "acetylcholine" (Ach), and "transforming growth factor" (TGF)." Additional papers and book chapters were identified by a manual search of references from the key articles.From above we found that the molecules NGF, NCAM, MMP, Ach and TGF may have prognostic significance in, and offer clues to the mechanism of CCA neural invasion.Cholangiocarcinoma's increasing worldwide incidence is especially poignant in view of both the lacking effective therapies, and the fact that it is commonly diagnosed in advanced stages. As CCA neural invasion often appears early, more complete characterization of its molecular pathology could lead to the identification of targets for the diagnosis and therapy of this devastating malignancy.Cholangiocarcinoma (CCA) is a malignant tumor originating from biliary tract epithelial cells. Among primary liver tumors, CCA incidence is only less than that of liver cancer[1,2], and it is becoming the most common hepatic tumor-induced death[3].Due to its difficulty of diagnosis and high fatality rate, cholangiocarcinoma is extremely destructive, currently surgery is the only therapeutic mode offering a cure. A simplified tether model for molecular motor transporting cargo

Abstract: Molecular motors are proteins or protein complexes which function as transporting engines in biological cells. This paper models the tether between motor and its cargo as a symmetric linear potential. Different from Elston and Peskin's work for which performance of the system was discussed only in some limiting cases, this study produces analytic solutions of the problem for general cases by simplifying the transport system into two physical states, which makes it possible to discuss the dynamics of the motor--cargo system in detail. It turns out that the tether strength between motor and cargo should be greater than a threshold or the motor will fail to transport the cargo, which was not discussed by former researchers yet. Value of the threshold depends on the diffusion coefficients of cargo and motor and also on the strength of the Brownian ratchets dragging the system. The threshold approaches a finite constant when the strength of the ratchet tends to infinity.

Abstract: This paper proposed a Memetic algorithm, which simulated the process of culture evolution, to solve vehicle routing problem with time windows(VRPTW). To convert VRPTW into continuous problem, it designed a real coding method. The memetic algorithm helped the particles which had certain learning capacity accelerate convergence rate by local search strategy. Meanwhile, because of helping some particles which fell into the local optimum escape from local optimum by tabu search,it enhanced the diversity of swarm. The experimental result shows that the proposed algorithm can get the solution more effectively and it is an effective method for VRPTW.

Abstract: A new algorithm of image Layer-presentation was proposed. The key concept of the algorithm was in that the image grayscale function f (x,y), which was comparatively irregular, was approximated by a series of high-regular grayscale functions gn(x,y). The algorithm had the feature of fast convergence and so it was a good approximator. Due to its high-regularity, gn might be stored in a quite small memory space. Thus, the algorithm gives a new way for data processing and reconstruction of biological images.

Abstract: Biomolecular motors are tiny engines that transport materials at the microscopic level within biological cells. In recent years, Elston and Peskin et al have investigated the effect of the elastic properties of the tether that connects the motor to its cargo at the speed of the motor. In this paper we extend their work and present a tether in the form of symmetric linear potential.Our results show that when the driving mechanism is an imperfect Brownian ratchet, the average speed decreases as the stiffness of the tether increases in the limit of large motor diffusion coefficient, which is similar to the results of Elston and Peskin.However, a threshold for the stiffness of the tether connecting the motor to its cargo is found in our model. Only when the tether is stiffer than the threshold can the motor and its cargo function co-operatively, otherwise, the motor and its cargo depart from each other. This result is more realistic than that of the spring model of Elston and Peskin.

Abstract: This paper proposed a segmentation method for MELK images based on a watershed algorithm, which was applied to segment the MELK images that include little information. Employed several techniques to solve the over-segmentation problem in the watershed algorithm. Firstly, used a Gaussian lowpass filter to enhance the MELK images. Provided weighted difference operators to obtain the gradient images, and processed the gradient images by a watershed algorithm after thresholded them by the Ostu method. At last, segmented MELK images based on a region merging algorithm called RAG method. The experimental results indicate that the proposed method is feasible to segment the MELK images which include little information.

Abstract: We have investigated two types of Zn_ 1-x Cd_xSe quantum islands in CdSe/ZnSe heterostructure with different size and Cd composition using micro-photoluminescence and micro-Raman spectroscopy. The micro-photoluminescence spectra at 4.2 K indicate that the photoluminescence peaks of the islands have a large red-shift of 166 meV when the thickness of CdSe deposition layer increased from 1.8 ML to 2.3 ML, which can't be explained by considering the change of quantum confinement potential only. We also found two other important mechanisms which may lead to the large red-shift by comparing the micro-photoluminescence and micro-Raman spectra. First, the sheet density of large Zn_ 1-x Cd_xSe islands which have lower exciton ground state energy will increase and the large islands will dominant gradually the photoluminescence properties of the samples with increasing thickness of the CdSe layer. Second, the increase of Cd composition in the Zn_ 1-x Cd_xSe quantum islands contributes to the large red-shift as well. This is verified by resonant Raman scattering spectra of the samples.

Abstract: We have investigated the temperature_dependent photoluminescence of two types of quantum islands in ultrathin CdSe/ZnSe layers. At low temperatures, excitons in these two types of quantum islands can transfer by tunneling. The hopping process of the localized excitons gradually becomes the dominant mechanism with increasing temperature. We found that the small islands are nano_scale clusters with quasi_zero_dimensional properties, which are different from those of the large islands. Experiments indicated that these two types of islands are optically correlated although their excitonic properties are different.

Abstract: Bacterium strain EVA17 was isolated from an oil-contaminated soil, and identified as Sphingomonas sp. based on analysis of 16S rDNA sequence,cellular fatty acid composition and physiological-chemical tests. The salicylate hydroxylase and catechol 2,3-dioxygenase(C23O) were detected in cell-free lysates, suggesting a pathway for phenanthrene catabolism via salicylate and catechol. Alignment showed that both of the C23O and GST genes of the strain EVA17 had high similarity with homologues of strains from genus Sphingomonas. The phylogenetic analysis based on 16S rDNA and C23O gene sequence indicated that EVA17 should be classified into genus Sphingomonas, although the two phylogenetic trees were slightly different from each other. The results of co-amplification and sequence determination indicated that GST gene should be located upstream of the C23O gene.
