Changes in the anterior segment of eyes of children with different genetic background under different accommodations

Abstract: 目的 研究和分析不同遗传背景下,相同屈光状态儿童在不同的调节刺激下眼前节结构的变化。方法 采集正视眼儿童(+0.50~-0.50 DS)60例(60眼),分成2组:A组30例,儿童父母均为正视;B组30例,父母均为高度近视(≥6.0 DS)。所有受试儿童行散瞳主觉验光,调节放松和不同程度调节刺激下测量前房深度、晶状体厚度及暗瞳直径,比较两组间的差异。结果 两组儿童的年龄、调节幅度和裸眼视力差异均无统计学意义(P>0.05)。调节放松和诱发3D调节时两组间各测量指标的差异无统计学意义(P>0.05)。诱发6D调节时,A组前房深度变浅值和晶状体变厚值均大于B组(P<0.05),但暗瞳直径缩小值两组之间的差异无统计学意义(P>0.05)。结论 在6D调节刺激下,父母均为高度近视的儿童眼前节结构中前房深度以及晶状体厚度变化值均小于父母均为正视的儿童,这种差异是否造成今后屈光转归的不同尚需进一步研究。
: Objective To investigate and analyze the changes in the anterior segment of eyes of children with different genetic background and same refraction under different accommodations. Methods A total of 60 emmetropic children (60 eyes, +0.50~-0.50 DS) were enrolled and divided into A group (n=30) and B group (n=30). Parents of children of A group were emmetropic and parents of children of B group were highly myopic (≥6.0 DS). All children underwent subjective refraction after mydriasis. The anterior chamber depth, crystalline lens thickness, and dark pupil diameter were measured under relaxation and different accommodations. The difference between two groups was compared. Results The differences of age, accommodation amplitude, and naked vision between two groups were not statistically significant (P>0.05). The differences of measurements under relaxation and accommodation of 3D between two groups were not statistically significant (P>0.05). The decrease of anterior chamber depth and the increase of crystalline lens thickness of A group under accommodation of 6D were larger than those of B group (P<0.05), while the difference of the decrease of dark pupil diameter between two groups was not statistically significant (P>0.05). Conclusion Changes of anterior chamber depth and crystalline lens thickness in the anterior segment of eyes of children with highly myopic parents under accommodation of 6D are smaller than those of children with emmetropic parents. Whether this difference leads to the difference of refraction in the future still needs further study

Abstract: 尿素和丙二醇合成碳酸丙烯酯(PC)在热力学上是可进行的.根据ZnO对尿素和丙二醇的催化反应性能,讨论了合成PC的反应历程.结果表明,该反应是分步进行的,首先尿素分解生成氨气和异氰酸,异氰酸与ZnO作用形成异氰酸物种,在丙二醇的作用下生成中间产物羟丙基氨基甲酸酯(HPC),然后由HPC脱氨气生成PC. ZnO在尿素分解和HPC脱氨气转化为PC的反应过程中起催化作用.

Abstract: RNA biogenesis, including biosynthesis and maturation of rRNA, tRNA and mRNA, is a fundamental process that is critical for cell growth, division and differentiation. Previous studies showed that mutations in components involved in RNA biogenesis resulted in abnormalities in gametophyte and leaf development in Arabidopsis. In eukaryotes, RNases P/MRP (RNase mitochondrial RNA processing) are important ribonucleases that are responsible for processing of tRNA, and transcription of small non-coding RNAs. Here we report that Gametophyte Defective 1 (GAF1), a gene encoding a predicted protein subunit of RNases P/MRP, AtRPP30, plays a role in female gametophyte development and male competence. Embryo sacs were arrested at stages ranging from FG1 to FG7 in gaf1 mutant, suggesting that the progression of the gametophytic division during female gametogenesis was impaired in gaf1 mutant. In contrast, pollen development was not affected in gaf1. However, the fitness of the mutant pollen tube was weaker than that of the wild-type, leading to reduced transmission through the male gametes. GAF1 is featured as a typical RPP30 domain protein and interacts physically with AtPOP5, a homologue of RNases P/MRP subunit POP5 of yeast. Together, our data suggest that components of the RNases P/MRP family, such as RPP30, play important roles in gametophyte development and function in plants.
YAO is a nucleolar WD40-repeat protein critical for embryogenesis and gametogenesis in Arabidopsis

Abstract: Here we report the characterization of a mutation, yaozhe (yao), that causes zygote arrest and misplacement of cell plate of the zygote, leading to early embryo lethality. In addition, gametophyte development is partially impaired. A small portion of the mutant embryo sacs are arrested at four-nucleate stage with aberrant nuclear positioning. Furthermore, the competence of male gametophytes is also compromised. YAO encodes a nucleolar protein with seven WD-repeats. Its homologues in human and yeast have been shown to be components of the U3 snoRNP complex and function in 18S rRNA processing. YAO is expressed ubiquitously, with high level of expression in tissues under active cell divisions, including embryo sacs, pollen, embryos, endosperms and root tips.Phenotypic analysis indicated that YAO is required for the correct positioning of the first zygotic division plane and plays a critical role in gametogenesis in Arabidopsis. Since YAO is a nucleolar protein and its counterparts in yeast and human are components of the U3 snoRNP complex, we therefore postulate that YAO is most likely involved in rRNA processing in plants as well.The model plant Arabidopsis forms multicellular male and female gametophytes, namely pollen grains and embryo sacs that contain the sperm cells and egg cell respectively. During embryo sac development, the functional megaspore, one of the four meiotic products, undergoes three consecutive rounds of mitotic division to produce an eight-nucleate embryo sac, which cellularizes simultaneously to form a seven-celled female gametophyte composed of one egg, two synergids, three antipodal cells and a diploid central cell [1-4]. The pollen grain in Arabidopsis is a tricellular structure that contains two sperms and a large vegetative cell. Optimal Approximation of Linear Systems by Multi-agent Genetic Algorithm

Abstract: The problem of optimally approximating linear systems is solved by the multi-agent genetic algorithm(MAGA).In MAGA,each possible solution,a real-valued vectorin the search space,is considered as an agent,and all agents live in a latticelike environ-ment,with each agent being fixed at a lattice-point.In order to increase energies,theycompete or cooperate with their neighbors,and they can also use knowledge.Therefore,four evolutionary operators are designed for simulating the intelligent behaviors of agents,such as competition,cooperation,self-learning and so on.Making use of these agent-agentinteractions,MAGA realizes minimizing the objective function value.At the same time,asearch-space expansion scheme is adopted to find the regions where the optimal parameterslocate.In experiments,two linear systems,a stable one and an unstable one,are used totest the performance of MAGA,and a comparison is made between MAGA and two recentalgorithms.The results show that MAGA can find high quality approximate models withlow computational cost.
Chemical Composition and Antimicrobial Activity of the Essential Oil of Kumquat (Fortunella crassifolia Swingle) Peel

Abstract: The aim of this study was to determine the main constituents of the essential oil isolated from Fortunella crassifolia Swingle peel by hydro-distillation, and to test the efficacy of the essential oil on antimicrobial activity. Twenty-five components, representing 92.36% of the total oil, were identified by GC-MS analysis. The essential oil showed potent antimicrobial activity against both Gram-negative ( E. coli and S. typhimurium) and Gram-positive ( S. aureus, B. cereus, B. subtilis, L. bulgaricus, and B. laterosporus) bacteria, together with a remarkable antifungal activity against C. albicans. In a food model of beef extract, the essential oil was observed to possess an effective capacity to control the total counts of viable bacteria. Furthermore, the essential oil showed strongly detrimental effects on the growth and morphological structure of the tested bacteria. It was suggested that the essential oil from Fortunella crassifolia Swingle peel might be used as a natural food preservative against bacteria or fungus in the food industry.
Identifying Sources of Nitrate in Surface Water of Jiulong River Watershed

Abstract: Two representative catchments, Wuchuang catchment and Xiandu catchment, were selected from Jiulong River Watershed as study areas to trace sources of nitrate in surface water by using 15N isotope technique. In the spring of 2005, the surface water samples were twice collected from the catchments and determined for dissolved total nitrogen (DTN), nitrate nitrogen (NO3(-)-N), ammonia nitrogen (NH4+-N) and delta 15N value of nitrate. The results showed that in the samples from Xiandu catchment, the concentration ranges of DTN, NO3(-)-N and NH4+-N were 1.47-5.31 mg/L, 0.83-4.05 mg/L and 0.21-0.36 mg/L respectively, and the delta 15N values of nitrate ranged from 2.54 per thousand to 7.92 per thousand; in the samples from Wuchuang catchment, the concentration ranges of DTN, NO3(-)-N and NH4+-N were 1.14-5.56 mg/L, 0.96-1.46 mg/L and 0.12-1.28 mg/L respectively, and the delta 15N values of nitrate were between -0.19 per thousand and 5.89 per thousand. In contrast with the typical ranges of delta 15N isotopic signature of nitrate from different sources and in consideration of the information of local crop planting circumstances and application of fertilizers, the data indicated that the first, the nitrate in the surface water of Xindu catchment mainly came from inorganic fertilizers and soil organic nitrogen, and some originated from organic fertilizers; the second, the nitrate in the surface water of Wuchuang catchment mostly flowed from inorganic fertilizers and soil organic nitrogen, and few derived from organic fertilizers; the third, the sources of nitrate varied with the sampling location and time and were dependent on the kinds of local crops and their planting seasons.
The Construction of Shuttle Vectors of Brevibacillus brevis-Escherichia coli

Abstract: The 5' region of the cell wall protein(CWP) gene containing multiple tandem promoters and the signal peptide-coding sequence was isolated by PCR from Br. brevis 50, and used to construct the shuttle vector pBKE50, which included the replication origin of pUB110 and the erythromycin-resistance gene of pGK12. The alpha-amylase gene of Bacillus subtilis 168 was ligated to pBKE50, producing plasmid pBKE50/alpha-amy. After the resulting plasmid was introduced into Br. brevis 50, soluble and biologically active alpha-amylase was secreted directly into the culture medium. The expression level of alpha-amylase in the recombinant Br. brevis 50 was twice higher than that of the donor strain.
Establishment of the screening method and isolation of PQQ producing strains

Abstract: Pyrroloquinoline quinone (PQQ) is a cofactor of some oxido-reductases with many important physiological effects and potential pharmaceutical applications. The glucose dehydrogenase of Escherichia coli,being a candidate for enzymic detection of PQQ,is known to be a quinoprotein which is obligately dependant on PQQ as cofactor. The gdh gene of E. coli was amplified and cloned into plasmid pET28a. The recombinant GDH was overexpressed in soluble form in E. coli BL21(DE3). A bioassay method was established for determination of PQQ by the purified GDH. A screening model was set up for the enrichment of methylotrophic bacteria. Together with the above bioassay method,over 2000 soil samples were screened for the isolation of high-yielding PQQ producing strains. A methylotrophic strain,named MP606,was thus isolated. The PQQ production of MP606 is determined to be 113mg/L without conditional optimization and genetic breeding. The PQQ crystal was obtained from the culture supernatant which has been identified by HPLC,absorption spectra assay,and enzymatic analysis. The 16S rDNA of MP606 was amplified and sequenced. According to the comparison of 16S rDNA sequences,overall similarity value between strain MP606 and 12 typical methylotrophic bacteria is above 95%. The highest value is with two strains of Methylovorus,which reached at 99%.
