Purification and Partial Characterization of Polyphenol Oxidase from Sapodilla Plum (Achras sapota)  [PDF]

Abstract: The browning of fruits can be considered as an enzymatic oxidation which is believed to be one of the main causes of quality loss during post-harvest handling. The enzymes responsible for this are the oxidoreductases; the polyphenol oxidase (PPO) (monophenol, o-diphenol, oxygen oxidoreductase; EC 1.14.18.1) belongs to this group. This enzyme, which is found in the sapodilla plum (Achras sapota), was purified using a phenylsepharose and a SephacrylS-200 columns. The molecular weight of the purified enzyme was estimated to be about 66 kDa by gel filtration and 29 kDa by SDS-PAGE. A single protein band was found using the latter system (SDS-PAGE), which shows that the PPO of the pulp of the sapodilla plum may be composed of two protein subunits with similar molecular weight. The optimum pH was 7.0 and the optimum temperature 60. The most effective inhibitors tested were ascorbic acid, sodium metabisulfite and acetic acid.

Abstract: background: body dissatisfaction (bd) is considered as a predisposing, triggering, perpetuating and/or prognosis factor of eating disorders (ed). objective: to compare the presence of bd and psychopathological and/or behavioural features between female subjects with and without ed. method: comparatively the eatingattitudes test (eat-40), the eating disorder inventory (edi) and the body shape questionnaire (bsq) were administered to 52 ed female patients and to 51 female students without ed. results: patients reached significantly higher rates than female students without ed on bsq, eat-40, edi and subscales (p = < 0.001), except perfectionism (p = 0.015). bd assessed by edi (17 versus 4; p < 0.001) y el bsq (152 versus 58.5; p < 0.001) scored significantly higher in patients, mainly in anorexics on eat-40, bsq, bd, ineffectiveness and perfectionism (p < 0.001); bulimics on global edi, drive for thinness, interoceptive awareness, bulimia, maturity fears and interpersonal distrust (p < 0,001). the or of bd (edi) was 1.17 versus bsq (or = 1.05) (p < 0.001). the eat global score was directly and significantly correlated to bsq (r = 0.672; p < 0.001) in patients and to bd (edi) (r = 0.639; p < 0.001) in female students without ed. conclusions: bd is prevalent in an extreme degree within all the subtypes of ed. anorexics exhibited a higher bd as well as ineffectiveness and perfectionism. a higher bd is positively related to a higher ed symptomatology. the risk of developing an ed in a young woman that exhibits some degree of bd is very high.

Abstract: Antecedentes: La insatisfacción corporal (IC) es considerada factor predisponente, desencadenante, perpetuante y pronóstico de los trastornos alimentarios (TCA). Objetivos: Comparar la presencia de IC y características psicopatológicas/conductuales entre mujeres con y sin TCA. Método: A 52 pacientes con TCA y 51 estudiantes sin TCA, se les aplicó comparativamente el Test de Actitudes Alimentarias (EAT-40), el Inventario de Desórdenes Alimentarios (EDI) y el Cuestionario de Silueta Corporal (BSQ). Resultados: Las pacientes alcanzaron puntajes significativamente más elevados que las estudiantes en el BSQ, EAT-40, EDIy sub escalas (p = < 0,001), excepto perfeccionismo (p = 0,015). La IC medida por el EDI (17 versus 4; p < 0,001) y el BSQ (152 versus 58,5; p < 0,001) arrojó puntajes significativamente más altos en las pacientes, principalmente las anorécticas en el EAT-40, BSQ, IC, sentimientos de inutilidad personal y perfeccionismo (p < 0,001); las bulímicas en el EDI global, motivación por la delgadez, consciencia interoceptiva, bulimia, temor a la madurez y desconfianza interpersonal (p < 0,001). El ítem IC del EDI mostró un OR de 1,17, versus el BSQ (OR= 1,05) (p < 0,001). El puntaje global del EAT-40 se correlacionó directa y significativamente con el BSQ (r = 0,672; p < 0,001) en las pacientes y estudiantes con el ítem IC del EDI (r = 0,639; p < 0,001). Conclusiones: La IC está presente en grado extremo en todos los subtipos de TCA. Las anorécticas mostraron mayor IC acompa ada de sentimientos de inutilidad personal y perfeccionismo. Una mayor IC se relaciona positivamente con una mayor sintomatología compatible con TCA. El riesgo de padecerlo de una joven que exhibe algún grado de IC es altísimo. Abstract:

The extraction of anthocyanins present in the skin of the dragon fruit was performed using trifluoroacetic acid (TFA) plus a mixture of methanol, acetic acid and water; the anthocyanins were then purified with a LC-18 cartridge, using methanol acidified with TFA as eluent, reaching concentrations of 44.3865 ± 1.3125 mg/100g of sample. The extracts were put through stability tests under different storage conditions, modifying the pH of the extracts (pH of 1, 4 and 6), the temperature (4℃, 25℃ and 68℃) and the absence and presence of light for a time period of 4 days; the tests indiated that anthocyanins remain more stable at a temperature of 4℃ with a pH of 4 in the absence of light, retaining up to 80% of the pigment. Three anthocyanins were partially identified in the extracts by high performance liquid chromatography (HPLC); they were: cyanidin 3-O-glucoside, cyanidin 3,5 O-glucoside and pelargonidin 3,5 O-glucoside.

First remarks on the nesting biology of Hypodynerus andeus (Packard) (Hymenoptera, Vespidae, Eumeninae) in the Azapa valley, northern Chile

Abstract: first remarks on the nesting biology of hypodynerus andeus (packard) (hymenoptera, vespidae, eumeninae) in the azapa valley, northern chile. some aspects about the nesting biology of the potter wasp hypodynerus andeus (packard, 1869) are reported for the first time. observations were carried out at the azapa valley, coastal desert of northern chile. a total of sixty nests were collected and examined, each composed by 1-14 cells, most of them found attached to concrete lamp posts. the only preys recorded in the cells were geometridae (lepidoptera) caterpillars and the presence of the parasitoid anthrax sp. (diptera, bombyliidae) was also recorded. a number of arthropods belonging to different groups, mainly spiders, were found occupying empty nests.

Abstract: purulent pericarditis is an infrequent disease which affects the pericardium and has a high mortality rate. the diagnosis is suspected in a febrile patient with right cardiac failure and a primary infectious source. however, not all patients present in this form therefore the diagnosis and treatment can be difficult and a real challenge. this report is of a patient with this disease and other etiologic possibilities are discussed.

Abstract: the causistics of 52 patients with orbital tumors surgically treated at the institute of neurology and neurosurgery of cuba for 10 years, with the direct participation of the author in the cases attended during the last 4 years, is analyzed. the diagnostic and therapeutic elements of these occupying processes were studied in detail. the most common tumors were the meningiomas, the glyomas and the hemangiomas. the clinical and neuroophthalmological study oriented towards the topographic diagnosis in most of the cases and suggested, in some of them, the nature of the injury. the computerized axial tomography was the most used complementary study method, and it also provided the greatest number of data of interest. the types of lesions found in these patients are discussed and the main therapeutic modalities used are described, specially the routes of surgical approach to the orbit (56 operations), depending on the location or other characteristics of the injury

Abstract: Se analiza la casuística de 52 pacientes con tumores orbitarios tratados quirúrgicamente en el Instituto de Neurología y Neurocirugía de Cuba durante un período de diez a os, con participación directa del autor en los casos atendidos durante los últimos 4 a os, en la que se profundiza en los elementos diagnósticos y terapéuticos de estos procesos ocupativos. Los tipos tumorales más frecuentes fueron los meningiomas, los gliomas y los hemangiomas. El estudio clínico y neurooftalmológico orientó en el diagnóstico topográfico en la mayoría de los casos y sugirió en algunos la naturaleza de la lesión. La tomografía axial computadorizada fue el método de estudio complementario más utilizado y que más datos de interés aportó. Abstract: Durante tres a os se estudiaron los principales indicadores productivos de la vaquería “Modelo” perteneciente a la Estación Experimental de Zootecnia de la Universidad Central “Marta Abreu” de Las Villas. Inicialmente las vacas disponían de 48 cuartones de 0,31 Ha cada uno, con predominio de pastos naturales. La inclusión paulatina del clon CT-115 (Pennisetum purpureum) primero y del banco de proteínas después, en la alimentación de las vacas permitió incrementar los rendimientos de 4,68 a 6,00 L/vaca/día; así mismo el rendimiento L/Ha/a o se incrementó desde 1806 hasta 2405 litros en el a o 2000. Aunque de forma más discreta también se observó cierta mejoría en los indicadores reproductivos. Se concluye que la inclusión del clon CT-115 para el pastoreo en la época de seca y la utilización de los bancos de proteínas, permite aumentar la cantidad y calidad de la biomasa ofertada a las vacas lecheras, lográndose indicadores productivos más favorables. During three years the main productive indicators of "Modelo" dairy belonging to the Animal Husbandry Experimental Station at Las Villas “Marta Abreu” Central University, were studied. Initially the cows had 48 paddocks of 0,31 Ha each one, with prevalence of native grasses. The gradual inclusion of the clon CT-115 (Pennisetum purpureum) first and of the proteins bank later, in the feeding of the cows it allowed to increase the yields from 4,68 to 6,00 L/cow/day; likewise the yield L/Ha/year was increased from 1806 up to 2405 liters in the year 2000. Although in a more discreet way certain improvement was also observed in the reproductive indicators. We concluded that the inclusion of the clon CT-115 for grazing in dry period and the use of the proteins banks, allows to increase the quantity and quality of the biomass offered to dairy cows, achieving better productive indicators

Abstract:

Coastal zones are very dynamic and fragile environments, constituting a landscape ever more heterogeneous, fragmented and with increasing levels of complexity due to the changing relationship between man and nature. Integrated coastal zone management therefore requires detailed knowledge of the system and its components, based—to a large extent—on technical and scientific information. However, the information generated must be in line with the political requirements necessary for decision-making and planning. Thus the use of indicators to give a simplified view of the many components of the territory, and at the same time to provide important information about patterns or trends, becomes a tool of the utmost importance. These indicators can be understood as measurable characteristics of the environment, which facilitate comprehension of the processes occurring at different scales and serve as a reference to inform the population and support decision-making. The aim of the present note is to demonstrate briefly the need to develop geographical-environmental and natural risk indicators to facilitate comprehension of the dynamic of spatial and temporal landscape patterns, particularly in coastal environments. This approach offers an historical summary of the natural, socio-economic and political processes which currently make up the territory, and which without doubt will continue to influence it in the future. At the same time, it is proposed that information should be integrated on the basis of this framework with a view to generating spatial decision support systems in a context of planning and integrated management of the coastal zones of Chile.
