Triterpenes and triterpenoidal glycosides from the fruits of Ilex paraguariensis (Maté)

Abstract: a new triterpenoid glycoside, 3b-o-a-l-arabinopyranosyl rotundic acid, named here mateside (4), and the known triterpenes ursolic acid (1), acetylursolic acid (2), 23-hydroxyursolic acid (3), the saponins ziyu-glycoside i (5) and ilexoside ii (6), were isolated from fruits of ilex paraguariensis. their structures were established on the basis of 1d and 2d nmr spectroscopic methods. the threshold bitterness values for mateside and ilexoside ii were also determined and compared with saponins obtained from leaves of i. paraguariensis and other ilex species. the high bitterness of ilexoside ii, one of the main saponins found the fruits, but not in the leaves of i. paraguariensis, suggests that the utilization of the fruits in the maté production can produce a strong taste variation and unknown physiological effects.

An HPLC-DAD method to quantification of main phenolic compounds from leaves of Cecropia Species

Abstract: an efficient and reproducible hplc-dad method was developed and validated for the simultaneous quantification of major compounds (chlorogenic acid, isoorientin, orientin and isovitexin) present in the leaves of two cecropia species, c. glaziovii and c. pachystachya. from the leaves of c. glaziovii and c. pachystachya were isolated the c-glycosylflavones isoorientin and isovitexin and identified on both species chlorogenic acid (3-o-caffeoylquinic acid) and the o-glycosylflavonol isoquercitrin. the c-glycosylflavone orientin was isolated only from c. pachystachya. chlorogenic acid was the major compound in both species (11.1 mg g-1 of extract of c. glaziovii and 27.2 mg g-1 of extract of c. pachystachya) and for the flavonoids quantified, isovitexin was the main c-glycosylflavonoid for c. glaziovii (4.6 mg g-1 of extract) and isoorientin the main one for c. pachystachya (17.3 mg g-1 of extract).

Abstract: five glycosides were isolated from leaves of p. alata. the structures 1-5 were obtained through extensive spectral analyses as 3-o-b-d-glucopyranosyl-stigmasterol (1), 3-o-b-d-glucopyranosyl-oleanolic acid (2), 3-o-b-d-glucopyranosyl-(1?3)-b-d-glucopyranosyl-oleanolic acid (3), 3-o-b-d-glucopyranosyl-(1?2)-b-d-glucopyranosyl-oleanolic acid (4) and 9,19-cyclolanost-24z-en-3b,21,26-trihydroxy-3,26-di-o-gentiobiose (5). comparison of the tlc profiles of the hydroethanolic extracts from leaves of other passiflora species found in the south of brazil (p. actinia, p. caerulea, p. edulis var. flavicarpa, p. elegans, p. foetida, p. misera and p. tenuifila) showed that only p. alata presented saponin accumulation.
Saponins and Sapogenins from Brachiaria decumbens Stapf

Abstract: four steroidal saponins and three sapogenins were identified from aerial parts of brachiaria decumbens. their structures were established by chemical and spectroscopic means (1h and 13c nmr, hmbc, hmqc) as 3b-methoxy-lanost-9(11)-ene (1), diosgenin (2a), yamogenin (2b), 3-o-b-d-glucopyranosyl-24(s)-ethyl-22e -dihydrocholesterol (3a), 3-o-b-d-glucopyranosyl-24(r)-ethyl-22e -dihydrocholesterol (3b), dioscin (4a) and 3-o-{a-l-rhamnopyranosyl-(1?4)-[a-l-rhamnopyranosyl-(1 ?2)]-b-d-glucopyranosyl}-25(s)-spirost-5-en-3 b-ol (4b). all these compounds were isolated for the first time from b. decumbens and compound 4b is described for the first time as far as we know.
HPLC method to assay total saponins in Ilex paraguariensis aqueous extract

Abstract: ilex paraguariensis st. hilaire is a south american tree from which leaves and twigs are used to prepare a commonly consumed tea in several south american countries. the plant is known as "erva-mate" in portuguese or "yerba mate" in spanish. considering the potential use of the saponins as surfactant as well as its therapeutic potential, the present work was designed to propose extraction and quantitation methods for the saponins present in ilex paraguariensis. the saponins were extracted by decoction, hydrolyzed and quantified by a hplc method with uv detection. the saponins concentration was expressed in ursolic acid (total saponins). the method showed linearity for ursolic acid in the range of 13.5 to 135 μg ml-1. the aqueous extract presented total saponins concentration of 352 μg ml-1. the results also suggest the possibility of use of similar method for assaying triterpenoid saponins in other plants.

Abstract: ilex paraguariensis a. st. -hil. is a native species of southern south america. the caffeine content in ilex paraguariensis leaves has been considered an important quality parameter for maté-derived products. in this work different extraction methods for the determination of these methylxantines are compared. the influence of the extraction conditions on the methylxanthine yields was evaluated. extraction by decoction with acidic aqueous solution (h2so4 4n) presented the higher efficiency in the theobromine extraction. the extraction in a soxhlet with acidic aqueous solution and decoction with acidic aqueous solution showed the highest caffeine yield. for the concomitant theobromine and caffeine quantification, the decoction with acidic aqueous solution is suggested.

Abstract: objective: to investigate the adequacy of the content and format of the patient information section in package inserts of commonly prescribed drugs at the internal medicine service of a school hospital. methods: forty eight package inserts were collected from six pharmacies of the city of porto alegre in june 1998. the presence of mandatory notification and other information required by brazilian laws that regulate inserts content was examined. results: no one package inserts contained all the notification required by law. warnings about medication storage and expiration date were the most frequently information found in package inserts analyzed. conclusion: important information for the drug user was not presented in package inserts analyzed, limiting the purpose of inserts as an instrument for patient education.

Abstract: objective: to evaluate the knowledge of the generic designation, use and composition of analgesic medications containing aspirin. methods: a total of124 interviews were carried out between december 1999 and january 2000, in two neighborhoods of the city of porto alegre, southern brazil. the interview was held with the person who came to answer the door at each of the homes that was drawn. the data collection instruments comprised a set of five different pharmaceutical specialties containing acetylsalicylic acid , and an interview consisting of two open questions concerning the differences and similarities between the products. results: three major knowledge-level groups were characterized on the basis of the information that the interviewees were able to provide. the group that was knowledgeable about the matter comprised 14 individuals (11%). the group with limited knowledge contained 61 people (49)%. those who had no knowledge of the matter at all formed a group of 49 people (40%). conclusions: taking the results as a whole, they indicate that most people (about 90% of the sample investigated) are simply not aware of what the active substance is, even in pharmaceutical specialties that they use frequently.

Abstract: objective: to describe drugs used during pregnancy by women attending prenatal clinics of the national public health system (sus) in brazilian cities. methods: using a structured questionnaire, 5,564 pregnant women between the week 21 to 28 who attended prenatal visits of the sus in six brazilian cities were interviewed. the interview questions were grouped in "guided use" to cover pain, cramps, nausea, cough, and others, and "guided medicine" to cover vitamins, iron, and fluoride. the food and drug administration gestational risk classification (1991-1995) was applied. results: of a total of 5,564 women, 4,614 (83.8%) used at least one drug during pregnancy, with a total of 9,556 drugs used. the drugs most frequently used were vitamins associated with anti-anemics (33.5%), gastrointestinal drugs (31.3%), analgesics and anti-inflammatory drugs (22.2%), anti-anemics (19.8%), and antibiotics (11.1%). regarding gestational risk, 3,243 drugs used (34%) belonged to category a risk, 1,923 (22.6%) to category b, 3,798 (39.7%) to category c, 289 (3.0%) to category d, and 55 (0.6%) to category x. conclusions: a large variation in drug use across the cities was observed, especially for anti-anemics and vitamins associated with anti-anemics, revealing the lack of a national consensus regarding the use of these drugs during pregnancy. there was no literature data about safety during pregnancy for 12.9% of the drugs used. this percentage, plus the 26.9% of category c drugs, shows that 40% of the drugs used during pregnancy do not belong to the approved safety categories. however, only 3% of the 9,956 drugs used were clearly contraindicated during pregnancy.
