Abstract: Enzymatic activities are important to be quantified in products as enzymatic cleaners, which are used in medical and surgical devices reprocessing. Enzymatic activities are critical for the proper chemical cleaning that intends to remove solid organic dirt from inaccessible sites. The most important enzyme for this purpose is the protease, which is able to dissolve the main dirt attached to medical and surgical instruments. In this context, this study contributes to the development of a new proteolytic activity quantification method and its validation. The methodology is based on colorimetry and uses a UV-Vis spectrophotometer to measure the substrate hydrolysis by the blue color intensity, employing Protazyme AK tablets as substrate.

Abstract: glyoxal, which can be obtained from biomass (as by lipids oxidation), is non-toxic and non-volatile. it was used as a substitute of formaldehyde, which does not have these properties, in the synthesis of a novolac-type phenolic resin, using oxalic acid as a catalyst, which can also be obtained from renewable sources. the glyoxal-phenol resin was used in the preparation of composites reinforced with microcrystalline cellulose (mcc 30, 50, and 70% w/w). scanning electron microscopy (sem) images of the fractured surfaces showed that the composites presented a good reinforcement/matrix interface. this can be attributed to the high surface area of the mcc and also to the presence of polar groups (hydroxyl) in both cellulose and matrix, which allowed the formation of hydrogen bonds, leading to a good adhesion between the components present at the interface. dynamic mechanical thermoanalysis (dmta) showed that all of the obtained composites have high storage modulus at room temperature. moreover, the composite reinforced with 30% of mcc showed the lowest water absorption, almost the same as that of the phenolic thermoset, which is used in industrial applications. the results showed that composites with good properties can be prepared using high proportions of materials obtained from biomass.

Abstract: based on the multidimensional model proposed by marsh and shavelson (1985), this study tries to verify in what manner different level of sport participation (people who never play sport; abandoned sport and still plays sport) are associated to different self-concept structures. simultaneously, the study pretends to access how sport participation is related with different levels in the various domains of self-concept. the portuguese version of sdq iii (faria & fontaine, 1992) was administrated to 218 subjects (male and female), with ages between 17 and 35 years, non-praticants (n=69), past-praticants (n=72) and praticants (n=77) of different kind of sport. the differences between these groups in differents domains of self-concept were analyse, well as the differences between gender and age were explored. the results revealed that global self-concept was higher in sport participants and that men and women differ significantly in terms of physical appearance and competence.

Abstract: The present work is a study about degradation occurring with red ceramics whenever they are exposed to the environmental conditions of a cold and humid climate. Four different types of clay from Rio Grande do Sul were studied: two of them from the Serra Geral formation, and the other two from the formation of Strada Nova, which are commonly used to make roof tiles. The clay was characterised to identify its chemical and mineralogical composition. Also the physical properties were researched. In the ceramic processing, the pressure with four different sintering temperatures (850oC, 900oC, 950oC, and 1000oC) at three different intervals (3, 12, and 24 hours) were used. The ceramic materials thus obtained were characterised according to their physical properties (water absorption, total density, porosity, and porous size), and mechanical (mechanical resistance over four points). For the characterisation of the degradation mechanism, some trials were carried out to obtain the coefficient of saturation and on frost and defrost cycles. The microstructural analysis was made with a scanning electronic microscopy. On the basis of the results obtained, a model for the degradation mechanism is presented, and a degradation factor which relates the total volume of mesopores to the standard size of mesopores is suggested. It was concluded that the red ceramic products more prone to be frost-resistant are those presenting a degradation factor below 100. Se estudia la degradación que sufre la cerámica roja cuando se expone a la acción de los climas fríos e húmedos. La experimentación se ha realizado con cuatro arcillas de la son expuestas a los climas fríos y húmedos. Se investigaron cuatro diferentes arcillas de la provincia del Rio Grande do Sul, que se emplean tradicionalmente en la fabricación de tejas. Dos de ellas pertenecen a la formación de la Serra Geral y las otras dos de la formación Estrada Nova. La caracterización de las arcillas ha incuido: composición química y mineralógica, granulometría, plasticidad, humedad de moldeo, contracción lineal de secado e resistencia a la flexión de piezas secas. Las probetas han sido moldeadas por prensado, y cocidas a 850o, 900o, 950o y 1000oC con tres tiempos de permanencia a la temperatura máxima (3, 12 y 24 horas). En las probetas cocidas se ha determinada la capacidad de absorción de agua, la densidad total, la porosidad abierta, la distribución del tama o de los poros y la resistencia a la flexión en cuatro puntos. Procura-se compreender o processo de implanta o, abertura, funcionamento e fechamento dessa escola étnica italiana, processo este que ocorreu n o em detrimento à escola pública, mas paralelo a ela. Busca-se identificar os sujeitos que fizeram parte daquele processo de escolariza o étnico e reconhecer os impactos do nacionalismo na busca da desconstru o das fun es daquela escola, bem como as estratégias utilizadas pelo grupo para a manuten o da identidade étnica. Exploring MIA-QSARs for farnesyltransferase inhibitory effect of antimalarial compounds refined by docking simulations  [PDF]

Abstract: Two series of farnesyltransferase (FTase) inhibitors were grouped and their antimalarial activi-ties modeled by means of multivariate image analysis applied to quantitative structure-activity relationship (MIA-QSAR). A reliable model was achieved, with r2 for calibration, external prediction and leave-one-out cross-validation of 0.96, 0.87 and 0.83, respectively. Therefore, biological activities of congeners can be estimated using the QSAR model. The bioactivities of new compounds based on the miscellany of substructures of the two classes of FTase inhibitors were predicted using the MIA-QSAR model and the most promising ones were submitted to ADME (absorption, distribution, metabolism and excretion) and docking evaluation. Despite the smaller interaction energy of the two most promising, predicted compounds in comparison to the two most active compounds of the data set, one of the proposed structures did not violate any Lipinski’s rule of five. Therefore, it is either a potential drug or may drive synthesis of similar, improved compounds.
The Expression of Soluble and Active Recombinant Haemophilus influenzae IgA1 Protease in E. coli

Abstract: Immunoglobulin A1 (IgA1) proteases from Haemophilus influenzae are extracellular proteases that specifically cleave the hinge region of human IgA1, the predominant class of immunoglobulin present on mucosal membranes. The IgA1 proteases may have the potential to cleave IgA1 complexes in the kidney and be a therapeutic agent for IgA1 nephropathy (IgAN), a disease characterized by deposition of the IgA1 antibody in the glomerulus. We have screened for the expression of recombinant H. influenzae IgA1 protease by combining various expression plasmids, IgA1 protease constructs, and E. coli strains under multiple conditions. Using the method we have developed, approximately 20–40 mg/L of soluble and active H. influenzae IgA1 protease can be produced from E. coli strain C41(DE3), a significant increase in yield compared to the yield upon expression in H. influenzae or other related bacteria.
Expression of FoxA and GATA transcription factors correlates with regionalized gut development in two lophotrochozoan marine worms: Chaetopterus (Annelida) and Themiste lageniformis (Sipuncula)

Abstract: FoxA and GATA456 transcripts are predominantly restricted to gut tissue, and together show regional expression spanning most of the alimentary canal in each of these lophotrochozoans, although neither FoxA nor GATA456 is expressed in the posterior intestine of Chaetopterus. In both species, FoxA is expressed at the blastula stage, transiently in presumptive endoderm before formation of a definitive gut tube, and throughout early larval development in discrete foregut and hindgut domains. GATA456 genes are expressed during endoderm formation, and in endoderm and mesoderm associated with the midgut in each species. Several species-specific differences were detected, including an overlap of FoxA and GATA456 expression in the intestinal system of Themiste, which is instead complimentary in Chaetopterus. Other differences include additional discrete expression domains of FoxA in ectodermal trunk cells in Themiste but not Chaetopterus, and expression of GATA456 in anterior ectoderm and midgut cells unique to Chaetopterus.This study of gene expression in a sipunculan contributes new comparative developmental insights from lophotrochozoans, and shows that FoxA and GATA456 transcription factors are part of an ancient patterning mechanism that was deployed during early evolution of the metazoan through gut. The common utilization of FoxA and GATA456 throughout gut formation by species with contrasting life history modes indicates that both genes are core components of a gut-specific gene regulatory network in spiralians. Despite a highly conserved pattern of early development, and probably similar ontogenic origins of gut tissue, there are molecular differences in gut regionalization between lophotrochozoan species.Formation of a through gut was a key innovation during the radiation of the Metazoa.
