Canonical Wnt Signaling Promotes Early Hematopoietic Progenitor Formation and Erythroid Specification during Embryonic Stem Cell Differentiation

Abstract: The generation of hematopoietic stem cells (HSCs) during development is a complex process linked to morphogenic signals. Understanding this process is important for regenerative medicine applications that require in vitro production of HSC. In this study we investigated the effects of canonical Wnt/β-catenin signaling during early embryonic differentiation and hematopoietic specification using an embryonic stem cell system. Our data clearly demonstrates that following early differentiation induction, canonical Wnt signaling induces a strong mesodermal program whilst maintaining a degree of stemness potential. This involved a complex interplay between β-catenin/TCF/LEF/Brachyury/Nanog. β-catenin mediated up-regulation of TCF/LEF resulted in enhanced brachyury levels, which in-turn lead to Nanog up-regulation. During differentiation, active canonical Wnt signaling also up-regulated key transcription factors and cell specific markers essential for hematopoietic specification, in particular genes involved in establishing primitive erythropoiesis. This led to a significant increase in primitive erythroid colony formation. β-catenin signaling also augmented early hematopoietic and multipotent progenitor (MPP) formation. Following culture in a MPP specific cytokine cocktail, activation of β-catenin suppressed differentiation of the early hematopoietic progenitor population, with cells displaying a higher replating capacity and a propensity to form megakaryocytic erythroid progenitors. This bias towards erythroid lineage commitment was also observed when hematopoietic progenitors were directed to undergo myeloid colony formation. Overall this study underscores the importance of canonical Wnt/β-catenin signaling in mesodermal specification, primitive erythropoiesis and early hematopietic progenitor formation during hematopoietic induction.
A time to look within curricula—Nursing students’ perception on sexuality and gender issues  [PDF]

Abstract: Nursing students could be viewed as dynamic change agents given the knowledge and skills they would be equipped with. Amongst all areas to be addressed in their clientele of the hospital or community setting, one key area that a nurse could focus on is sexuality. This however is often neglected for simple reasons such as lack of knowledge, embarrassment, fear of intrusion on privacy, or it is perceived as an un-important concern of the patient. Focus groups discussions combined with self report were conducted on 84 nursing students selected by the faculty from three institutions of nursing to assess their perceptions related to sexuality and gender issues, and thus extrapolate on their learning needs within the curriculum. Student expressed discomfort in caring for patients who had sexual expressions and relationships different from the accepted cultural norm; perceived sexuality primarily as heterosexual relationship; helplessness when faced with sexual harassment in the health care field and gaps in their present curricula in relation to contextualization of sexuality and gender issues.

Abstract: Military communities differ in how they implement ethics training.Some have well developed programmes while others have only a few stand-alone presentations. For this reason it is difficult to produce a template to suit every situation.Leadership training within the Military is generally of a very high standard and it is particularly important in relation to the development of high moral standards, whereas the training in ethics various considerably throughout the military world.Therefore, even though we regard our ethics programme to be of a very high standard there is always the need to review what we have in place and update it. Codes of Ethics need to be revised from time to time and we can gain valuable insight from sharing and comparing our training programmes with other militaries.

Abstract: This paper examines the semiosic development of energy to information within a dyadic reality that operates within the contradictions of both classical and quantum physics. These two realities are examined within the three Peircean modal categories of Firstness, Secondness and Thirdness. The paper concludes that our world cannot operate within either of the two physical realities but instead filiates the two to permit a semiosis or information-generation of complex systems.

Abstract: The success of a profession will be determined upon its education and training. A profession is required to encompass: a core body of knowledge; a set of ethical codes of practice; and have practitioners with humanistic qualities. In order to maintain the success of a profession it is necessary to have continuing education, which enhances professional development. Continuing professional education includes a form of self-regulation, which ensures the maintenance of a minimum standard of practice in this ever-changing workplace, and by regulating this standard, the discipline becomes more accountable to the client and the profession as a whole. In Australia, the Nuclear Medicine society's continuing education programs and Universities offering postgraduate programs promote continuing education. If technologists are to successfully keep up with developments in instrumentation, protocols and changing health care requirements, we must ensure that the education of practitioners does not cease at certification of entry to the workplace.
Reshaping Spectatorship: Immersive and Distributed Aesthetics

Abstract: Although discourses of immersive aesthetics and distributed aesthetics may evoke associations with different media, creative processes, modes of audience engagement and even political ideologies, artists using these aesthetics often share similar interests in transforming and enhancing notions of the body and perception through technological intervention. This paper undertakes a comparison between immersive and distributed aesthetics in relation to Virtual Reality (VR) and Networked Art (net.art), particularly networked installation art. It focuses on the ways in which both VR and networked installations immerse the viewer in states of perceptual and cognitive transition. Central to this article is the argument that VR and net.art are able to generate immersive experiences in the viewer by creating the sensation of being (tele-)present in an electronically mediated environment that is illusionistic and sometimes remote from the physical body of the participant. Furthermore, the immersive and distributed aesthetics generated by specific VR and net.art projects have revolutionary consequences for traditional aesthetic theories of spectatorship and art appreciation that assert the need for critical and physical distance.

Abstract: Globalization is an irreversible development and the main purpose of this paper is to rethink how Mainland China responds to the challenges of globalization in rural area. To achieve this aim, this study will be divided into three parts. First of all, it will examine the phenomenon of privatization and dumping in rural China as a result of globalization. It will then try to consider the feasible top-down administrative changes of Chinese government. The last part will explore the new modalities of political engineering and social transformation of the state-society relation resulted from the bottom-up resistance to globalization.
Optimally splitting cases for training and testing high dimensional classifiers

Abstract: We develop a non-parametric algorithm for determining an optimal splitting proportion that can be applied with a specific dataset and classifier algorithm. We also perform a broad simulation study for the purpose of better understanding the factors that determine the best split proportions and to evaluate commonly used splitting strategies (1/2 training or 2/3 training) under a wide variety of conditions. These methods are based on a decomposition of the MSE into three intuitive component parts.By applying these approaches to a number of synthetic and real microarray datasets we show that for linear classifiers the optimal proportion depends on the overall number of samples available and the degree of differential expression between the classes. The optimal proportion was found to depend on the full dataset size (n) and classification accuracy - with higher accuracy and smaller n resulting in more assigned to the training set. The commonly used strategy of allocating 2/3rd of cases for training was close to optimal for reasonable sized datasets (n ≥ 100) with strong signals (i.e. 85% or greater full dataset accuracy). In general, we recommend use of our nonparametric resampling approach for determing the optimal split. This approach can be applied to any dataset, using any predictor development method, to determine the best split.The split sample approach is a widely used study design in high dimensional settings. This design divides the collection into a training set and a test set as a means of estimating classification accuracy. A classifier is developed on the training set and applied to each sample in the test set. In practice, statistical prediction models have often been developed without separating the data used for model development from the data used for estimation of prediction accuracy [1]. The Importance of the PI3K/AKT/MTOR Pathway in the Progression of Ovarian Cancer

Abstract: Ovarian cancer is the fifth most common cause of death due to cancer in women despite being the tenth in incidence. Unfortunately, the five-year survival rate is only 45%, which has not improved much in the past 30 years. Even though the majority of women have successful initial therapy, the low rate of survival is due to the eventual recurrence and succumbing to their disease. With the recent release of the Cancer Genome Atlas for ovarian cancer, it was shown that the PI3K/AKT/mTOR pathway was one of the most frequently mutated or altered pathways in patients’ tumors. Researching how the PI3K/AKT/mTOR pathway affects the progression and tumorigensis of ovarian cancer will hopefully lead to new therapies that will increase survival for women. This review focuses on recent research on the PI3K/AKT/mTOR pathway and its role in the progression and tumorigensis of ovarian cancer.
