Folículo Dominante e Resposta Superovulatória em Novilhas da Ra a Nelore

Abstract: Dezessete novilhas da ra a Nelore foram superovuladas a partir do nono dia do ciclo estral, sendo oito novilhas com 250 UI (T1) e nove com 500 UI (T2) de FSH, em oito doses decescentes. Os animais foram monitorados por meio de ultra-sonografia durante o período superovulatório e no dia da coleta de embri es. N o houve diferen a na taxa de recupera o e qualidade das estruturas coletadas entre os animais dos dois tratamentos. Foram coletados 4,16 embri es viáveis/doadora nos animais do T1 e 3,15 embri es viáveis/doadora nos animais do T2. A presen a do folículo dominante no início do tratamento superovulatório n o interferiu na resposta superovulatória. O diametro do maior e do segundo maior folículo e número de folículos subordinados entre os animais dos tratamentos durante a superovula o n o diferiram. O diametro do maior folículo e do segundo maior e o número de folículos subordinados foram de 11,36 mm, 8,91 mm e 16,93 e de 11,59 mm, 9,42 mm e 17,44 para os tratamentos com 250 e 500 UI de FSH, respectivamente.

Abstract: Objetivou-se avaliar os efeitos da vitrifica o em ovócitos de bovinos após o cultivo in vitro, utilizando o etilenoglicol como crioprotetor. Ovócitos obtidos de ovários de vacas abatidas em matadouro foram distribuídos aleatoriamente em três tratamentos. Tratamento 0 (testemunha): ovócitos n o-desnudados e n o-congelados. Tratamento 1: vitrifica o de ovócitos imaturos n o desnudados, desidratados previamente por cinco minutos em três solu es contendo 20, 20 e 40% de etilenoglicol, acrescidas de 0,3 mol L-1 de trehalose e 20% de PVP, em meio de Talp Hepes. Tratamento 2: vitrifica o de ovócitos imaturos desnudados, conforme o Tratamento 1. Após o descongelamento (imers o em banho-maria a 30oC por 20 segundos), os ovócitos foram reidratados gradativamente, mantendo-os por 6 minutos em cada uma das solu es a seguir, sucessivamente: meio Talp Hepes com 20% de etilenoglicol + 0,3 mol L-1 de trehalose + 10% de PVP e meio Talp Hepes sem etilenoglicol, trehalose e PVP, onde foram lavados três vezes. Posteriormente, os ovócitos foram cultivados a 38,5oC, com 95% de umidade e atmosfera de 5% de CO2 por 24 horas. Após o cultivo, os ovócitos foram fecundados e os embri es cultivados in vitro por sete dias. Foi encontrada uma taxa de matura o nuclear de 81 (68/84), 19 (7/36) e 0% (0/31), nos Tratamentos 0, 1 e 2, respectivamente. As taxas de clivagem e de desenvolvimento embrionário foram de 56,4 (102/181) e 54,9% (56/102), 1,7 (1/60) e 0,0% (1/60), 0,0 (0/71) e 0,0% (0/71), nos Tratamentos 0, 1 e 2, respectivamente. Esses resultados indicam que o procedimento de vitrifica o, segundo os protocolos utilizados, n o é indicado para a criopreserva o de ovócitos de bovinos.

Abstract: seventeen nelore heifers were submitted to a superovulation protocol, beginning on the ninth day of the estrous cycle. eight heifers were superovulated with 250 iu (t1) and nine with 500 iu (t2) of fsh. the animals were monitored by ultrasonography during the superovulation period and on the embryo collection day. there were no differences on the recovery rate and quality of the structures collected among the animals of the two treatments. animals treated with 250 (t1) and 500 iu (t2) of fsh produced 4.16 and 3.15 embryos viables/donor, respectively. the presence of the dominant follicle in the beginning of the superovulatory treatment did not affect the superovulatory response. the largest diameter follicle and second largest one and number of subordinate follicles during the superovulation from the animals of the treatments did not differ among treatments. the diameter for the largest follicle, the second largest and the number of subordinate follicles for the animals treated with 250 versus 500 iu, were: 11.36 x 11.59 mm; 8.91 x 9.42 mm and 16.93 x 17.44 mm, respectively.

Abstract: the objective of this work was to evaluate the effects of increasing levels of urea in the diets in postpartum crossbred dairy cows on plasma level of n-urea (nup) and uterine ph in the estrus and luteal phase. sixteen crossbred cows (holstein-zebu) with body condition score at parturition between 3.5 and 4.0 (1 to 5 scale) were assigned in four treatments with diets made up with corn silage and concentrate in the proportion of 60:40 as: t1=0.0; t2=0.7; t3=1.4; and t4=2.1% of urea in the dry matter. the uterine ph was measured four hours after the diet intake, during the estrus and luteal phase from the second or third estrous cycle postpartum period in dairy cows. simultaneously, blood samples were collected for determination of the level of plasma urea. the dry matter intake from 70 to 110 days of lactation reduced from 0.7% of urea. the levels of plasma n-urea were not affected in the samples collected during the estrus (19.59 mg/dl). the uterine ph during estrus (6.66) and luteal phase (6.97) were not affected by the different levels of urea.

Abstract: the objective was to evaluate the effects of vitrification of immature bovine oocytes after in vitro culture, by the use of cryoprotectors ethylene glycol. oocytes from cows ovaries from slaughters houses were randomly alocated into three treatments. treatment 0 (control): frozen-thawed undesnude oocytes; treatment 1, immature vitrificated undesnude oocytes dehydrated for 5 minutes in each of the following solutions of 20, 20 and 40% of ethylene glycol, respectively, associated to 0.3 mol l-1 of trehalose and 20% of pvp, in media talp hepes, and, treatment 2, the same as treatment 1, but desnudes oocytes. after frozen-thawed of the oocytes (imersion in water bath at 30oc for 20 seconds), the oocytes were gradually rehydrated, in the following sequence of solutions: media talp hepes with 20% of ethylene glycol + 0.3 mol l-1 of trehalose + 10% of pvp and media talp hepes without ethylene glycol, trehalose and pvp, were washed three times. ultimately, the oocytes were cultured at 38.5oc, with 95% umidity and atmosphere of 5% of co2 for 24 hours. after culture, the oocytes were fertilized and the embryos cultured in vitro for seven days. the nuclear maturation were 81 (68/84), 19 (7/36) and 0% (0/31), for treatments 0, 1 and 2, respectively. the cleavage and development rates were: 56.4(102/181) and 54,9% (56/102), 1,7. (1/60) and 0,0% (1/60), 0,0 (0/71) and 0,0% (0/71), for the treatments 1, 2 e 3, respectively. these results show that the vitrification procedures, by the used protocols, are not indicated for bovine oocytes cryopreservation.
Uso de dimetil-formamida associada ou n?o ao glicerol na criopreserva??o de sêmen caprino

Abstract: the objective of this trial was to evaluate the efficiency of dimethylformamide on cryopreservation of goat semen using semen physical analyses and complementary tests. alpine and saanen bucks from the dairy goat experimental station- dzo were used. semen samples were frozen with a yolk-skimmed milk diluent and cryoprotectors with different concentrations as follows: 7% glycerol (t1), 3.5% glycerol plus 3.5% dimethylformamide (t2) or 5% dimethylformamide (t3). semen was centrifuged, diluted, and bottled in 0.5 ml straws followed by cooling for 40 minutes until reach 5.0°c and kept at this temperature for additional 80 minutes. samples were then exposed to liquid nitrogen vapor for 15 minutes and finally frozen. thawing was done in a waterbath at 37°c for 50 seconds. the following variables were evaluated in vitro: sperm progressive motility and vigor, acrosomal integrity and reaction, and plasmatic membrane integrity. the cryopreservation procedures reduced 30.0% of the initial motility and 19% of plasmatic membrane integrity that was evaluated using the hyposmotic swelling test (host). it was observed an increaseof 3% in acrosomal lesions when samples were submitted to a slow thermoresistance test (trt). no significant differences in sperm motility were observed among treatments at 0, 5, and 60 minutes of trt as well as in sperm vigor at all trt times. however, at 120 minutes of trt a significant difference in sperm motility was found comparing t1 and t2 while t3 was intermediate. there were no significant differences on plasmatic membrane integrity, acrosomal integrity, and post-thawing acrossomal reaction across treatments. it can be concluded that dimethylformamide can be a viable alternative for goat semen cryopreservation used alone or in association with glycerol.

Abstract: the objective of this study was to evaluate the relationship between complementary (hiposmotic, thermo-resistance and acrosome reaction tests) and conventional evaluations (physical and morphologic aspects) of bovine frozen/thawed semen and conception rates. average values for spermatic motility evaluated by thermo-resistance test were of 53.48% (post-thawed), 43.69% (60 minutes), 35.88% (120 minutes) and 33.04% (180 minutes). the percentage of reactive cells observed for the hiposmotic test was 37.89%. average intensity was observed for post-thawing spermatic motility, positively correlated to hiposmotic test (0.21). however, correlation between motility in the 180 minutes and hiposmotic test was high (0.64). the percentage of cells presenting post-thawing acrosome reaction was 9.85%, which was negative correlated to post-thawing (-0.25) and after three hours of incubation (-0.46) spermatic motility. no correlation was observed for complementary tests and post-thawed motility and the conception rate. no one of the parameters evaluated in this study was individually able to indicate the fertilizing capacity of the frozen/thawed semen.

Abstract: this study aimed at the evaluation of the effects from cryopreservation of bovine oocytes in vitro matured, by using ethylene glycol (eg) associated to trehalose and polyvinylpyrrolidone (pvp), of ovary oocytes of slaughtered cows, randomly assigned to three treatments. treatment 0 (t0 - control): oocytes that were desnuded and not vitrified. treatment 1 (t1): cryopreservation of in vitro matured oocytes with cumulus oophorus. tratamento 2 (t2): cryopreservation of in vitro matured desnuded oocytes. the percentage of recovered oocytes after cryopreservation and with normal morphology was different for vitrified oocytes (94.7 and 76.8%; 69.5 and 48.85% for t1 and t2, respectively). the main changes ultrastructural in vitrificated oocytes were prematurely released of cortical granules. later, all normal oocytes were fecundated and cultivated at 38.5oc in atmosphere with 5% co2 for seven days. the fecundation and cleavage rates for treatments were different (56.2, 41.7 and 12.5%; 36.3, 0.0 and 0.0%, for t0, t1 and t2, respectively). morulas and blastocysts were obtained only in t0 (34.5%). these results indicate that, the used protocols, for vitrification procedure is not indicated for cryopreservation of matured bovine oocytes.

Abstract: this study aimed to evaluate reproductive characteristics (sexual behavior, scrotal perimeter and quali-quantitative aspects of semen) of young and adults alpine and saanen male goats exposed to an artificial photoperiod (16 hours of light and 8 hours of darkness for 2 months). the young animals showed greater libido than the adults, based on the smallest time of reaction. frequency of manifestation of the behavior traits varied between breeds and ages and higher intensity was observed on adult animals. scrotal perimeter was smaller for alpine goats than for saanen goats at the same age. the scrotal perimeter increased from 30.53 cm in the first period (15 days after the end of the photoperiod exposure) to 31.53 cm in the second period (to the end of the experiment). quality of semen of alpine goats was better than that of saanen goats at the same age. according to the reproductive traits evaluated, alpine and saanen goats exposed to similar conditions of this study are sensible to seasonal variation during the year.

Abstract: the freezing ability of semen from alpine and saanen goats submitted to an artificial photoperiod exposure were evaluated by the thermo-resistance (ttr) and hypoosmotic (host) tests and integrity of the acrosome using eight male goats, four of each breed and two age groups (young and adults). the quality of semen during the freezing process was superior for the young animals of both breeds. spermatic motility immediately after thawing showed a positive effect on ttr; animals with higher motility after collection (86.2% vs 79.3%) and after thawing (37.7% vs. 32.0%) had a higher seminal longevity. both fresh and frozen semen did not differ between breeds and ages by the host test. there was a 14% decrease (p<0.05) in host values for frozen semen compared the fresh semen (38.0 vs. 52.0%, respectively). the spermatic progressive mobility was more sensible to cryopreservation than the integrity of the spermatic membrane, indicating that spermatic mobility is more affected by the frozen process than the plasmatic membrane. the level of acrosome damage (edema, partial and total detachment) was acceptable (44.5%) after thawing and after finishing the ttr.
