Core-Germplasm Construction of Rosa rugosa Collections by CDDP Molecular Markers  [PDF]

Abstract: A method for constructing core-germplasm of Rosa rugosa in China based on molecular marker data was probed and the optimum core germplasm was established. Studies were initiated to analyze the genetic diversity of 120 rugged roses from 6 different Chinese source populations based on CDDP marker, and a preliminary construction of the core collection was established using stepwise UPGMA clustering sampling method. The 26 core collection resources of R. rugosa collections in China have 20% germplasm samples of initial collection, the retention ratio of polymorphic loci, effective number of alleles (Ne), Nei’s genetic diversity (H) and Shannon information index (I) were respectively 97.52%, 104.16%, 108.38% and 106.18%. The results of t-test showed that no significant difference was found in genetic diversity indexes between the core collection and the original collection. These results also demonstrated that the core collection could stand for original collection excellently. The results show that CDDP molecular technology can be successfully applied to the construction of core germplasm resources of rugged roses.
Effects of Different Treatments on Physiological Characteristics of Cephalotaxus sinensis Seeds  [PDF]

Abstract: Cephalotaxus sinensis seeds can’t germinate even in the appropriate environment. However, numerous studies have showed that cold stratification and gibberellin acid (GA) can break the seed dormancy and promote seed germination effectively. To investigate the effect of cold stratification and different concentrations of exogenous GA3 on dormancy breaking in seeds of Cephalotaxus sinensis, we monitored germination rates and changes in soluble sugar, starch, amylase, soluble protein, free amino acid during cold stratification. The results showed that seeds stratified for 5 months germinated to 12.7%, while those disposed with 200, 400, 600 mg/L of GA3 germinated to 29.2%, 21.7%, and 18.4%, respectively. Free amino acid content was enhanced significantly, whereas soluble sugar content decreased during 45 days and then increased constantly. Additionally, the main reserves such as starch, protein decreased significantly during cold stratification, and cold stratification induced increases in the activities of α-amylase, (α + β)-amylase. The preliminary results show that the combination of GA3 and cold stratification has better effect to break seed dormancy.
The Causes of Dormancy and the Changes of Endogenous Hormone Content in Cephalotaxus sinensis Seeds  [PDF]

In many wild species, seeds are dormancy at maturity and will not germinate even under favorable environment conditions. Dormancy is a complex trail that is determined by many factors. Some studies have shown that cold stratification and the application of gibberellic acid (GA) can break seed dormancy and promote seed germination. The present study investigated the causes of plant dormancy and the effect of cold stratification and different concentrations of exogenous GA3 in regulating Cephalotaxus sinensis seed germination. Results showed that C. Sinensis seeds have good water permeability, which suggested that seed coats were not the main cause that inhibited the seed germination. There were germination inhibitions in all parts of seeds, and the order of inhibitory effect was: testa < endosperm (embryo), which indicated that existence of germination inhibitions was the main reason causing seed dormancy. Endogenous GAs and IAA (indole-3-acetic acid) content increased, while ABA (abscisic acid) content decreased over the experiments. ZR (zeatin riboside) content decreased in the early phase of cold stratification, but rebounded by the end of the experimental period. The changes of endogenous hormone indicated that GA, IAA and ZR played a positive role in seed germination, whereas ABA was associated with seed dormancy. Besides, the relative ratio of GA/ABA, IAA/ABA and ZR/ABA may play a more important role than their absolute level during the seed development.
Genetic Diversity Analysis of Cotoneaster schantungensis Klotz. Using SRAP Marker  [PDF]

Abstract: In order to detect the genetic relationship of different Cotoneaster schantungensis natural populations, 16 samples were selected for genetic diversity analysis based on SRAP markers. Twelve pairs primers were selected out, 93 bands were amplified, of which 91 bands (97.85%) were polymorphic. At species level, the average value of Nei’s genetic diversity (H) was 0.2213, Shannon’s information index (I) was 0.3596, whereas at population level, they were 0.1955, 0.3121, the percentage of polymorphic loci was 75.27%. The genetic differentiation coefficient in populations (Gst) was 0.0788, gene flow Nm was 5.8467, indicating that there was 7.88% of the variation among populations and 92.12% of the variance within populations and the variation within populations was relatively high. These results indicate that C. schantungensis has high genetic diversity and low differentiation among populations. Although most Cotoneaster species are apomictic, C. schantungensis, however, may be an exception.

Abstract: In the flavonoid biosynthesis pathway, Chalcone synthase (CHS) is involved in the formation of the pigment and has been shown to be a rate-limiting enzyme for the synthesis of flavonoids. In this study, a PCR approach was used to clone a Chalcone synthases cDNA from flower of sweet osmanthus “Chenghong Dangui” and it was designated as OfCHS (O. fragrans, CHS). The cDNA was 1383 bp long and a coding sequence (CDS) of 1173 bp encoding a polypeptide of 391 amino acids with an estimated molecular mass of 39.9 kDa. The theoretical isoelectric point was 6.23. Phylogenetic analysis demonstrated that OfCHS clustered with Olea europaea, Solenostemon scutellarioides, Perilla frutescens, Antirrhinum majus and Digitalis lanata. We also detected the expression of OfCHS in different tissues in “Dangui” and in two cultivars with varied coloration, “Zi Yingui” and “Chenghong Dangui” at different floral stages using quantitative real-time PCR. We observed that OfCHS transcript was higher in leaves than in petals in “Dangui”. The transcripts of OfCHS in “Zi Yingui” petals were higher than those in “Dangui” at three stages especially at xianyan stage and there was no significant difference between the two cultivars in the full flowering stage. “Chenghong Dangui” has a relatively high anthocyanin content compared to “Zi Yingui”. The relative amount of anthocyanin of “Chenghong Dangui” initially increases, and then decreases during the bloom period. However, the expression of CHS is the highest at the initial flowering stage. These data suggest that the OfCHS does not play a key role in the accumulation of total flavonoid in this cultivar. These data could contribute to explain the different accumulation of flavonoids in petals of the two cultivars.

Abstract: In order to find a method to break dormancy of Rosa rugose seeds and interspecific hybrid seeds between Rosa rugose and Rosa hybrid quickly, and accelerate the breeding process of interspecific hybridization between Rosa rugose and Rosa hybrid, the influence of concentrated acid, seed maturity, GA3 (gibberellin) and low temperature (4°C) on seed germination of Rosa rugose from Muping was researched under aseptic condition. The results showed that aseptic germination can significantly shorten the germination time of Rosa rugose seeds and raise its germination ratio. Abstract: In order to reveal the phenomenon of R. rugose pollination incompatibility, the full-length cDNA sequence of S Locus F-box Gene was cloned for the first time from the pollen of Rosa rugose “Zilong wochi” with RT-PCR and RACE methods and named as RrSLF. The full-length cDNA is 1236 bp with an open reading frame of 1122 bp, encoding 343 amino acids. The derived protein has a molecular weight of 43.7 kD, a calculated pI of 6.24, an F-box conserved domain at position 343 - 741, and belongs to F-box family. The derived protein is a Hydrophobicity protein secreted into the cytoplasm. There is no transmembrane domain and no signal peptide cleavage site, twenty-one Ser phosphorylation sites, seven Thr phosphorylation sites, seven Tyr phosphorylation sites, two N-glycosylation sites, and no O-glycosylation sites. Abstract: This paper deals with the floristic components,geographical distribution and ecological environments of the Pteridophyte in Shandong. The floristic relationships between Shandong and some other regions are also disscussed. The results show that: 1. The floristic elements are relatively not rich, however, the geographical elements are varied. All the 40 genera can be classified into 10 areal-types, mainly composed of the North Temperate(30.0%), the Pantropical(22.55%), and the Cosmopolitan(20.0%).The areal-types of temperate account for 42.5%, taking a predominant position, while those of tropic account for 37.5%. 2. Endemism is remarkable, 12 species of Pteridophyte, which make up 11.3% of the total numbers, are endemic to this province. 3. The geographical distribution is fairly uneven, The number of species are decreased from South to North and from East to West. 4. The similar quotents of Pteridophyte flora between Shandong and Liaoning, Beijing, Henan, Qinling, Japan, Hainan, Xizang are 46.4%, 45.3% , 41.2%, 26.0%, 24.8%,4.7%, 3.8% respectively in sequence order. This means that this flora is most similar to Liaoning, and then to Beijing and Henan.

Abstract: Taishan mountain is located in central Shandong Province.There are 757 species seed plants belonging to 407 genera and 100 families (3 sp.of 3 gen.among 2 fam.of gymnosperms and 754 sp.of 404 gen.among 98 fam.of angiosperms) in this flora.In the 407 genera, there are 11 world monotypic genera and 40 world oligotypic ones, which make up 12.5% of the total.Most of them are phylogenetically ancient or primitive.This means the seed plants flora of Taishan mountain is characteristically of ancient origin.According to Prof.C.Y.Wu's theory on the areal-types of the genera of China's seed plants flora, all the 407 genera in this flora can be classified to 15 types, mainly comosing of North Temperate(34.5%),Pantropic(19.1%) ,Old World Temperate(14.3%)and East Asia(10.1%).The genera of temperate areal-types make up 72.2%,taking a predominated position, while those of tropical ones only acount for 27.8%,which shows that the seed plants flora of Taishan mountain is of a temperate feature,and has something with tropical flora.
Molecular phylogeny and biogeography of the highly specialized grade schizothoracine fishes (Teleostei: Cyprinidae) inferred from cytochrome b sequences

Abstract: We recovered the phylogenetic relationships among 23 species and subspecies of the highly specialized grade schizothoracine fishes distributing at 36 geographical sites in the Tibetan Plateau and its surrounding regions by analyzing sequences of cytochrome b genes. Furthermore, we estimated the possible divergent times among lineages based on a historical geological isolation event in the Tibetan Plateau. The molecular data revealed that the highly specialized grade schizothoracine fishes were not a monophyletic group, but were the same as genera Gymnocypris and Schizogypsis. Our results indicated that the molecular phylogenetic relationships apparently reflected their geographical and historical associations with drainages, namely species from the same and adjacent drainages clustered together and had close relationships. The divergence times of different lineages were well consistent with the rapid uplift phases of the Tibetan Plateau in the late Cenozoic, suggesting that the origin and evolution of schizothoracine fishes were strongly influenced by environment changes resulting from the upheaval of the Tibetan Plateau.
